We studied the compartmentalization of cadmium and zinc in the oligochaete Tubifex tubifex. The subcellular distribution was followed over time and levels of metallothionein-like proteins were measured. The impact of the speciation on the trophic transfer was studied by calculating the assimilation efficiencies of metals from Tubificidae fed to carp. It was found that carp were able to assimilate 9.8% of the cadmium. The expected assimilated amount of cadmium, based on the subcellular fractions which are thought to be trophically available, is however 72%. The zinc assimilation results suggest that the debris fraction is at least partially available to predators. Differential centrifugation techniques provide information about the tissue compartmentalization in aquatic organisms but it is not straightforward to directly link internal speciation in prey items to the actual assimilation in the predator. The possible impact that the compartmentalization of cadmium in T. tubifex will have on the toxicity to the organism is also discussed.  相似文献

Separate focus on crop fertilization or feeding practices inadequately describes nitrogen (N) loss from mixed dairy farms because of (1) interaction between animal and crop production and between the production system and the manager, and (2) uncertainties of herd N production and crop N utilization. Therefore a systems approach was used to study N turnover and N efficiency on 16 conventional and 14 organic private Danish farms with mixed animal (dairy) and crop production. There were significant differences in N surplus at the farm level (242 kg. N/ha. vs. 124 kg. N/ha. on conventional and organic dairy farms respectively) with a correlation between stocking rate and N surplus. N efficiency was calculated as the output of N in animal products divided by the net N import in fodder, manure and fertilizer. N turnover in herd and individual crops calculated on selected farms showed differences in organic and conventional crop N utilization. This is explained via a discussion of the rationality behind the current way of planning the optimum fertilizer application in conventional agriculture. The concept of marginal N efficiency is insufficient for correcting problems of N loss from dairy farms. Substantial reductions in N loss from conventional mixed dairy farms is probably unlikely without lower production intensity. The concept of mean farm unit N efficiency might be a way to describe the relation between production and N loss to facilitate regulation. This concept is linked to differing goals of agricultural development—i.e. intensification and separation vs. extensification and integration. It is discussed how studies in private farms—using organic farms as selected critical cases—can demonstrate possibilities for balancing production and environmental concern.  相似文献

A rapid and simple analytical method for the determination of organochlorines, such as polychlorinated biphenyls (PCBs) and selected organochlorinated pesticides (OCPs) and organobromines, such as polybrominated diphenyl ethers (PBDEs), in sediment samples was optimised using CRM 536 (PCBs in freshwater sediment). The method involved a hot Soxhlet extraction that reduced the extraction time to 2 h. Elemental sulphur, which is present in sediments and may interfere during the analysis, was removed by means of copper powder added to the sediment during extraction and into the clean-up cartridge. The analysis of PCBs and OCPs was accomplished by gas chromatography with electron capture or mass spectrometric detection. Similar quantitative results for PCB congeners in CRM 536 were obtained using a 50-m capillary column and a 10-m narrow bore column suited for fast analysis. The analysis of PBDEs was done by mass spectrometry in negative chemical ionisation mode. Concentrations of organic pollutants in two sediment cores (approximately 50 cm depth) from the Scheldt river (south of Antwerp, Belgium) showed a relative steady state for PCBs and DDTs, with a slight decrease in the top layers, suggesting a slight decline in their concentrations due to restrictions in their usage. On the contrary, PBDEs were showing an increase in their concentrations in the top layers (up to 270 and 8400 ng/g dry weight for sum of tri- to hexa-BDE congeners and for BDE 209, respectively). This suggests an increasing trend in the concentrations of PBDEs in the Belgian environment.  相似文献

Field trials concerning the establishment of plant cover on a deposit of wastes from the Ammeberg zinc mine in central Sweden were carried out during 1976-1985. Different soil conditioners and manures were applied and plant species cultivars were evaluated with regard to plant biomass, vigour, durability and content of zinc, lead and cadmium. Sewage sludge and topsoil led to better establishment of grasses than did municipal waste, straw and hydraulic seeding. After 2 years, Festuca rubra and Poa pratensis dominated the swards. Other species (Dactylis glomerata, Bromus inermis, Lolium perenne, Phleum nodosum, Festuca pratensis and F. arundinacea) constituted only a minor part of the stand. After 10 years, F. rubra was the most dominant species, while native Agrostis tenuis had invaded 20-50% of the area within the plots. Merlin was the clearly dominant red fescue cultivar. The concentration of zinc in shoots (616 mg kg(-1) dw) was about 10% of that in the soil. Zinc concentration decreased with increasing biomass above ground. It increased with age in Scots pine needles and was very high in birch leaves. Grasses survived longer than legumes in the zinc sand waste. Among the surviving grasses was a group with high (3800 mg kg(-1) dw) and a group with low (320 mg kg(-1) dw) zinc concentrations. The low group included Merlin red fescue and Sobel creeping bent. The cultivar Merlin contained a much lower zinc concentration than the other cultivars of red fescue (375 and 624 mg kg(-1) dw, respectively). A large amount of root biomass was present in plots with dominating Merlin red fescue (1715 g m(-2)), 97% of which was concentrated in the top 10 cm of the soil. The concentration of zinc in the roots was very high (13 000-25 000 mg kg(-1) dw). Nitrate fertilizer, especially ammonium nitrate, and acidic water (pH 4.3) increased zinc leaching.  相似文献

The molecular weight changes in abiotically and biotically degraded LDPE and LDPE modified with starch and/or prooxidant were compared with the formation of degradation products. The samples were thermooxidized for 6 days at 100°C to initiate degradation and then either inoculated with Arthobacter paraffineus or kept sterile. After 3.5 years homologous series of mono- and dicarboxylic acids and ketoacids were identified by GC-MS in abiotic samples, while complete disappearance of these acids was observed in biotic environments. The molecular weights of the biotically aged samples were slightly higher than the molecular weights of the corresponding abiotically aged samples, which is exemplified by the increase in from 5200 g/mol for a sterile sample with the highest amount of prooxidant to 6000 g/mol for the corresponding biodegraded sample. The higher molecular weight in the biotic environment is explained by the assimilation of carboxylic acids and low molecular weight polyethylene chains by microorganisms. Assimilation of the low molecular weight products is further confirmed by the absence of carboxylic acids in the biotic samples. Fewer carbonyls and more double bonds were seen by FTIR in the biodegraded samples, which is in agreement with the biodegradation mechanism of polyethylene.  相似文献

Due to the low cell concentration, cultures from early amniotic fluid specimens usually require 2–3 weeks in culture prior to karyotyping. The purpose of this study was to evaluate the culture quality of amniotic fluid cells from early pregnancy, obtained by a new filter technique. The hypothetical advantage of the technique was that the increased cell yield might reduce the culture time before karyotyping. Culture quality was assessed by the number of colonies, the percentage of colonies containing mitoses in filter and control cultures, and the culture time. The setting was a consecutive clinical trial. One hundred samples were obtained from ongoing pregnancies at 11–14 weeks of gestation (mean 12·8 weeks). By circulating a mean of 26 ml of amniotic fluid through a cell filter system leading the cell-free fluid back to the amniotic cavity, the cell yield was increased in the sample of 7 ml corresponding to the dead space of the filter system. The culture results were compared with control cultures from 5 ml samples drawn from the same pregnancies prior to recirculation. The cultures from the first flushing of the filter system yielded 2·6 times more colonies and in total 4·2 times more colonies were found in the three cultures grown from each filter sample when compared with the control cultures. Moreover, the filter cultures showed significantly more colonies with mitoses. The mean culture time was 8·0 days for the filter cultures, from which the karyotypes were analysed. The controls would have needed more time in culture to fulfil the diagnostic criteria for karyotyping. One case of 47,XY, + 21 was found; the rest had normal karyotypes. We conclude that the filter technique improves the culture quality of early amniotic fluid samples and allows early arrest of the cultures.  相似文献

A technique for sampling first trimester chorionic villi for prenatal diagnosis by transabdominal fine needle biopsy is described. Specimens of chorionic villi were obtained from 49 out of 58 women, a success rate of 84.5 per cent. No fetal or maternal complications were demonstrated in the period before abortion. The procedure is useful for obtaining fetal tissue for culturing, DNA analysis and direct chromosome analysis.  相似文献

The concentration of marine antifoulant 4,5-dichloro-2-n-octyl-4-isothiazolin-3-one (DCOIT; the active ingredient in Sea-Nine 211 Antifouling Agent) leaching into a Danish Harbor from two painted ships was quantitated at varying distances from the ships. Sediment and suspended particulate matter were also analyzed for DCOIT. Water samples were concentrated on-site using C-18 solid phase extraction and subsequently analyzed by gas chromatography-tandem mass spectrometry. A strong decline in DCOIT water concentration as a function of distance from the ships was observed. The highest concentration (maximum 283 ng/l) was measured in the immediate vicinity of the ships and the concentration declined rapidly to less than the limit of detection (5 ng/l) at 400 m from the ships' surfaces. The measured decline curve was compared to that calculated using a one-dimensional model (ECoS). The comparison indicates that the primary mechanism of dissipation of DCOIT is not dilution resulting from dispersion but degradation with a rate constant in the order of 1 h(-1). Thus the field results correlate with the earlier microcosm studies demonstrating that DCOIT biodegrades rapidly in a marine environment.  相似文献
