State of the art of culturomics and metagenomics to study resistome was presented.The combination of culturomics and metagenomics approaches was proposed.The research directions of antibiotic resistance study has been suggested. Pharmaceutical residues, mainly antibiotics, have been called “emerging contaminants” in the environment because of their increasing frequency of detection in aquatic and terrestrial systems and their sublethal ecological effects. Most of them are undiscovered. Both human and veterinary pharmaceuticals, including antibiotics, are introduced into the environment via many different routes, including discharges from municipal wastewater treatment plants and land application of animal manure and biosolids to fertilize croplands. To gain a comprehensive understanding of the widespread problem of antibiotic resistance, modern and scientific approaches have been developed to gain knowledge of the entire antibiotic-resistant microbiota of various ecosystems, which is called the resistome. In this review, two omics methods, i.e. culturomics, a new approach, and metagenomics, used to study antibiotic resistance in environmental samples, are described. Moreover, we discuss how both omics methods have become core scientific tools to characterize microbiomes or resistomes, study natural communities and discover new microbes and new antibiotic resistance genes from environments. The combination of the method for get better outcome of both culturomics and metagenomics will significantly advance our understanding of the role of microbes and their specific properties in the environment. 相似文献
Male Xenopus laevis frogs produce underwater advertisement calls that attract gravid females and suppress calling by male competitors. Here we
explore whether groups of males establish vocal ranks and whether auditory cues alone suffice for vocal suppression. Tests
of male–male pairs within assigned groups reveal linear vocal dominance relations, in which each male has a defined rank.
Both the duration over which males interact, as well as the number of competitive opportunities, affect linearity. Linear
dominance across the group is stable for about 2 weeks; rank is dynamic. Males engage in physical interactions (clasping)
while paired but clasping and vocal rank are not correlated. Playbacks of advertisement calls suppress calling and calls from
high- and low-ranking males are equally effective. Thus, auditory cues alone suffice to suppress vocal behavior. Playback
intensities equivalent to a nearby male advertising effectively suppress calling while low-intensity playbacks are either
ineffective or stimulate vocal behavior. X. laevis advertisement calls are biphasic, composed of alternating fast and slow click trills. Approximately half the males tested
are more vocally suppressed by all slow than by all fast trills; thus, these males can distinguish between the two phases.
The fully aquatic family Pipidae diverged from terrestrial ancestors approximately 170 mya. Vocal suppression in the X. laevis mating system may represent the translation of an ancient anuran social strategy to underwater life. 相似文献
Focus groups have achieved a profile in the analysis of public values unparalleled since the emergence of national opinion polls. While they have been used in a variety of public policy, academic and political settings it is their employment within the field of environmental policy, specifically the land-use planning system, that is the focus of this paper. Using evidence from empirical research it is argued that the research-policy interface of environmental focus groups and planning is currently both undertheorised and underpractised. Undertheorised in that much of the literature on focus groups concentrates on their conduct rather than how they can inform policy. Underpractised because focus group data are failing to have much impact on policy. This paper concentrates on the second of these propositions by showing that while some of the problems with quantitative public environmental values surveys can be overcome by focus groups, the nature and content of the data they produce are not easily assimilated by existing planning structures. In an era when the role of the public, in all its diversity, is being emphasised in policy circles, and collaborative projects between academia and policy communities on environmental issues are becoming increasingly commonplace, early warning signs of tensions between research findings and policy development need to be heeded. A failure to acknowledge these problems may well lead to impoverished environmental policy in planning, a further marginalisation of publics from policy processes and an unjust devaluation of the focus-group method. 相似文献
Europe has a long history of human pressure on freshwater ecosystems. As pressure continues to grow and new threats emerge, there is an urgent need for conservation of freshwater biodiversity and its ecosystem services. However, whilst some taxonomic groups, mainly vertebrates, have received a disproportionate amount of attention and funds, other groups remain largely off the public and scientific radar. Freshwater mussels (Bivalvia, Unionida) are an alarming example of this conservation bias and here we point out six conceptual areas that need immediate and long-term attention: knowledge, threats, socioeconomics, conservation, governance and education. The proposed roadmap aims to advance research, policy and education by identifying the most pressing priorities for the short- and long-term conservation of freshwater mussels across Europe. 相似文献
The main objective of this study is to quantify the potential overpressures due to Vapor Cloud Explosions (VCEs) and the potential gas buildup by using Computation Fluid Dynamics (CFD) for onshore or offshore facilities.A series of CFD simulations and analyses have been performed for the various vapor dispersion scenarios, covering different release rates and release locations. The overpressure that could result from the potential VCE is assessed by CFD simulation for the largest explosive transient gas cloud. The results from the analyses also comprise an extensive picture of probable leak scenarios having the potential to make an explosive gas cloud.The CFD analysis results could be applied to provide input for detailed risk-based design and risk analysis, to find safe and cost-optimal design against explosions. 相似文献
The A-01 wetland treatment system (WTS) is a surface flow wetland planted with giant bulrush [Schoenoplectus californicus (C.A. Mey.) Palla] that is designed to remove Cu and other metals from the A-01 National Pollution Discharge Elimination System (NPDES) effluent at the Savannah River Site near Aiken, SC. Copper, Zn, and Pb concentrations in water were usually reduced 60 to 80% by passage through the treatment system. The Cu concentrations in the wetland sediments increased from about 4 to 205 and 796 mg kg(-1), respectively, in the organic and floc sediment layers in cell 4A over a 5-yr period. Metal concentrations were higher in the two top layers of sediment (i.e., the floc and organic layers) than in the deeper inorganic layers. Sequential extraction was used to evaluate remobilization and retention of Cu, Pb, Zn, Mn, and Fe in the wetland sediment. Metal remobilization was determined by the potentially mobile fraction (PMF) and metal retention by the recalcitrant factor (RF). The PMF values were high in the floc layer but comparatively low in the organic and inorganic layers. High RF values for Cu, Zn, and Pb in the organic and inorganic layers indicated that these metals were strongly bound in the sediment. The RF values for Mn were lower than for the other elements especially in the floc layer, indicating low retention or binding capacity. Retention of contaminants was also evaluated by distribution coefficient (Kd) values. Distribution coefficient (Kd) values were lower for Cu and Zn than for Pb, indicating a smaller exchangeable fraction for Pb. 相似文献
The multi-scale approach to conserving forest biodiversity has been used in Sweden since the 1980s, a period defined by increased reserve area and conservation actions within production forests. However, two thousand forest-associated species remain on Sweden’s red-list, and Sweden’s 2020 goals for sustainable forests are not being met. We argue that ongoing changes in the production forest matrix require more consideration, and that multi-scale conservation must be adapted to, and integrated with, production forest development. To make this case, we summarize trends in habitat provision by Sweden’s protected and production forests, and the variety of ways silviculture can affect biodiversity. We discuss how different forestry trajectories affect the type and extent of conservation approaches needed to secure biodiversity, and suggest leverage points for aiding the adoption of diversified silviculture. Sweden’s long-term experience with multi-scale conservation and intensive forestry provides insights for other countries trying to conserve species within production landscapes.
