The primary aim of our research was to investigate the applicability of activated sludge models (ASM) for aerobic thermophilic processes, especially autothermal thermophilic aerobic digestion (ATAD). The ASM3 model (Gujer et al., 1999) theoretically seems to be the most suitable, because storage plays an important role in a batch-feed cycle system like ATAD. The ASM3 model was extended with an activation step of the thermophilic organisms. This model was calibrated and verified by independent test results, demonstrating its ability to describe the process. The growth (microH = 26.04 day(-1)), storage (k(STO) = 20.39 day(-1)), hydrolysis (kH = 11.15 day(-1)) and decay rates (b 9H,O2) = 1.28 day(-1), b(STO,O2 = 1.10 day-1)) obtained from calibration are significantly higher at 55 degrees C than at mesophilic temperatures, justifying the faster metabolism at higher temperatures. An inert fraction of the biomass (characterized by the model parameter f(i) = 0.4) was found to be significantly greater than in the mesophilic case. The development of process-based models to estimate ammonia emissions from animal feeding operations (AFOSs) is sought to replace costly and time-consuming direct measurements. Critical to process-based model development is conducting sensitivity analysis to determine the input parameters and their interactions that contribute most to the variance of the model output. Global and relative sensitivity analyses were applied to a process-based model for predicting ammonia emissions from the surface of anaerobic lagoons for treating and storing manure. The objectives were to compare global sensitivity analysis (GSA) to relative (local) sensitivity analysis (RSA) on a process-based model for ammonia emissions. Based on the first-order coefficient, both GSA and RSA showed the model input parameters in order of importance in process model for ammonia emissions from lagoon surfaces were: (i) pH, (ii) lagoon liquid temperature, (iii) wind speed above the lagoon surface, and (iv) the concentration of ammoniacal nitrogen in the lagoon. The GSA revealed that interactions between model parameters accounted for over two-thirds of the model variance, a result that cannot be achieved using traditional RSA. Also, the GSA showed that parameter interactions involving liquid pH had more impact on the model output variance than the single parameters: (i) temperature, (ii) wind speed, or (iii) total ammoniacal nitrogen. This study demonstrates that GSA provides a more complete analysis of model input parameters and their interactions on the model output compared to RSA. The neutral red retention assay has been proposed to determine the lysosomal membrane stability in immune cells. Nevertheless, this assay implies many examinations under a microscope at short time intervals and therefore the analysis of few samples. The present study proposes two more rapid, efficient, and sensitive sample analyses using flow cytometry method. Lysosomal presence and lysosomal membrane integrity (LMI) were evaluated on the three-spined stickleback, Gasterosteus aculeatus (L.), a well-described model fish species for aquatic ecotoxicology studies. After development of the two biomarkers, they were validated by ex vivo contamination with endosulfan and copper and by in situ sampling. These immunomarkers were clearly modulated by pollutants and their variations seemed to be correlated with leucocyte mortality. Thus, from a practical point of view, lysosomal presence and LMI may provide novel and efficient means of evaluating immune capacities and indicating the toxic effects of environmental pollution. 相似文献

There is general consensus that carbon (C) sequestration projects in forests are a relatively low cost option for mitigating climate change, but most studies on the subject have assumed that transaction costs are negligible. The objectives of the study were to examine transaction costs for forest C sequestration projects and to determine the significance of the costs based on economic analyses. Here we examine four case studies of active C sequestration projects being implemented in tropical countries and developed for the C market. The results from the case studies were then used with a dynamic forest and land use economic model to investigate how transaction costs affect the efficiency and cost of forest C projects globally. In the case studies transaction costs ranged from 0.38 to 27 million US dollars ($0.09 to $7.71/t CO2) or 0.3 to 270 % of anticipated income depending principally on the price of C and project size. The three largest cost categories were insurance (under the voluntary market; 41–89 % of total costs), monitoring (3–42 %) and regulatory approval (8–50 %). The global analysis indicated that most existing estimates of marginal costs of C sequestration are underestimated by up to 30 % because transaction costs were not included. 相似文献
