We developed quantitative PCR (qPCR) assays to distinguish each of the four clades (AD) of dinoflagellate endosymbionts (genus Symbiodinium) commonly found in Caribbean corals. We applied these primer sets, which target portions of the multi-copy ribosomal DNA (rDNA) gene family, to assess the presence/absence of symbionts in clade D (as indicated by the detection of clade D DNA). We detected these symbionts in five of six Caribbean host species/genera (21% of samples analyzed, N = 10 of 47 colonies), from which clade D had rarely or never been observed. This suggests that Symbiodinium in clade D are present in a higher diversity of coral species than previously thought. This qPCR-based approach can improve our understanding of the total microbial diversity associated with corals, particularly in hosts thought to be relatively specific, and has many other potential applications for studies of coral reef ecology and conservation.  相似文献

The pelagic copepod Calanus pacificus ranges nearly continuously across temperate-boreal regions of the North Pacific Ocean and is currently divided into three subspecies—C. pacificus oceanicus, C. pacificus californicus, C. pacificus pacificus—based on subtle morphological differences and geographic location. The relation between geography and genetic differentiation was examined for 398 C. pacificus individuals sampled from six widely distributed locations across the North Pacific, including an open ocean site and coastal sites on both sides of the North Pacific basin. For each individual copepod, the DNA sequence was determined for a 421-bp region of the mitochondrial coxI gene (mtCOI). A total of sixty-three different mtCOI sequences, or haplotypes, were detected, with a sequence divergence between haplotypes of 0.2–3.1%. The number and distribution of haplotypes varied with sampling location; 12 haplotypes were distributed across multiple sampling locations, and 51 occurred at only one location. Five genetically distinct populations were detected based on F ST values. Haplotype minimum spanning networks, nucleotide divergence and F ST values indicated that individuals from coastal sites in the North Pacific Ocean were more closely related to each other than to individuals from the open ocean site at Station P. These results provide genetic support for the designation of two subspecies—a coastal subspecies that consists of what is currently referred to as C. p. pacificus and C. p. californicus and an open ocean subspecies C. p. oceanicus. This work also indicates that planktonic copepods with potentially high dispersal capacity can develop genetically structured populations in the absence of obvious geographic barriers between proximate locales within an ocean basin.  相似文献

One potential trade-off that bold individuals face is between increased predation risks and gains in resources. Individuals experiencing high predation and hungry individuals (or individuals with low body condition) are predicted to show increased boldness. We examined one behavioral trait previously reported to be associated with boldness (the time individual fish needed to emerge from shelter) in various populations of mollies (Poecilia spp.). Our study system included several southern Mexican surface streams with high piscine predation and high food availability, sulfidic surface streams with high avian predation, in which the inhabiting fish show reduced body condition, and a sulfidic cave, where predation and body condition are low. Our comparison revealed very short times to emerge from the start box in populations from non-sulfidic streams. In sulfidic habitats (whether surface or cave), it took individual Poecilia mexicana considerably longer to emerge from the start box, and the same difference was also found in an independent comparison between P. mexicana and the closely related, highly sulfide-adapted Poecilia sulphuraria. Fish reared under common garden conditions (in the absence of predators and hydrogen sulfide) showed intermediate boldness scores to the extremes observed in the field. Our data thus indicate that (a) boldness is shaped by environmental conditions/experiential effects, but is not heritable, (b) predation affects boldness in the predicted direction, but (c) low body condition leads to reduced boldness. Extremophile Poecilia spp. spend most of their time surfacing to survive under sulfidic and hypoxic conditions, which exposes them to increased levels of predations, but the fish forage on the bottom. Hence, in this system, increased boldness does not increase foraging success. We argue that energy limitation favors reducing energetically costly behaviors, and exploring novel environments may be just one of them.  相似文献

Using a biopsy dart system, samples of skin tissue were collected from southern right whales (Eubalaena australis) in 1995 on two wintering grounds, southwest Australia (n = 20) and the Auckland Islands of New Zealand (n = 20); and on offshore feeding grounds at Latitudes 40 to 43°, south of Western Australia (n = 5). A variable section of the mitochondrial DNA control-region (289 nucleotides) was amplified and sequenced from these 45 individuals (21 males, 20 females and 4 of unknown sex), distinguishing a total of seven unique sequences (i.e. mtDNA haplotypes). Two haplotypes were found on both wintering grounds (including a common type representing 45% of each sample), and five types were unique to only one wintering ground. An analysis of variance adapted for molecular information revealed significant genetic differentiation between the two wintering grounds (p = 0.017). The feeding-ground sample was too small for statistical comparison with the wintering grounds, but included two haplotypes found only in the Auckland Islands as well as the common haplotype found on both wintering grounds. The nucleotide diversity and differentiation of mtDNA among the right whales was similar to that among humpback whales (Megaptera novaeangliae) from the same regions (Baker et al. 1998), but haplotype diversity was significantly reduced, perhaps as a result of more intensive hunting during the last century and continued illegal hunting during this century. Researchers have noted that current water quality protection strategies, like nutrient management plans, lack a sound hydrological underpinning for pollutant transport processes. This is especially true for areas like the northeastern U.S. where copious research has shown that variable source area hydrology largely governs runoff generation. The goal of this study was to develop a scientifically justified method to identify the locations that generate overland flow. Furthermore, this methodology must be computationally simple enough that it can be utilized or incorporated into nutrient management plans and other established water quality tools. We specifically tested the reliability of the 'distance from a stream,'D(s), and the 'topographic index,'lambda, to predict areas with a high propensity for generating overland flow, i.e. hydrologically sensitive areas (HSA). HSAs were defined by their probability of generating runoff, P(sat), based on 30 year simulations using a physically based hydrological model. Using GIS, each location's P(sat) was correlated with D(s) and lambda. We used three Delaware Co., NY watersheds in the New York City watershed system with areas varying in size from 1.6 to 37 km2 and with forested and agricultural land uses. The topographic index gave stronger, more regionally consistent correlations with P(sat) than did D(s). Equations correlating lambda and P(sat) for each month are presented and can be used to estimate hydrological sensitivity in the region surrounding our study watersheds, i.e. in Delaware Co. This work is currently being incorporated into an Internet Mapping System to facilitate user-friendly, on-line identification of HSAs.  相似文献

Offset schemes help avoid or revert habitat loss through protection of existing habitat (avoided deforestation), through the restoration of degraded areas (natural regrowth), or both. The spatial scale of an offset scheme may influence which of these 2 outcomes is favored and is an important aspect of the scheme's design. However, how spatial scale influences the trade-offs between the preservation of existing habitat and restoration of degraded areas is poorly understood. We used the largest forest offset scheme in the world, which is part of the Brazilian Forest Code, to explore how implementation at different spatial scales may affect the outcome in terms of the area of avoided deforestation and area of regrowth. We employed a numerical simulation of trade between buyers (i.e., those who need to offset past deforestation) and sellers (i.e., landowners with exceeding native vegetation) in the Brazilian Amazon to estimate potential avoided deforestation and regrowth at different spatial scales of implementation. Allowing offsets over large spatial scales led to an area of avoided deforestation 12 times greater than regrowth, whereas restricting offsets to small spatial scales led to an area of regrowth twice as large as avoided deforestation. The greatest total area (avoided deforestation and regrowth combined) was conserved when the spatial scale of the scheme was small, especially in locations that were highly deforested. To maximize conservation gains from avoided deforestation and regrowth, the design of the Brazilian forest-offset scheme should focus on restricting the spatial scale in which offsets occur. Such a strategy could help ensure conservation benefits are localized and promote the recovery of degraded areas in the most threatened forest landscapes.  相似文献

Aerosol organic nitrogen over the remote Atlantic Ocean   总被引:1,自引:0,他引:1  
Water soluble organic nitrogen (WSON) has been measured in aerosols collected on three research cruises on the Atlantic Ocean from approximately 55°N to 45°S. Results are interpreted using air mass back trajectories and results for other aerosol components. WSON concentrations range from <1 to ~40 (median 5.6) nmol m?3 with significant WSON concentrations in both fine (<1 μm) and coarse mode (>1 μm) aerosol. Concentrations of WSON were highest in samples containing Saharan dust, suggesting a locally significant source associated with soil dust. More generally WSON concentrations were highest in air which had recently crossed continental areas. In the whole data set, WSON is well correlated to total soluble nitrogen and represents approximately 25% of total nitrogen. This correlation implies a significant anthropogenic contribution to the organic nitrogen.  相似文献
