The common methods for estimating food consumption in the environment involve either (1) performing experiments in the laboratory and relating food concentrations used to those in the environment, or (2) analyzing gut contents of animals directly from the environment and estimating food consumption by some knowledge of gut residence time. Both techniques involved compromises and assumptions. The greater precision and speed of the laboratory approach is counterbalanced by the impossibility of duplicating the natural feeding milieu. We compared predation rates of the ctenophores from gut content analysis of ctenophores in 1 300 m3 enclosures, with the estimates of expected predatory effects based on controlled laboratory experiments, over a wide range of ctenophore size and food concentration. Procedures have been developed for the extraction and high-performance liquid chromatography (HPLC) analysis of paralytic shellfish poisoning (PSP) toxins from Protogonyaulax spp. grown in batch culture. Using these procedures, the toxin content of two isolates of P. tamarensis (NEPCC 183 and 255) and one isolate of P. catenella (NEPCC 355) were examined. Total toxin and individual toxin concentrations were measured for each isolate during the exponential and stationary phases of growth in batch culture. The total toxicity of each isolate as measured by HPLC analysis was found to agree with toxicity as determined by the standard mouse bioassay. Two of the isolates (255 and 355) were found to be toxic and the third (183) was non-toxic. The toxic isolates (255 and 355) both showed higher average total PSP toxin content during the exponential phase (35 and 23 fmol toxin cell-1, respectively) than during the stationary phase (21 and 8 fmol toxin cell-1, respectively). These cultures differed dramatically in their toxin composition. P. tamarensis (255) contained a large proportion of the N(21) sulfo toxins (B1, B2, C1, C2) while P. catenella (355) contained primarily Gonyautoxins 1 through 4. The percent composition of individual toxins was found to be constant throughout the growth cycle for both toxic isolates, even though the total toxin concentration varied. The Mae Tao Clinic, located on the Thailand-Burma border, has provided health services for illegal migrant workers in Thailand and internally displaced people from Burma since 1989. In 2001, the clinic launched a project with the primary aim of improving reproductive health services and the secondary aim of building clinic capacity in monitoring and evaluation (M&E). This paper first presents the project's methods and key results. The team used observation of antenatal care and family-planning sessions and client exit interviews at baseline and follow-up, approximately 13 months apart, to assess performance on six elements of quality of care. Findings indicated that improving programme readiness contributed to some improvement in the quality of services, though inconsistencies in findings across the methods require further research. The paper then identifies lessons learned from introducing M&E in a resource-constrained setting. One key lesson was that a participatory approach to M&E increased people's feelings of ownership of the project and motivated staff to collect and use data for programme decision-making to improve quality. 相似文献

This research combines laboratory and field studies with computer simulation to characterize the amount of plant-available nitrogen (PAN) released when municipal biosolids are land-applied to agronomic crops. In the laboratory studies, biosolids were incubated in or on soil from the land application sites. Mean biosolids total C, organic N, and C to N ratio were 292 g kg(-1), 41.7 g kg(-1), and 7.5, respectively. Based on CO2 evolution at 25 degrees C and optimum soil moisture, 27 of the 37 biosolids-soil combinations had two decomposition phases. The mean rapid and slow fraction rate constants were 0.021 and 0.0015 d(-1), respectively, and the rapid fraction contained 23% of the total C assuming sequential decomposition. Where only one decomposition phase existed, the mean first order rate constant was 0.0046 d(-1). The mean rate constant for biosolids stored in lagoons for an extended time was 0.00097 d(-1). The only treatment process that was related to biosolids treatment was stabilization by storage in a lagoon. Biosolids addition rates (dry basis) ranged from 1.3 to 33.8 Mg ha(-1) with a mean value of 10.6 Mg ha(-1). A relationship between fertilizer N rate and crop response was used to estimate observed PAN at each site. Mean observed PAN during the growing season was 18.9 kg N Mg(-1) or 37% of the biosolids total N. Observed PAN was linearly related to biosolids total N. Predicted PAN using the computer model Decomposition, actual growing-season weather, actual analytical data, and laboratory decomposition kinetics compared well with observed PAN. The mean computer model prediction of growing-season PAN was 19.2 kg N Mg(-1) and the slope of the regression between predicted and observed PAN was not significantly different from unity. Predicted PAN obtained using mean decomposition kinetics was related to predicted PAN using actual decomposition kinetics suggesting that mean rate constants, actual weather, and actual analytical data could be used in estimation of PAN. There was a linear relationship between predicted N mineralization for the growing season and for the first year. We describe feeding behavior of Aurelia aurita (Linnaeus) using gut content analyses of field-collected specimens and a mesocosm experiment. The field studies were conducted in Narragansett Bay, Rhode Island, USA from March to April 1988, and the mesocosm studies were done at the Marine Ecosystems Research Laboratory at the University of Rhode Island. Patterns of prey selection changed with medusa diameter. Smaller medusae (12 mm diameter) consumed mostly hydromedusan prey whereas larger medusae (up to 30 mm diameter) ingested greater numbers of copepod prey. While larger medusae did feed on copepods, their diet also contained more barnacle nauplii and hydromedusae than expected from the relative abundances of these prey types in plankton samples. A marginal flow mechanism of feeding by A. aurita provided an explanation for the patterns of prey selection we observed in medusae of different sizes and among widely divergent prey types. Our data indicated that large prey, with escape speeds slower than the marginal flow velocities around the bell margins of A. aurita, made up a substantial fraction of the daily ration when they were available. Such prey species may be more important to nutrition than the more abundant copepods and microzooplankton. Because of the considerable uncertainties associated with modeling complex ecosystem processes, it is essential that every effort be made to test model performance prior to relying on model projections for assessment of future surface water chemical response to environmental perturbation. Unfortunately, long-term chemical data with which to validate model performance are seldom available. The authors present here an evaluation of historical acidification of lake waters in the northeastern United States, and compare historical changes in a set of lakes to hindcasts from the same watershed model (MAGIC) used to estimate future changes in response to acidic deposition. The historical analyses and comparisons with MAGIC model hindcasts and forecasts of acid-base response demonstrate that the acidic and low-ANC lakes in this region are responsive to strong acid inputs. However, the model estimates suggest lakewater chemistry is more responsive to atmospheric inputs of sulfur than do the estimates based on paleolimnological historical analyses. A 'weight-of-evidence approach' that incorporates all available sources of information regarding acid-base response provides a more reasonable estimate of future change than an approach based on model projections alone. The results of these analyses have important implications for predicting future surface water chemical change in response to acidic deposition, establishing critical loads of atmospheric pollutants, and other environmental assessment activities where natural variation often exceeds the trends under investigation (high noise-to-signal ratio). A weight-of-evidence approach was used by the US National Acid Precipitation Assessment Program (NAPAP) to assess the sensitivity of chemistry and biology of lakes and streams to hypothesized changes in sulfate deposition over the next 50 years. The analyses focused on projected effects in response to differences in the magnitude and the timing of changes in sulfate deposition in the north-eastern United States, the Mid-Appalachian Highlands, and the Southern Blue Ridge Province. A number of tools was used to provide the weight of evidence that is required to have confidence in an assessment that has many uncertainties because of the complexity of the systems for which the projections of future conditions were made and because of limited historical data. The MAGIC model provided the projections of chemical changes in response to alternative deposition scenarios. Projected chemical conditions were input into biological models that evaluate effects on fish populations. The sensitivity of water chemistry and brook trout resources to the hypothesized changes in deposition was found to be greatest in the Adirondacks and Mid-Atlantic Highlands. Under the hypothesized sulfur deposition reduction scenarios, chemical conditions suitable for fish were projected to improve 20-30 years sooner than with the scenario that assumed no new legislated controls. Other lines of evidence, e.g. other models, field observations, and paleolimnological findings, were used to evaluate uncertainty in the projections. Model parameter/calibration uncertainty for the chemical models and population sampling uncertainty were explicitly quantified. ABSTRACT: A technique for using the rotifer Philodina acuticornis as a bioassay organism is described. The rotifer was exposed to a range of concentrations for each of 14 toxicants. The effects of the heavy metals cadmium, chromium, cobalt, copper, lead, mercury, nickel, silver and zinc were studied. Based upon 96 hours exposure in soft water the sensitivity of the rotifer to the metals from the most toxic to least toxic was: cadmium, mercury and copper, zinc, silver, nickel (chloride), chromium, nickel (sulfate), lead and colbalt. In hard water with 96 hours exposure the most to least toxic respectively were: cadmium, copper, mercury, chromium and lead. The 48 hour EC50 value suggests that zinc will follow mercury in relative toxicity when Philodina is tested in hard water. In a comparison of the toxicity of the chloride and sulfate salts of cadmium, nickel and zinc in soft water cadmium sulfate and zinc sulfate were more toxic after 96 hours; nickelous chloride was more toxic than nickelous sulfate. Increased water hardness decreased the toxicity of the heavy metals studied. The results suggest that this rotifer may be more sensitive than the bluegill sunfish to the salts of cadmium, copper, nickel, zinc and chromium and less sensitive to lead. Data for cobalt, silver and mercury were not available. Philodina was extremely tolerant of ammonium chloride and phenol. The feasibility and economics of using an inexpensive, readily cultured and available organism such as Philodina acuticornis as a bioassay organism were discussed. 相似文献
