Molecular methods are useful both to monitor natural communities of bacteria, and to track specific bacterial markers in complex environments. Length-heterogeneity polymerase chain reaction (LH-PCR) and terminal restriction fragment length polymorphism (T-RFLP) of 16S rDNAs discriminate among 16S rRNA genes based on length polymorphisms of their PCR products. With these methods, we developed an alternative indicator that distinguishes the source of fecal pollution in water. We amplify 16S rRNA gene fragments from the fecal anaerobic genus Bacteroides with specific primers. Because Bacteroides normally resides in gut habitats, its presence in water indicates fecal pollution. Molecular detection circumvents the complexities of growing anaerobic bacteria. We identified Bacteroides LH-PCR and T-RFLP ribosomal DNA markers unique to either ruminant or human feces. The same unique fecal markers were recovered from polluted natural waters. We cloned and sequenced the unique markers; marker sequences were used to design specific PCR primers that reliably distinguish human from ruminant sources of fecal contamination. Primers for more species are under development. This approach is more sensitive than fecal coliform assays, is comparable in complexity to standard food safety and public health diagnostic tests, and lends itself to automation and high-throughput. The Khorezm region is located in the northwest of Uzbekistan, approximately 350 km from the current shore of the Aral Sea. It comprises a large‐scale irrigation system that conveys water from the river Amu Darya to agricultural land cropped mainly with cotton, wheat, and rice. Khorezm's water resources are vulnerable as they depend on upstream developments and are indispensable to rural livelihoods and state budgets. Since water scarcity is expected to increase in the future, sustainable water management is a necessity. Hence, the objectives of the paper are to: (1) conceptualize the distinctive features of water management in Khorezm; (2) provide an integrated analysis of water management by establishing linkages between society, technical infrastructure, and the bio‐physical environment; and (3) make policy and technology recommendations for improved water management. To conceptualize water management in Khorezm, the paper distinguishes three types of practices: formal practices, strategic practices, and discursive practices. Based on these, it presents an analysis of water management on the state water management level, the water user association level, and the farmer and field level. For each level, recommendations are given. The paper concludes that elements of integrated water resources management (IWRM) such as transparency, accountability, participation, and technical efficiency are relevant to improve water management in Khorezm, as elsewhere. In addition, it underlines the need to create legal space for agency and innovation. Both similarities and differences in summertime atmospheric photochemical oxidation appear in the comparison of four field studies: TEXAQS2000 (Houston, 2000), NYC2001 (New York City, 2001), MCMA2003 (Mexico City, 2003), and TRAMP2006 (Houston, 2006). The compared photochemical indicators are OH and HO2 abundances, OH reactivity (the inverse of the OH lifetime), HOx budget, OH chain length (ratio of OH cycling to OH loss), calculated ozone production, and ozone sensitivity. In terms of photochemical activity, Houston is much more like Mexico City than New York City. These relationships result from the ratio of volatile organic compounds (VOCs) to nitrogen oxides (NOx), which are comparable in Houston and Mexico City, but much lower in New York City. Compared to New York City, Houston and Mexico City also have higher levels of OH and HO2, longer OH chain lengths, a smaller contribution of reactions with NOx to the OH reactivity, and NOx-sensitivity for ozone production during the day. In all four studies, the photolysis of nitrous acid (HONO) and formaldehyde (HCHO) are significant, if not dominant, HOx sources. A problematic result in all four studies is the greater OH production than OH loss during morning rush hour, even though OH production and loss are expected to always be in balance because of the short OH lifetime. The cause of this discrepancy is not understood, but may be related to the under-predicted HO2 in high NOx conditions, which could have implications for ozone production. Three photochemical indicators show particularly high photochemical activity in Houston during the TRAMP2006 study: the long portion of the day for which ozone production was NOx-sensitive, the calculated ozone production rate that was second only to Mexico City's, and the OH chain length that was twice that of any other location. These results on photochemical activity provide additional support for regulatory actions to reduce reactive VOCs in Houston in order to reduce ozone and other pollutants. 相似文献

Radical chemistry in the nocturnal urban boundary layer is dominated by the nitrate radical, NO3, which oxidizes hydrocarbons and, through the aerosol uptake of N2O5, indirectly influences the nitrogen budget. The impact of NO3 chemistry on polluted atmospheres and urban air quality is, however, not well understood, due to a lack of observations and the strong impact of vertical stability of the boundary layer, which makes nocturnal chemistry highly altitude dependent.Here we present long-path DOAS observations of the vertical distribution of the key nocturnal species O3, NO2, and NO3 during the TRAMP experiment in Summer 2006 in Houston, TX. Our observations confirm the altitude dependence of nocturnal chemistry, which is reflected in the concentration profiles of all trace gases at night. In contrast to other study locations, NO3 chemistry in Houston is dominated by industrial emissions of alkenes, in particular of isoprene, isobutene, and sporadically 1,3-butadiene, which are responsible for more than 70% of the nocturnal NO3 loss. The nocturnally averaged loss of NOx in the lowest 300 m of the Houston atmosphere is ~0.9 ppb h?1, with little day-to-day variability. A comparison with the daytime NOx loss shows that NO3 chemistry is responsible for 16–50% of the NOx loss in a 24-h period in the lowest 300 m of the atmosphere. The importance of the NO3 + isoprene/1,3-butadiene reactions implies the efficient formation of organic nitrates and secondary organic aerosol at night in Houston. 相似文献

The use of ion exchange membranes for isotope analyses on soil water sulfate: laboratory experiments
To investigate the potential use of anion exchange membranes (plant root simulator [PRS] probes) for isotope investigations of the soil sulfur cycle, laboratory experiments were performed to examine the sulfate exchange characteristics and to determine the extent of sulfur and oxygen isotope fractionation during sulfate sorption and desorption on the probes in aqueous solutions and simulated soil solutions. The sulfate-exchange tests in aqueous solutions under varying experimental conditions indicated that the amount of sulfate exchanged onto PRS probes increased with increasing reaction time, initial sulfate concentration, and the number of probes used (= surface area), whereas the percentage of removal of available sulfate was constant irrespective of the initial sulfate concentration. The competition of nitrate and chloride in the solution lowered the amount of exchanged sulfate. The exchange experiments in a simulated soil under water-saturated and water-unsaturated conditions showed that a considerable proportion of the soil sulfate was exchanged by the PRS probes after about 10 d. There was no evidence for significant sulfur and oxygen isotope fractionation between soil sulfate and sulfate recovered from the PRS probes. The species Agrostis stolonifera, Brassica napus and Trifolium repens representing different ecological strategies, were selected to study the effect of Zn contamination on Zn tolerance, uptake and accumulation patterns. Parallel tests were carried out with increasing concentrations of Zn in a semi-field lysimeter and hydroponics in the climate chamber. A significant reduction in biomass production or root length and an increase in shoot Zn concentration was observed for all species at increasing external Zn concentrations. However, shoot biomass production, Zn tolerance and Zn accumulation differed significantly among the tested species. The results in both experimental set-ups were quite similar concerning Zn tolerance and accumulation and improved the validity of the findings. The rather specific responses of the different plant species to Zn contamination interfere with the more generic approach used in risk assessment studies.
