Potable water is a resource out of reach for millions worldwide,and the available water may be chemically and microbiologically compromised. This is particularly acute in Africa,where water-networks may be non-existent or restricted to a small fraction of the urban population,as in the case of Guinea-Bissau,West Africa. This study was carried out seasonally in Bolama(11°N),where unprotected hand-dug wells with acidic water are the sole source of water for the population. We inspected the free-living bacterial community dynamics by automated rRNA intergenic spacer analyses,quantitative polymerase chain reaction and cloning approaches.The results revealed a clear seasonal shift in bacterial assemblage composition and microbial abundance within the same sampling site. Temperature,pH and turbidity,together with the infiltration and percolation of surface water,which takes place in the wet season,seemed to be the driving factors in the shaping and selection of the bacterial community and deterioration of water quality. Analysis of 16 S rDNA sequences revealed several potential pathogenic bacteria and uncultured bacteria associated with water and sediments,corroborating the importance of a culture-independent approach in drinking water monitoring.  相似文献

The influence of different nutrient sources on the seasonal variation of nutrients and phytoplankton was assessed in the northern area of the Perth coastal margin, south–western Australia. This nearshore area is shallow, semi-enclosed by submerged reefs, oligotrophic, nitrogen-limited and receives sewage effluent via submerged outfalls. Analysis of 14 year of field observations showed seasonal variability in the concentration of dissolved inorganic nitrogen and phytoplankton biomass, measured as chlorophyll-a. For 2007–2008, we quantified dissolved inorganic nitrogen inputs from the main nutrient sources: superficial runoff, groundwater, wastewater treatment plant effluent, atmospheric deposition and exchange with surrounding coastal waters. We validated a three-dimensional hydrodynamic-ecological model and then used it to assess nutrient-phytoplankton dynamics. The model reproduced the temporal and spatial variations of nitrate and chlorophyll-a satisfactorily. Such variations were highly influenced by exchange through the open boundaries driven by the wind field. An alongshore (south–north) flow dominated the flux through the domain, with dissolved inorganic nitrogen annual mean net-exportation. Further, when compared with the input of runoff, the contributions from atmospheric-deposition, groundwater and wastewater effluent to the domain’s inorganic nitrogen annual balance were one, two and three orders of magnitude higher, respectively. Inputs through exchange with offshore waters were considerably larger than previous estimates. When the offshore boundary was forced with remote-sensed derived data, the simulated chlorophyll-a results were closer to the field measurements. Our comprehensive analysis demonstrates the strong influence that the atmosphere–water surface interactions and the offshore dynamics have on the nearshore ecosystem. The results suggest that any additional nutrient removal at the local wastewater treatment plant is not likely to extensively affect the seasonal variations of nutrients and chlorophyll-a. The approach used proved useful for improving the understanding of the coastal ecosystem.  相似文献

The quality of results of an environmental monitoring plan is limited to the weakest component, which could be the analytical approach or sampling method. Considering both the possibilities and the fragility that sampling methods offer, this environmental monitoring study focused on the uncertainties caused by the time component. Four time series of nutrient concentration at two sampling points (PB1 and PB2) in the Ribeir?o Garcia basin in Blumenau, Brazil, which were significantly correlated to the spatial component, were considered with a 2-hour resolution to develop efficient sampling models. These models were based on the time at which there was the highest tendency toward adverse environmental effects. Fourier spectral analysis was used to evaluated the time series and resulted in two sampling models: (1) the SMCP (sampling model for critical period) that operated with 100% efficiency for registering the highest concentration of nutrients and was valid for 83% of the studied parameters; and (2) the SMGCP (sampling model for global critical period) that operated with 83 and 50% efficiency for PB1 and PB2, respectively.  相似文献

Nematodes present high potential as a biological indicator of soil quality. In this work, it was evaluated relations between soil physical properties and nematode community under sugarcane cropping and remaining of Atlantic Forest areas in Northeastern Pernambuco, Brazil. Soil samples were collected from September to November 2009 along two 200-m transects in both remaining of Atlantic Forest and sugarcane field at deeps of 0–10, 10–20, 20–30, 30–40, and 40–50 cm. For soil characterization, it was carried out analysis of soil size, water content, total porosity, bulk density, and particle density. The level of soil mechanical resistance was evaluated through a digital penetrometer. Nematodes were extracted per 300 cm3 of soil through centrifugal flotation in sucrose being quantified, classified according trophic habit, and identified in level of genus or family. Data were analyzed using Pearson correlation at 5% of probability. Geostatistical analysis showed that the penetration resistance, water content, total porosity, and bulk density on both forest and cultivated area exhibited spatial dependence at the sampled scale, and their experimental semivariograms were fitted to spherical and exponential models. In forest area, the ectoparasites and free-living nematodes exhibited spherical model. In sugarcane field, the soil nematodes exhibited pure nugget effect. Pratylenchus sp. and Helicotylenchus sp. were prevalent in sugarcane field, but in forest, there was prevalence of Dorylaimidae and Rhabditidae. Total amount of nematode did not differ between environments; however, community trophic structure in forest presented prevalence of free-living nematodes: omnivores followed by bacterial-feeding soil nematodes, while plant-feeding nematodes were prevalent in sugarcane field. The nematode diversity was higher in the remaining of Atlantic Forest. However, the soil mechanical resistance was higher under sugarcane cropping, affecting more directly the free-living nematodes; especially Dorylaimidae which was the most sensible to changes in soil physical properties.  相似文献

We report two cases of exencephaly diagnosed by transvaginal ultrasonography at 8 weeks 4 days and at 9 weeks 3 days of gestation. Both cases presented an irregular cephalic pole, and, in the case seen at 8 weeks 4 days, brain vesicles were also absent, whereas in the case seen at 9 weeks 3 days, the midline echo was indistinguishable with disorganized choroid plexuses. In both cases, anencephaly was evident at 11 and 12 weeks' gestation and the postmortem confirmed the diagnosis. Copyright © 2005 John Wiley & Sons, Ltd.  相似文献
