Monoterpenes are structurally diverse natural compounds that play an essential role in the chemical ecology of a wide array of organisms. A key enzyme in monoterpene biosynthesis is geranyl diphosphate synthase (GPPS). GPPS is an isoprenyl diphosphate synthase that catalyzes a single electrophilic condensation reaction between dimethylallyl diphosphate (C5) and isopentenyl diphosphate (C5) to produce geranyl diphosphate (GDP; C10). GDP is the universal precursor to all monoterpenes. Subsequently, monoterpene synthases are responsible for the transformation of GDP to a variety of acyclic, monocyclic, and bicyclic monoterpene products. In pheromone-producing male Ips pini bark beetles (Coleoptera: Scolytidae), the acyclic monoterpene myrcene is required for the production of the major aggregation pheromone component, ipsdienol. Here, we report monoterpene synthase activity associated with GPPS of I. pini. Enzyme assays were performed on recombinant GPPS to determine the presence of monoterpene synthase activity, and the reaction products were analyzed by coupled gas chromatography–mass spectrometry. The functionally expressed recombinant enzyme produced both GDP and myrcene, making GPPS of I. pini a bifunctional enzyme. This unique insect isoprenyl diphosphate synthase possesses the functional plasticity that is characteristic of terpene biosynthetic enzymes of plants, contributing toward the current understanding of product specificity of the isoprenoid pathway.  相似文献

We describe three cases in which we used fluorescence in situ hybridization (FISH), polymerase chain reaction (PCR) and comparative genomic hybridization (CGH) to characterize Y chromosome structural anomalies, unidentifiable by conventional G-banding. Case 1 was a 46,X,+mar karyotype; FISH analysis revealed an entire marker chromosome highlighted after hybridization with the Y chromosome painting probe. The PCR study showed the presence of Y chromosome markers AMG and SY620 and the absence of SY143, SY254 and SY147. CGH results confirmed the loss of Yq11.2-qter. These results indicated the presence of a deletion: del(Y)(q11.2). Case 2 was a 45,X [14]/46,XY[86] karyotype with a very small Y chromosome. The PCR study showed the presence of Y chromosome markers SY620 and AMG, and the absence of SY143, SY254 and SY147. CGH results showed gain of Yq11.2-pter and loss of Yq11.2-q12. These results show the presence of a Yp isodicentric: idic(Y)(q11.2). Case 3 was a 45,X,inv(9)(p11q12)[30]/46,X,idic(Y)(p11.3?),inv(9)(p11q12)[70] karyotype. The FISH signal covered all the abnormal Y chromosome using a Y chromosome paint. The PCR study showed the presence of Y chromosome markers AMG, SY620, SY143, SY254 and SY147. CGH only showed gain of Yq11.2-qter. These results support the presence of an unbalanced (Y;Y) translocation. Our results show that the combined use of molecular and classical cytogenetic methods in clinical diagnosis may allow a better delineation of the chromosome regions implicated in specific clinical disorders. The Caribbean is highly vulnerable to coastal hazards. Based on their short recurrence intervals over the intra-American seas, high-category tropical cyclones and their associated effects of elevated storm surge, heavy wave impacts, mudslides and floods represent the most serious threat. Given the abundance of historical accounts and trigger mechanisms (strike-slip motion and oblique collision at the northern and southern Caribbean plate boundaries, submarine and coastal landslides, volcanism), tsunamis must be considered as well. This paper presents interdisciplinary multi-proxy investigations of sediment cores (grain size distribution, carbonate content, loss-on-ignition, magnetic susceptibility, microfauna, macrofauna) from Washington-Slagbaai National Park, NW Bonaire (Leeward Antilles). No historical tsunami is recorded for this island. However, an allochthonous marine layer found in all cores at Boka Bartol reveals several sedimentary criteria typically linked with tsunami deposits. Calibrated 14C data from these cores point to a palaeotsunami with a maximum age of 3,300 years. Alternative explanations for the creation of this layer, such as inland flooding during tropical cyclones, cannot entirely be ruled out, though in recent times even the strongest of these events on Bonaire did not deposit significant amounts of sediment onshore. The setting of Boka Bartol changed from an open mangrove-fringed embayment into a poly- to hyperhaline lagoon due to the establishment or closure of a barrier of coral rubble during or subsequent to the inferred event. The timing of the event is supported by further sedimentary evidence from other lagoonal and alluvial archives on Bonaire.  相似文献

In this paper we review the published, scientific literature addressing the response of nutrients, sediment, pathogens, and cyanobacterial blooms to historical and potential future changes in air temperature and precipitation. The goal is to document how different attributes of water quality are sensitive to these drivers, to characterize future risk, to inform management responses, and to identify research needs to fill gaps in our understanding. Results suggest that anticipated future changes present a risk of water quality and ecosystem degradation in many United States locations. Understanding responses is, however, complicated by inherent high spatial and temporal variability, interactions with land use and water management, and dependence on uncertain changes in hydrology in response to future climate. Effects on pollutant loading in different watershed settings generally correlate with projected changes in precipitation and runoff. In all regions, increased heavy precipitation events are likely to drive more episodic pollutant loading to water bodies. The risk of algal blooms could increase due to an expanded seasonal window of warm water temperatures and the potential for episodic increases in nutrient loading. Increased air and water temperatures are also likely to affect the survival of waterborne pathogens. Responding to these challenges requires understanding of vulnerabilities, and management strategies to reduce risk.  相似文献

A public mobilization approach known as nikinake drives implementation and technology upscaling in Ethiopia's agricultural extension. This study investigates and describes the processes and effectiveness of nikinake as an extension method used for natural resource management (NRM). The paper draws on empirical field research conducted in Oromia and the southern region of Ethiopia by looking at nikinake in the context of a watershed management campaign in 2015 and 2016. Nikinake is used as an approach to mobilize the public and to promote the skills of farmers and development actors. In principle, the implementation of NRM is voluntary; however, it is largely planned top‐down and enforced through state actors and informal institutions. This study suggests effective integration of social mobilization with reliable extension and a paradigm shift in emphasis from spatial coverage to an effective outcome. Additionally, sustainability and scalability of NRM interventions could be ameliorated by improving experts’ technical skills, raising farmers’ awareness, improving an incentive system, building trust, and better integrating past watershed management and future planning activities. We reflect on the significance of the nikinake experience in Ethiopia for a broader theory of extension‐as‐mobilization for rural development. From the Ethiopian case, a more general recommendation emerges for extension‐as‐mobilization schemes. For long‐term development, it is worthwhile to consider the fit between yearly campaigns as ad hoc project organizations and the existing pattern of actors and institutions responsible for rural development.  相似文献

The ability of social insects to discriminate against non-nestmates is vital for maintaining colony integrity, and in most social insect species, individuals act aggressively towards non-nestmates that intrude into their nest. Our experimental field data revealed that intra-colony aggression in the primitive bulldog ant Myrmecia nigriceps is negligible; our series of bioassays revealed no significant difference in the occurrence of aggression in trials involving workers from the same, a close (less than 300 m) or a far (more than 1.5 km) nest. Further, non-nestmate intruders were able to enter the nest in 60% of our trials; a similar level was observed in trials involving nestmates. These results suggest that workers of M. nigriceps are either unable to recognize alien conspecifics or that the costs of ignoring workers from foreign colonies are sufficiently low to favor low levels of inter-colony aggression in this species.  相似文献
