Our experience with the prenatal detection of the Lesch-Nyhan syndrome (LNS; hypoxanthine-guanine phosphoribosyltransferase (HGPRT) deficiency) in three fetuses at risk is reported. Enzyme activities were measured in cultured amniocytes in two pregnancies, and in tissues and cultures obtained from chorionic villus sampling (CVS) in a third pregnancy. In all tissues the specific activities of HGPRT and adenine phosphoribosyltransferase (APRT) were determined and APRT/HGPRT ratios were calculated. In addition to the enzyme assays, the rate of purine synthesis de novo was assessed in the two amniocyte cultures, and the rate of [14C]hypoxanthine incorporation into nucleotides and sensitivity to azaguanine were measured in one of the amniocyte cultures. We report the diagnosis of normal fetuses by study of amniocytes in two pregnancies and of LNS using CVS in one pregnancy. In all three cases the diagnosis was confirmed.  相似文献

Organizational justice researchers tend to treat as synonyms the terms “justice” and “fairness”. We discuss different definitional arguments, concluding that these two concepts are distinct. The global biodiversity crisis requires an engaged citizenry that provides collective support for public policies and recognizes the consequences of personal consumption decisions. Understanding the factors that affect personal engagement in proenvironmental behaviors is essential for the development of actionable conservation solutions. Zoos and aquariums may be some of the only places where many people can explore their relations with wild animals and proenvironmental behaviors. Using a moderated‐mediation analysis of a survey of U.S. zoo and aquarium visitors (n = 3588), we explored the relationship between the sense of connection to animals and self‐reported engagement in proenvironmental behaviors related to climate change and how this relationship is affected by certainty that climate change is happening, level of concern about climate change, and perceptions of effectiveness in personally addressing climate change. We found a significant, directional relationship between sense of connection to animals and self‐reported proenvironmental behaviors. Political inclination within the conservative to liberal spectrum did not affect the relationship. We conclude that a personal sense of connection to animals may provide a foundation for educational and communication strategies to enhance involvement in proenvironmental actions.  相似文献

The Clean Air Act Amendments of 1990 establish new criminal and civil liability provisions that can potentially impose substantial fines and penalties. This article reviews the Act's new enforcement and liability provisions. The authors recommend that because of the new enforcement tools available to the federal government, the regulated community should implement effective self-auditing and compliance programs at facilities to help reduce the risk of criminal liability.  相似文献

In pursuit of socioeconomic development, many countries are expanding oil and mineral extraction into tropical forests. These activities seed access to remote, biologically rich areas, thereby endangering global biodiversity. We examined how protection of biodiversity and economic revenues can be balanced in biologically valuable regions. Using spatial data on oil profits and predicted species and ecosystem extents, we optimized the protection of 741 terrestrial species and 20 ecosystems of the Ecuadorian Amazon across a range of opportunity costs (i.e., sacrifices of extractive profit). We also applied spatial statistics to remotely sensed, historic deforestation data to focus the optimization on areas most threatened by imminent forest loss. Giving up 5% of a year's oil profits (US$221 million) allowed for a protected area network that retained an average of 65% of the extent of each species and ecosystem. This performance far exceeded that of the network produced by simple optimization for land area (which required a sacrifice of approximately 40% of annual oil profits [US$1.7 billion]) and used only marginally less land to achieve equivalent levels of ecological protection. We identified what we call emergency conservation targets: regions that are essential components of a cost-effective conservation reserve network but at imminent risk of destruction, thus requiring urgent and effective protection. Governments can use our methods when evaluating extractive-led development options to responsibly manage the associated ecological and economic trade-offs and protect natural capital.  相似文献

We examined the impact of exposing natural populations of marine bacteria (from seawater collected near Woods Hole, Massachusetts, USA) to multiple nitrogen and carbon sources in a series of batch growth experiments conducted from 1989 through 1990. The substrate C:N ratio (C:Ns) was varied from 1.5:1 to 10:1 either with equal amounts of NH 4 + and different amino acids or an amino acid mixture, all supplemented with glucose to maintain the C:Ns ratio equal to that of the respective amino acid, or with combinations of glucose and NH 4 + alone. A common feature of the experiments involving amino acids was the concurrent uptake of NH 4 + and amino acids that persisted as long as a readily assimilable carbon source (glucose in our case) was taken up. There was no net regeneration of NH 4 + , even though catabolism of amino acids occurred. Regeneration of NH 4 + was evident only after glucose was completely utilized, which usually occurred at the end of exponential growth. The contribution of15NH 4 + to total nitrogen uptake by the end of exponential growth varied from ~60 to 80% when individual amino acids were present and down to ~24% when the amino acid mixture was added. These estimates are conservative because we did not account for possible isotope dilution effects resulting from amino acid catabolism. When NH 4 + and glucose were the sole nitrogen and carbon sources, there was a stoichiometric balance between glucose and NH 4 + uptake over a wide range of C:Ns ratios, leading to a constant bacterial biomass C:N ratio (C:NB) of ~4.5:1. As a result NH 4 + usage varied from 50% when the C:Ns ratio was 3.6:1, to 100% when the C:Ns ratio was 10:1. Gross growth efficiency varied from ~60% when NH 4 + plus glucose were added alone or with the amino acid mixture, to 47% when the individual amino acids were used in place of the mixture. It is thus evident that actively growing bacteria will act as sinks for nitrogen when a carbon source that can be assimilated easily is available to balance NH 4 + uptake, even when amino acids are available and are being co-metabolized.  相似文献

Based on a series of short-term incubations involving the marine diatom Chaetoceros simplex (Bbsm), precultured in NH 4 + -, NO 3 - -and urea-limited continuous cultures at several dilution rates, we found that both the short-term specific rate of 14CO2 uptake and the amount of CO2 fixed after 8- and 16-min incubations were unaffected by enrichment with NH 4 + , urea, or NO 3 - when NH 4 + or urea were the preconditioning forms of N, but were slightly suppressed when the cells were first grown on NO 3 - . Similar enrichments in the dark, however, led to significant CO2 uptake under all conditions of NH 4 + enrichment and to similarly enhanced CO2 uptake, but only at high growth rates, when urea was the source of enrichment nitrogen. Our light results are contrary to some contemporary findings, but there does seem to be agreement that photosynthetic rates of rapidly growing phytoplankton will not be affected by exposure to pulses of nitrogen. Enhanced dark uptake, in contrast, appears to be characteristic of phytoplankton under all degrees of N limitation, and, as such, may be useful as an “all or nothing” index of the nitrogen status of natural waters. There is some indication that the index may be useful in determining both the form of and the degree of N limitation as well.  相似文献

This review examines and evaluates the literature on the ability of inorganic arsenic compounds to cause cancer in humans and laboratory animals. The epidemiological data that supports the position that inorganic arsenical derivatives are carcinogenic in humans is convincing and difficult to deny because of their consistency. These data are from studies of different occupational exposures such as smelter and pesticide workers, as well as from studies of drinking water, wines and medicinal tonics that contained or were contaminated with inorganic compounds of arsenic. Indeed, positive dose-response relationships between cancer incidence or mortality with many inorganic arsenical substances have been shown. Despite the presence of data which confuse the interpretation and evaluation of epidemiological data, associated neoplasms of the lungs, skin and gastrointestinal systems have been observed as a result of exposure to inorganic arsenic compounds.The mechanism of carcinogenicity of inorganic arsenical substances in humans is unknown. Inorganic arsenic compounds are not carcinogenic in laboratory animals by most routes of administration. However, further studies (subchronic, chronic, carcinogenic) using intratracheal and other conventional routes in other animal species would appear to be warranted. Moreso, especially since there is no evidence that organic arsenic compounds are carcinogenic in numerous mammalian species. Inorganic derivatives of arsenic are not mutagenic but may be teraiogenic. This latter conclusion is dependent on the method of administration and size of the dose, as well as on the species of animal used for the study.  相似文献
