Toluene dioxygenase (tod) is a multicomponent enzyme system in Pseudomonas putida F1. Tod can mediate the degradation of Trichloroethylene (TCE), a widespread pollutant. In this study, we try to explore the TCE-regulated tod expression by using real-time qRT-PCR. The minimal culture media were supplemented with glucose, toluene, or a mixture of glucose/toluene respectively as carbon and energy sources. The TCE was injected into each medium after a 12-hour incubation period. The TCE injection severely affected bacterial growth when cultured with toluene or toluene/glucose mixtures. The cell density dropped 61 % for bacteria growing in toluene and 36 % for bacteria in the glucose/toluene mixture after TCE injection, but the TCE treatment had little effect on bacteria supplied with glucose alone. The decrease in cell number was caused by the cytotoxicity of the TCE metabolized by tod. The results from the real-time qRT-PCR revealed that TCE was capable of inducing tod expression in a toluene-dependent manner and that the tod expression level increased 50 times in toluene and 3 times in the toluene/glucose mixture after 6 hours of TCE treatment. Furthermore, validation of the rpoD gene as a reference gene for P. putida F1 was performed in this study, providing a valuable foundation for future studies to use real-time qRT-PCR in the analysis of the P. putida F1 strain. 相似文献

Particulate compositions including elemental carbon (EC), organic carbon (OC), water-soluble ionic species, and elemental compositions were investigated during the period from 2004 to 2006 in southern Taiwan. The correlation between the pollutant standard index (PSI) of ambient air quality and the various particle compositions was also addressed in this study. PSI revealed a correlation with fine (r = 0.74) and coarse (r = 0.80) particulate matter (PM). PSI manifested a significant correlation with the amount of analyzed ionic species (r approximately 0.80) in coarse and fine particles and a moderate correlation with carbon content (r = 0.63) in fine particles; however, it showed no correlation with elemental content. Although the ambient air quality ranged from good to moderate, the ionic species including chloride (Cl-), nitrate (NO3-), sulfate (SO4(2-)), sodium (Na+), ammonium (NH4+), magnesium (Mg2+), and calcium (Ca2+) increased significantly (1.5-3.7 times for Daliao and 1.8-6.9 times for Tzouying) in coarse PM. For fine particles, NO3-, SO4(2-), NH4+, and potassium (K+) also increased significantly (1.3-2.4 times for Daliao and 2.8-9.6 times for Tzouying) when the air quality went from good to moderate. For meteorological parameters, temperature evidenced a slightly negative correlation with PM concentration and PSI value, which implied a high PM concentration in the low-temperature condition. This reflects the high frequency of PM episodes in winter and spring in southern Taiwan. A rotating disk reactor that significantly increases the process efficiency has been designed and evaluated for application to photocatalytic decomposition of dye pollutants in aqueous solutions. In this process, a novel multi-layer rotating disk reactor (MLRDR) was presented. Photocatalyst (TiO2) particles are immobilized on the surfaces of disks. Within each layer of the reactor, methyl orange aqueous solution is allowed to flow from the center of the disk in a radial direction along the surface of the disk, which is rotating at high speed and is irradiated with UV lamps. The effluent is then directed to the center of another layer that lies underneath. Up to four stacked layers have been tested in this study, and the effects due to the number of the layers and volumetric flow rate on reaction conversion are investigated.Results and discussion
The efficiency of this photocatalytic reactor exhibits complex dependence on these parameters. With selected operating conditions, conversions greater than 95% can be achieved within seconds of residence time. Municipal solid waste management (MSWM) is an important environmental challenge and subject in urban planning. For sustainable MSWM strategies, the critical management factors to be considered include not only economic efficiency of MSW treatment but also life-cycle assessment of the environmental impact. This paper employed linear programming technique to establish optimal MSWM strategies considering economic efficiency and the air pollutant emissions during the life cycle of a MSWM system, and investigated the correlations between the economical optimization and pollutant emissions. A case study based on real-world MSW operating parameters in Taichung City is also presented. The results showed that the costs, benefits, streams of MSW, and throughputs of incinerators and landfills will be affected if pollution emission reductions are implemented in the MSWM strategies. In addition, the quantity of particulate matter is the best pollutant indicator for the MSWM system performance of emission reduction. In particular this model will assist the decision maker in drawing up a friendly MSWM strategy for Taichung City in Taiwan. Implications: Recently, life-cycle assessments of municipal solid waste management (MSWM) strategies have been given more considerations. However, what seems to be lacking is the consideration of economic factors and environmental impacts simultaneously. This work analyzed real-world data to establish optimal MSWM strategies considering economic efficiency and the air pollutant emissions during the life cycle of the MSWM system. 高盐废水盐度变化是影响微生物生长的重要因素之一。研究以中度嗜盐菌Halomonassp.STSY.3为例,测定其在不同盐度下生长对数期末OD600,并以葡萄糖为限制性基质,利用Monod方程,对菌株进行生长动力学拟合。结果表明,Halomonassp.STSY-3的最适生长盐度为7%(以NaCl计),此盐度下其OD600 为2.56,而0%和34%盐度下OD600分别为0.47和0.06;Halomonassp.STSY-3的生长动力学模型与实验数据能较好地拟合,在最适生长盐度下动力学模型参数μmax 为2.257h-1,Ks为0.0082g/L。比较不同盐度下STSY-3生长动力学参数,得出中度嗜盐菌STSY-3的生长存在3个明显分区:最适盐度区(盐度2%~9%)、嗜盐过渡区(盐度9%~20%和0.68%~1.76%)及生长抑制区(盐度≤2%或≥20%)。其中嗜盐过渡区0.68%~1.76%由于范围较小,实际意义小,忽略不计。 相似文献
