This work has examined cobalt(II) binding by a variety of solid humic acids (HAs) isolated from peat, plant and soil sources at temperatures down to 60K. The results confirm that X-ray absorption near-edge spectroscopy (XANES) measurements cannot distinguish between aquo and carboxylato ligands in the inner coordination sphere of Co(II). However, between 1 and 2 inner-sphere carboxylato ligands can be detected in all the peat, plant and soil-derived HA samples by extended X-ray absorption fine structure (EXAFS) measurements, indicating inner-sphere coordination of HA-bound Co(II). The precision of C(carboxylate) detection is limited by the extent and quality of the data and the contribution from inner-sphere O to the Fourier transformed peaks used to detect carbon. Putative chelate ring formation is consistent with a relatively negative entropy change in step A, the stronger Co(II) binding step by HA functional groups, and could relate to 'non-exchangeable' metal binding by HSs. 相似文献
Background, Aim and Scope
Numerous herbicides and xenobiotic organic pollutants are detoxified in plants to glutathione conjugates. Following this enzyme
catalyzed reaction, xenobiotic GS-conjugates are thought to be compartmentalized in the vacuole of plant cells. In the present
study, evidence is presented for long range transport of these conjugates in plants, rather than storage in the vacuole. To
our knowledge this is the first report about the unidirectional long range transport of xenobiotic conjugates in plants and
the exudation of a glutathione conjugate from the root tips. This could mean that plants possess an excretion system for unwanted
compounds which give them similar advantages as animals.
Materials and Methods:
Barley plants (Hordeum vulgare L. cv. Cherie) were grown in Petri dishes soaked with tap water in the greenhouse.
- Fluorescence Microscopy. Monobromo- and Monochlorobimane, two model xenobiotics that are conjugated rapidly in plant cells
with glutathione, hereby forming fluorescent metabolites, were used as markers for our experiments. Their transport in the
root could be followed sensitively with very good temporal and spatial resolution. Roots of barley seedlings were cut under
water and the end at which xenobiotics were applied was fixed in an aperture with a thin latex foil and transferred into a
drop of water on a cover slide. The cover slide was fixed in a measuring chamber on the stage of an inverse fluorescence microscope
(Zeiss Axiovert 100).
- Spectrometric enzyme assay. Glutathione S-transferase (GST) activity was determined in the protein extracts following established
methods. Aliquots of the enzyme extract were incubated with 1-chloro-2,4-dinitrobenzene (CDNB), or monochlorobimane. Controls
lacking enzyme or GSH were measured.
- Pitman chamber experiments. Ten days old barley plants or detached roots were inserted into special incubation chambers, either
complete with tips or decapitated, as well as 10 days old barley plants without root tips. Compartment A was filled with a
transport medium and GSH conjugate or L-cysteine conjugate. Compartments B and C contained sugar free media. Samples were
taken from the root tip containing compartment C and the amount of conjugate transported was determined spectro-photometrically.
Results:
The transport in roots is unidirectional towards the root tips and leads to exsudation of the conjugates at rates between
20 and 200 nmol min-1. The microscopic studies have been complemented by transport studies in small root chambers and spectroscopic
quantification of dinitrobenzene-conjugates. The latter experiments confirm the microscopic studies. Furthermore it was shown
that glutathione conjugates are transported at higher rates than cysteine conjugates, despite of their higher molecular weights.
This observation points to the existence of glutathione specific carriers and a specific role of glutathione in the root.
Discussion:
It can be assumed that long distance transport of glutathione conjugates within the plant proceeds like GSH or amino acid
transport in both, phloem and xylem. The high velocity of this translocation of the GS-X is indicative of an active transport.
For free glutathione, a rapid transport-system is essential because an accumulation of GSH in the root tip inhibits further
uptake of sulfur. Taking into account that all described MRP transporters and also the GSH plasmalemma ATPases have side activities
for glutathione derivatives and conjugates, co-transport of these xenobiotic metabolites seems credible.
- On the other hand, when GS-B was applied to the root tips from the outside, no significant uptake was observed. Thus it can
be concluded that only those conjugates can be transported in the xylem which are formed inside the root apex. Having left
the root once, there seems to be no return into the root vessels, probably because of a lack of inward directed transporters.
Conclusions:
Plants seem to possess the capability to store glutathione conjugates in the vacuole, but under certain conditions, these
metabolites might also undergo long range transport, predominantly into the plant root. The transport seems dependent on specific
carriers and is unidirectional, this means that xenobiotic conjugates from the rhizosphere are not taken up again. The exudation
of xenobiotic metabolites offers an opportunity to avoid the accumulation of such compounds in the plant.
Recommendations and Perspectives:
The role of glutathione and glutathione related metabolites in the rhizosphere has not been studied in any detail, and only
scattered data are available on interactions between the plant root and rhizosphere bacteria that encounter such conjugates.
The final fate of these compounds in the root zone has also not been addressed so far. It will be interesting to study effects
of the exuded metabolites on the biology of rhizosphere bacteria and fungi. 相似文献
The carcinogenic and toxic ptaquiloside (PTA) is a major secondary metabolite in Bracken fern (Pteridium aquilinum (L.) Kuhn) and was hypothesized to influence microbial communities in soil below Bracken stands. Soil and Bracken tissue were sampled at field sites in Denmark (DK) and New Zealand (NZ). PTA contents of 2.1 +/- 0.5 mg g(-1) and 37.0 +/- 8.7 mg g(-1) tissue were measured in Bracken fronds from DK and NZ, respectively. In the two soils the PTA levels were similar (0-5 microg g(-1) soil); a decrease with depth could be discerned in the deeper B and C horizons of the DK soil (weak acid sandy Spodosol), but not in the NZ soil (weak acid loamy Entisol). In the DK soil PTA turnover was predominantly due to microbial degradation (biodegradation); chemical hydrolysis was occurring mainly in the uppermost A horizon where pH was very low (3.4). Microbial activity (basal respiration) and growth ([3H]leucine incorporation assay) increased after PTA exposure, indicating that the Bracken toxin served as a C substrate for the organotrophic microorganisms. On the other hand, there was no apparent impact of PTA on community size as measured by substrate-induced respiration or composition as indicated by community-level physiological profiles. Our results demonstrate that PTA stimulates microbial activity and that microorganisms play a predominant role for rapid PTA degradation in Bracken-impacted soils. 相似文献
The mouthparts of Munida sarsi Huus, 1935, including the labrum and paragnaths, are described using LM and SEM, with special attention to the setal composition
and its function. We also used high-resolution macrovideo equipment to record the feeding processes and monitor setae in action.
This combination of morphological and preliminary behavioral data enabled us to arrive at a more comprehensive understanding
of the functional morphology. M. sarsi was found to be an opportunist and capable of handling and eating most kinds of animal tissue. The species can also be a
selective deposit feeder, sorting the sediment by quality and not by size. This sorting is performed by the setae on the medial
rim of the bases of maxilla 1, maxilla 2 and maxilliped 1. Many of the other mouthpart functions ranging from food detection
to ingestion are mapped with comparison to what has been found for other anomurans. Grooming was shown to be a significant
function of many mouthparts, especially the endopod of maxilliped 3. In general the mouthparts closest to the mouth have the
least complex movements and functional scheme, with the mandibles being able to perform only one movement, i.e. cutting or
crunching food objects. Some functions are attributed to the paragnath, the endopod of maxilla 2 and the epipod of maxilliped

The influence of moulting and ovarian maturation on cadmium accumulation in the tissues of female shore crabs Carcinus maenas exposed to 1 mg Cd l−1 in the water was investigated. Cadmium accumulation in all tissues examined was markedly increased in crabs in the postmoult
stages (A and B) compared to crabs in all other moult stages. During the moult cycle, average cadmium accumulation in the
midgut gland ranged from 29 μg Cd g−1 dw at premoult stage (D2) to 589 μg Cd g−1 dw at postmoult stage (A). Average cadmium concentrations in the haemolymph ranged from 0.56 μg Cd ml−1 at intermoult stage (C4) to 4.6 μg Cd ml−1 at postmoult stage (A), while the gills accumulated from 103 μg Cd g−1 dw in intermoult stage (C3) to 352 μg Cd g−1 dw in postmoult stage (A). Cadmium concentration in gills and haemolymph was also significantly higher in crabs in late premoult
stage (D3) compared to C4-crabs, while midgut gland cadmium concentration remained elevated in C1- and C3- intermoult stages relative to C4. During ovarian maturation the cadmium accumulation in midgut gland, gills, ovaries and haemolymph decreased. Average cadmium
concentration in the midgut gland decreased from 63 μg g−1 dw in ovarian Stage I to 19 μg g−1 dw in ovarian Stage VI. The same pattern was observed for gills, haemolymph and ovaries. The present study demonstrates that
cadmium accumulation in the female shore crab strongly depends on the physiological status of the animal. A possible association
between physiological calcium requirements and cadmium accumulation during moulting is discussed.

Silicon-based fertilizers and soil amendments can have direct and indirect positive influences on cultivated plants. The solid forms of Si-based substances, the most widespread in use, are efficient only at high application rates due to their low level of solubility. Several types of Si-based substances such as fumed silica, slags from the iron and steel industry, modified slags, and a Si-rich product were tested using barley and pea as silicon accumulative and non-accumulative plants, respectively, at two application rates. The plants were grown under toxic concentrations of heavy metals in a greenhouse. Si-rich materials high in water-soluble Si had a positive effect at both the low and high application rates, and for both plant species. This type of substance can be regarded as Si fertilizer, demonstrating greater efficiency at a low application rate and lessened efficiency at a high application rate for protection of the cultivated plants against accumulation of the heavy metals.
In a world where issues of food safety and food security are increasingly important, the social responsibility of central actors in the food chain—producers and the main grocery chains—becomes more pressing. As a response, these actors move from implicitly assuming social responsibilities implied in laws, regulations and ethical customs, towards explicitly expressing social responsibilities. In this paper, we discuss the ethical values relevant for the social responsibility of central food producers and retailers in Norway, one of the most subsidized and protected areas of food production in the world. How do the actors perceive and express their social responsibility, and—given their position in the local, national and global market—how should they handle these responsibilities? We analyze Tine and Nortura, two producers owned by farmer cooperatives with market regulator function, as well as Coop—a dominant grocery chain in Norway, with basis in the same public ownership model as the farmer-owned cooperative producers. While the complex roles of these key actors in the Norwegian food market have been criticized from several angles, we argue that these multifaceted roles put them in a good position to promote informed consumer choices in a globalized market. 相似文献
The destruction of land and forced migration during the Anfal attacks against the Kurds in Iraq in the late 1980s has been reported to have severe consequences for agricultural development. A reconstruction program to aid people in returning to their lands was launched in 1991. To assess the agricultural situation in the Duhok governorate during the pre-Anfal (A), post-Anfal (B), reconstruction (C), and present (D) periods, we mapped winter crops by focusing on inter-annual variability in vegetation greenness, using satellite images. The results indicate a decrease in cultivated area between period A and B, and a small increase between period B and C. This supports reports of a decline in cultivated area related to the Anfal campaign, and indicates increased activity during the reconstruction program. Period D showed a potential recovery with a cropland area similar to period A. 相似文献
Climate change generally requires species to migrate northward or to higher elevation to maintain constant climate conditions, but migration requirement and migration capacity of individual species can vary greatly. Individual populations of species occupy different positions in the landscape that determine their required range shift to maintain similar climate, and likewise the migration capacity depends on habitat connectivity. Here, we demonstrate an approach to quantifying species vulnerabilities to climate change for 419 rare vascular plants in Alberta, Canada, based on a multivariate velocity of climate change metric, local habitat fragmentation, and migration capacity. Climate change velocities indicated that future migration requirements ranged from 1 to 5 km/year in topographically complex landscapes, such as the Alberta Foothills and Rocky Mountains. In contrast, migration requirements to maintain constant climate in relatively flat Boreal Plains, Parkland, and Grassland ranged from 4 to 8 km/year. Habitat fragmentation was also highest in these flat regions, particularly the Parkland Natural Region. Of the 419 rare vascular plants assessed, 36 were globally threatened (G1–G3 ranking). Three globally threatened species were ranked as extremely vulnerable and five species as highly vulnerable to the interactions among climate change velocity, habitat fragmentation, and migration capacity. Incorporating dispersal characteristics and habitat fragmentation with local patterns in climate change velocity improves the assessment of climate change threats to species and may be applied to guide monitoring efforts or conservation actions. 相似文献
