Early signalling events that control the process of heavy metal-induced cell death are largely unknown in plants. In mammals protein tyrosine phosphorylation plays an important role in the activation of programmed cell death. We thus examined the involvement of tyrosine phosphorylation in Cu-induced rice cell death. This investigation demonstrates that Cu induces cell death and DNA fragmentation in rice root cells. In the presence of Cu, the level of phosphotyrosine accumulation declined in the band of 45 kDa, p45. To analyze the role of tyrosine dephosphorylation for the regulation of Cu-induced cell death more precisely, we increased levels of tyrosine phosphorylation using the protein tyrosine phosphatase inhibitor, sodium orthovanadate (Na(3)VO(4)). Treatment of rice roots with Na(3)VO(4) blocked Cu-induced cell death and protein tyrosine dephosphorylation. In addition, the antioxidant GSH and the calcium chelator EGTA significantly abolished Cu-induced cell death and protein tyrosine dephosphorylation. These results provide evidence that dephosphorylation of a tyrosine-phosphorylated protein, p45, is an important step in the Cu-triggered signalling transduction pathway.  相似文献

Chitosan is a natural high molecular polymer made from crab, shrimp and lobster shells. When used as coagulant in water treatment, not like aluminum and synthetic polymers, chitosan has no harmful effect on human health, and the disposal of waste from seafood processing industry can also be solved. In this study the wastewater from the system of cleaning in place (CIP) containing high content of fat and protein was coagulated using chitosan, and the fat and the protein can be recycled. Chitosan is a natural material, the sludge cake from the coagulation after dehydrated could be used directly as feed supplement, therefore not only saving the spent on waste disposal but also recycling useful material. The result shows that the optimal result was reached under the condition of pH 7 with the coagulant dosage of 25 mg/l. The analysis of cost-effective shows that no extra cost to use chitosan as coagulant in the wastewater treatment, and it is an expanded application for chitosan.  相似文献

铜绿微囊藻菌CCAP1450/4在室内人工培养的结果表明,较大的无机培养液表面、连续适当光照20uE/(m2·s)及适宜的温度(25℃)有利于该藻菌的生产.通过超声波击碎细胞,离心过滤C18净化柱分离,得到藻毒素.应用高效液相色谱分离主峰纯化藻毒素藻毒素在紫外光谱238nm处出现吸收高峰表明,铜绿微囊藻菌CCAP1450/4中藻毒素的主要成分是藻毒素LR.本研究还探索了简便、快速人工培养铜绿微囊藻菌及高效、叶靠分离纯化藻毒素LR的方法.  相似文献

农药化工厂生产苯肼、苯唑醇、乙基氯化物过程排放的废水是高氯难生物降解有机废水,采用三辛胺作萃取剂,用液-液萃取处理,三辛胺与水中Cl-离子形成萃合物而使Cl-转移到有机相.再经高效絮凝处理后,CODCr总去除率达89.8%,Cl-总去除率达83.2%,BOD/COD比从0.02上升到0.34,可生化性大幅度提高. 废水再经河水稀释进A/O池生化处理3d后,可达标排放.负载萃取液用5%NaOH水溶液反萃取.由于萃取剂回用降低了处理费用.  相似文献

研究垃圾组分PVC在焚烧过程中HCl的排放和脱除特性的实验在准等温、电加热的水平石英管反应器中进行.随着床温从700℃升高到900℃,HCl的转化率从82.5%平缓增加到88.3%,而过量空气系数的增加促使部分HCl转化为C12(1.9%~4.5%)的形式存在.钙基吸收剂CaCO3,Ca(OH)2,Ca(CH3COO)2对HCl的脱除效率高达68%~79%,而镁基吸收剂的脱氯效率却低于3%.小的吸收剂粒径、高的Ca/Cl比会促进脱氯效率的提高;烟气中CO2和H2O对脱氯效率的影响与它们在反应平衡中所起作用有关.高温下燃烧固氯机理反应的平衡常数的计算不仅诠释了实验结果,而且提供了脱氯吸收剂选择的依据.  相似文献

The mantle of the pearl oyster Pinctada fucata was adopted for the proliferation profile study in our work and a proliferation hot spot was found in the outer epithelia of mantle central zone using the BrdU immunohistochemistry method. This result contradicts the previous research that the mantle has numerous growth centers all over the mantle epithelium, with the same proliferation activity throughout the whole mantle outer epithelial cells. This is the first report on the different proliferation features on the whole mantle where Alcian Blue/PAS staining analysis and ultrastructural observation with the aid of transmission electron microscope (TEM) demonstrated distinct features of the epithelium in four different regions of the mantle. Results from the present investigation displayed that in the outer epithelium of the marginal zone in mantle outer fold, organelles such as mitochondria and endoplasmic reticulum (ER) were well-developed and double membrane bounded vesicles were present; in the outer epithelia of mantle central zone, stem-like cells with a high ratio of nucleus to cytoplasm and comparatively undeveloped organelles were detected. Together with the observations of the cell proliferation profile of different regions of the mantle, a hypothetic model for the proliferation and differentiation of the pearl oyster’s mantle is proposed: there exists a proliferation “hot spot” in the outer epithelial cells of central zone and the proliferation ability decreases progressively from this “hot spot” towards the marginal zone; the whole mantle’s differentiation occurs continuously with its growth and the direction is from the proliferation ‘hot spot’ (central zone) towards the marginal zone. Furthermore, another interesting result was found when the proliferation rate was investigated together with the tidal rhythm: the proliferation activity was found to be closely correlated with the tidal rhythm, indicating that the mantle outer epithelia’s proliferation rhythm might be the impetus of the shell’s daily growth bands.  相似文献

采集渤海海河河口水,在实验室受控条件下,研究了营养盐和光照条件对邻苯二甲酸二丁酯(DBP)生物降解的影响。实验结果表明:DBP在海河河口水中的生物降解符合一级动力学,微生物降解占总去除率的99%左右,微生物降解是DBP降解的主要途径。高营养水平时,DBP在光照、光暗条件下的生物降解速率常数分别为0.32d-1和0.41d-1;低营养水平时,DBP在光照、光暗条件下的生物降解速率常数分别为0.17d-1和0.20d-1。高营养条件下较高的微生物总量使DBP的生物降解速率明显大于相对低营养条件下的降解速率;在相同的营养条件下,光照能够改变水体中藻、菌的微生态结构,从而延迟DBP的降解。  相似文献

根据气泡与絮体的粘附理论和紊流气浮理论,设计了共凝聚气浮反应器的试验装置,并通过对洗衣废水进行试验,确定运行参数,考察设备性能,对设备进行改进.混凝剂与原水在输水管线中混合,混凝过程在气浮反应池中进行.溶气水释放的微气泡在微絮体的空隙内释放长大,与微絮体形成共聚复合体,在上浮过程中起到了“核”的作用,提高了絮体的上浮速度,使气浮达到了最优的去除效果.该设备采用了气液混合泵溶气,取代了传统气浮装置中的空压机、溶气罐和释放器,减小了占地面积,节省了投资费用.  相似文献
