Transcervical cell (TCC) samples have been shown to contain fetal cells amenable to molecular analysis. However, the presence of ‘contaminating’ maternal cells limits their use for prenatal diagnoses. In this report we show that clumps of fetal cells can be isolated from transcervical samples by micromanipulation and tested by fluorescence in situ hybridization (FISH) and polymerase chain reaction (PCR). Out of 129 clumps, isolated from mucus aspirates and transcervical lavages from 29 patients, 29 clumps from 11 patients were found to be exclusively of fetal origin as judged by the detection of chromosome 21-specific polymorphic DNA markers and Y-derived DNA sequences by PCR and FISH. One case of a male triploid fetus, diagnosed by the analysis of TCC samples obtained by mucus aspiration and lavage, was confirmed by testing clumps of cells isolated by micromanipulation.  相似文献

This paper reports a case of chromosomal mosaicism for trisomy 5 recovered from amniotic fluid cells and from skin fibroblasts of a liveborn dysmorphic male. Routine amniocentesis was performed at 16 weeks' gestation because of parental concern. Trisomy 5 cells were measured from 25 per cent of amniocytes from two culture vessels. No further invasive testing was performed until 32 weeks' gestation, at which time ultrasound examination showed fetus with intrauterine growth retardation. Fetal blood sampling was then performed, with only karyotypically normal cells recovered. At birth, the child was found to have multiple dysmorphic features and congenital anomalies, including an eventration of the diaphragm and ventricular septal defect, both of which required surgical correction. Chromosomal analysis of cord blood lymphocytes indicated 46,XY; however, 20 per cent of the cultured fibroblasts obtained from the chest skin at the incision site for diaphragmatic repair had a 47,XY,+5 karyotype. Trisomy 5 mosaicism may be another example of tissue-limited mosaicism. Fetal blood sampling can then be falsely reassuring. Furthermore, because some cell lines rarely appear in lymphocyte populations, cytogenetic analysis of multiple tissues warranted as part of the evaluation of individuals with developmental delay and dysmorphic features.  相似文献

Drawing on gender role theory and tournament theory, we examined the effects of sex and organizational culture preferences on the incomes of MBA graduates over an 8‐year period. We found that masculine culture preferences led to higher income 4 years after graduation and, in contrast to previous research, the effect was stronger for women. By 8 years after graduation, however, men's rate of income increase was significantly higher than women's, an effect that was mediated by hours worked per week. These findings clarify some of the conflicting results of previous research on the effects of gender roles on women's careers and suggest that a tournament‐like aspect of careers may account for higher incomes in organizations. The effects of maize (Zea mays L.), genetically modified to express the Cry1Ab protein (Bt), and an insecticide on soil microbial and faunal communities were assessed in a glasshouse experiment. Soil for the experiment was taken from field sites where the same maize cultivars were grown to allow comparison between results under glasshouse conditions with those from field trials. Plants were grown in contrasting sandy loam and clay loam soils, half were sprayed with a pyrethroid insecticide (deltamethrin) and soil samples taken at the five-leaf stage, flowering, and maturity. The main effect on all measured parameters was that of soil type and there were no effects of Bt trait or insecticide on plant growth. The Bt trait resulted in more soil nematodes and protozoa (amoebae), whereas insecticide application increased plant Bt concentration and altered nematode community structure. The only significant effects on soil microbial community structure, microarthropods, and larvae of a nontarget root-feeding Dipteran, were due to soil type and plant growth stage. The results indicate that, although there were statistically significant effects of the Bt trait on soil populations, they were small. The relative magnitude of the effect could best be judged by comparison with the insecticide treatment, which was representative of current best practice. The Bt trait had no greater effect than the insecticide treatment. Results from this glasshouse experiment were in broad agreement with conclusions from field experiments using the same plant material grown in the same soils.  相似文献

Repeated manure application can lead to excessive soil test P (STP) levels and increased P concentration in runoff, but also to improved water infiltration and reduced runoff. Research was conducted to evaluate soil P tests in prediction of P concentration in runoff and to determine the residual effects of composted manure on runoff P loss and leaching of P. The research was conducted from 2001 to 2004 under natural runoff events with plots of 11-m length. Low-P and high-P compost had been applied during the previous 3 yr, resulting in total applications of 750 and 1150 kg P ha(-1). Bray-P1 in the surface 5 cm of soil was increased from 16 to 780 mg kg(-1) with application of high-P compost. Runoff and sediment losses were 69 and 120% greater with no compost than with residual compost treatments. Runoff P concentration increased as STP increased, but much P loss occurred with the no-compost treatment as well. Agronomic soil tests were predictive of mean runoff P concentration, but increases in STP resulted in relatively small increases in runoff P concentration. Downward movement of P was not detected below 0.3 m. In conclusion, agronomic soil tests are useful in predicting long-term runoff P concentration, and risk of P loss may be of concern even at moderate soil P levels. The residual effect of compost application in reducing sediment and runoff loss was evident more than 3 yr after application and should be considered in P indices.  相似文献

Denitrification is a process that reduces nitrogen levels in headwaters and other streams. We compared nirS and nirK abundances with the absolute rate of denitrification, the longitudinal coefficient of denitrification (i.e., Kden, which represents optimal denitrification rates at given environmental conditions), and water quality in seven prairie streams to determine if nir-gene abundances explain denitrification activity. Previous work showed that absolute rates of denitrification correlate with nitrate levels; however, no correlation has been found for denitrification efficiency, which we hypothesise might be related to gene abundances. Water-column nitrate and soluble-reactive phosphorus levels significantly correlated with absolute rates of denitrification, but nir-gene abundances did not. However, nirS and nirK abundances significantly correlated with Kden, as well as phosphorus, although no correlation was found between Kden and nitrate. These data confirm that absolute denitrification rates are controlled by nitrate load, but intrinsic denitrification efficiency is linked to nirS and nirK gene abundances.  相似文献

Present methods for the determination of carbon monoxide are discussed including indicator tubes, the iodine pentoxide reaction and measurement by gas chromatography. In the gas chromatographic method an air sample is separated on a gas-solid chromatogra-phic column and the separated CO is converted to methane by hydrogenation at elevated temperature. The separated CO, in the form of methane, is passed into a hydrogen flame detector and measured. The conversion from CO to methane allows the use of a sensitive ionization detector in place of the thermal conductivity cell which is insufficiently sensitive for the measurement of trace amounts of CO. The optimum operating conditions are discussed. It is possible to determine one ppm CO in air. The iodine pentoxide reaction with CO has been combined with electrometric measurement. The iodine liberated is passed into a Ditte cell and the current generated is measured by an electrometerrecorder combination. This method is continuously direct reading with a permanent record. It is suitable for the continuous routine analysis of one ppm CO.  相似文献
