Bacteria of the genus Vibrio are ubiquitous in aquatic environments and can be found either in culturable or in a viable but nonculturable (VBNC) state. The genus comprises many pathogenic species accountable for water and food-borne diseases that prove to be fatal, especially in developing countries, as in Guinea-Bissau (West Africa), where cholera is endemic. In order to ascertain the abundance and structure of Vibrio spp. community in well waters that serve as the sole source of water for the population, quantitative polymerase chain reaction (qPCR), PCR-denaturant gradient gel electrophoresis (DGGE), and cloning approaches were used. Results suggest that Vibrio spp. were present throughout the year in acidic, freshwater wells with a seasonal community composition shift. Vibrio spp. abundance was in accordance with the abundance found in coastal environments. Sequences closely related to pathogenic Vibrio species were retrieved from well water revealing exposure of the population to such pathogens. pH, ammonium, and turbidity, regulated by the rain pattern, seem to be the variables that contributed mostly to the shaping and selection of the Vibrio spp. community. These results reinforce the evidence for water monitoring with culture-independent methods and the clear need to create/recover water infrastructures and a proper water resources management in West African countries with similar environmental conditions. 相似文献

In the present study, selected advanced oxidation processes (AOPs)—namely, photo-Fenton (with Fe2+, Fe3+, and potassium ferrioxalate—FeOx—as iron sources), solar photo-Fenton, Fenton, and UV/H2O2—were investigated for degradation of the antineoplastic drug mitoxantrone (MTX), frequently used to treat metastatic breast cancer, skin cancer, and acute leukemia. The results showed that photo-Fenton processes employing Fe(III) and FeOx and the UV/H2O2 process were most efficient for mineralizing MTX, with 77, 82, and 90 % of total organic carbon removal, respectively. MTX probably forms a complex with Fe(III), as demonstrated by voltammetric and spectrophotometric measurements. Spectrophotometric titrations suggested that the complex has a 2:1 Fe3+:MTX stoichiometric ratio and a complexation constant (K) of 1.47 × 104 M–1, indicating high MTX affinity for Fe3+. Complexation partially inhibits the involvement of iron ions and hence the degradation of MTX during photo-Fenton. The UV/H2O2 process is usually slower than the photo-Fenton process, but, in this study, the UV/H2O2 process proved to be more efficient due to complexing of MTX with Fe(III). The drug exhibited no cytotoxicity against NIH/3T3 mouse embryonic fibroblast cells when oxidized by UV/H2O2 or by UV/H2O2/FeOx at the concentrations tested. 相似文献

This paper presents an attempt to model the water balance in the metropolitan center landfill (MCL) in Salvador, Brazil. Aspects such as the municipal solid waste (MSW) initial water content, mass loss due to decomposition, MSW liquid expelling due to compression and those related to weather conditions, such as the amount of rainfall and evaporation are considered. Superficial flow and infiltration were modeled considering the waste and the hydraulic characteristics (permeability and soil-water retention curves) of the cover layer and simplified uni-dimensional empirical models. In order to validate the modeling procedure, data from one cell at the landfill were used. Monthly waste entry, volume of collected leachate and leachate level inside the cell were monitored. Water balance equations and the compressibility of the MSW were used to calculate the amount of leachate stored in the cell and the corresponding leachate level. Measured and calculated values of the leachate level inside the cell were similar and the model was able to capture the main trends of the water balance behavior during the cell operational period. 相似文献

Foodborne transmission gastroenteritis (AGE) outbreak occurred during a celebration lunch in July, 2016, Brazil. All stool samples tested were positive for noroviruses (NoV) and phylogenetic analysis revealed that strains were genetically close to GII.17 Kawasaki_2014. The objective of this study was to determine the concentrations of trace elements in sediment samples taken from impermeable urban areas of a city with predominantly agroindustrial development characteristics. For this, samples were taken every month from paved streets in the city of Toledo, Paraná State (PR), Brazil. Thus, trace element levels were obtained through analysis performed by optical emission spectrometry with inductively coupled plasma. Levels of total organic carbon were also obtained through the dry combustion method. When compared to local background levels, the trace element levels were shown to be enriched in the following percentage proportions: barium (Ba) (45.4%), copper (Cu) (48.6%), chromium (Cr) (37.2%), manganese (Mn) (81.6%), nickel (Ni) (0%), lead (Pb) (40%), zinc (Zn) (283.7%), magnesium (Mg) (34.3%), and sodium (Na) (250.2%), whereas the carbon (C) percentages were around 1%. The most significant enrichment factors occurred for Ba, Cr, Pb, Zn, Mg, and Na. The highest rates occurred in geoaccumulation where the sediments were enriched by zinc. According to the principal guidelines, the concentrations obtained for cadmium (Cd), Cr, Cu, Ni, and Zn can cause adverse environmental impacts. 相似文献

Gemycircularvirus (GemyCV) is a group of viruses which has been recently proposed as a new viral genus detected in fecal and environmental samples around the world. GemyCVs have been detected in human blood, brain tissue, cerebrospinal fluid, and stool sample. In the present study, we demonstrate for the first time, through molecular detection and characterization, the presence of GemyCVs in environmental samples from Brazil. Our results show a percentage of positivity ranging from 69 (25/36) to 97 % (35/36) in river water samples collected in Manaus, Amazon region, and wastewater from a wastewater treatment plant located in Rio de Janeiro, respectively, revealing GemyCVs as an important environmental contaminant. 相似文献

Shear strength characterization of MSW materials is a mandatory task when performing analyses related to landfill design and landfill geometry improvements. Despite the considerable amount of research focusing on MSW mechanical behavior there remain certain aspects which are not completely understood and deserve attention in particular the case of the undrained behavior of MSW. This paper presents the results of a comprehensive laboratory testing program using a large-scale triaxial apparatus at the Federal University of Bahia, Salvador, Brazil. The effect of factors such as confining pressure, unit weight, fiber content, rate of loading and over-consolidation on the MSW mechanical response were investigated. Tested samples presented typical MSW shear/strain curves (concave upward) in all the tests, despite the pore water pressure reaching levels almost equal to the confining pressure. The obtained results show that increasing confining stress, unit weight, loading rate, fiber content and over-consolidation lead to an increase in the MSW shear strength. The importance of the fibrous components in the waste behavior is highlighted and graphs showing the variation of the MSW shear strength with fiber content in different drainage conditions are shown. The authors believe these results could be of interest to many companies, especially considering the new trend of plastic material recycling (prior landfilling) for energy recovery purposes. 相似文献

The microbial production of fumaric acid by Rhizopus arrhizus NRRL 2582 has been evaluated using soybean cake from biodiesel production processes and very high polarity (VHP) sugar from sugarcane mills. Soybean cake was converted into a nutrient-rich hydrolysate via a two-stage bioprocess involving crude enzyme production via solid state fermentations (SSF) of either Aspergillus oryzae or R. arrhizus cultivated on soybean cake followed by enzymatic hydrolysis of soybean cake. The soybean cake hydrolysate produced using crude enzymes derived via SSF of R. arrhizus was supplemented with VHP sugar and evaluated using different initial free amino nitrogen (FAN) concentrations (100, 200, and 400 mg/L) in fed-batch cultures for fumaric acid production. The highest fumaric acid concentration (27.3 g/L) and yield (0.7 g/g of total consumed sugars) were achieved when the initial FAN concentration was 200 mg/L. The combination of VHP sugar with soybean cake hydrolysate derived from crude enzymes produced by SSF of A. oryzae at 200 mg/L initial FAN concentration led to the production of 40 g/L fumaric acid with a yield of 0.86 g/g of total consumed sugars. The utilization of sugarcane molasses led to low fumaric acid production by R. arrhizus, probably due to the presence of various minerals and phenolic compounds. The promising results achieved through the valorization of VHP sugar and soybean cake suggest that a focused study on molasses pretreatment could lead to enhanced fumaric acid production. 相似文献
