Mutualistic interactions with fungal endophytes and dinitrogen-fixing bacteria are known to exert key biological influences on the host plant. The influence of a fungal endophyte alkaloid on the toxicity of a plant has been documented in Oxytropis sericea. Oxytropis sericea is a perennial legume responsible for livestock poisoning in western North America. Livestock poisoning is attributed to the alkaloid swainsonine, which is synthesized inside the plant by the fungal endophyte Embellisia sp. In this study, the ability of Oxytropis sericea to form a dinitrogen-fixing symbiosis with Rhizobium and the effects of this symbiosis on the production of swainsonine by Embellisia sp. were evaluated in a greenhouse environment. Seeds of O. sericea were grown in plastic containers. Twenty-week-old O. sericea seedlings were inoculated with four strains of Rhizobium. Twenty weeks after inoculation, plant growth and root nodulation by Rhizobium were measured. Dinitrogen fixation was confirmed using an acetylene reduction assay (ARA) on excised root nodules. Dry leaves were analyzed for swainsonine content. A second set of plants was treated with fungicide to evaluate the effect of reduced fungal endophyte infection on plant growth and swainsonine production. All inoculated plants produced indeterminate nodules. The ARA indicated that 98% of the excised nodules were fixing dinitrogen. Rhizobium-treated plants had greater swainsonine concentrations than the non-inoculated controls. Plants that established from seeds treated with fungicide had lower biomass than non-fungicide-treated controls and plants treated with foliar fungicide. Plants treated with foliar fungicide and the controls had greater swainsonine concentrations than the plants that received seed fungicide. This greenhouse study is the first report of nodulation and dinitrogen fixation in O. sericea. It also demonstrates that dinitrogen fixation increases the production of swainsonine in O. sericea plants infected with Embellisia sp. Results from this study suggest that dinitrogen fixation affects swainsonine production and has the potential to support the symbiosis between Embellisia sp. and O. sericea when soil nitrogen is limited. Oxytropis sericea competitiveness appears to be facilitated by an ability to simultaneously associate with Rhizobium and a fungal symbiont. 相似文献

Eelgrass, Zostera marina, is generally regarded as the preferred habitat of bay scallops, but in some cases scallop populations have persisted or
increased in areas lacking eelgrass. This suggests that some other substrate(s) may serve important ecological functions for
bay scallops. One candidate is Codium fragile, a macroalgal species with which bay scallops are known to associate and in which we commonly find juvenile and adult bay
scallops in eastern Long Island, New York. In this study, we examined whether survival of planted bay scallops differed in
Codium, eelgrass, and Codium + eelgrass substrates at two sites during August and October of 2 years. Survival of tethered scallops and recoveries of
live free-planted individuals varied with scallop size, planting season and year, but no differences were observed between
the three substrates for a given scallop size and planting date. Crab (particularly Dyspanopeus
sayi) and whelk predation were implicated as important causes of tethered scallop mortalities while emigration and removal by
predators likely contributed to scallop losses. Densities of naturally recruited 0+ years scallops recovered by visual and
suction dredge sampling were similar in the eelgrass and Codium substrates. While our results suggest that Codium may offer some degree of predation refuge for bay scallops, further work needs to weigh the potential disadvantages of this
substrate (such as low DO levels, potential attachment and transport of scallops, and differences in current flow, food availability
and sedimentation relative to eelgrass) to determine if Codium may serve as a valuable habitat for bay scallops throughout their lifespan. 相似文献

We have developed and applied a process-based model, the Wetland Ecosystem Model (WEM), to evaluate the effects of a prescribed fire on the phosphorus (P) dynamics and cattail (Typha domingensis) growth in a P-enriched area in the Florida Everglades. The WEM couples major ecosystem processes including carbon (C), nitrogen (N) and P biogeochemical cycles, plant growth, hydrology, and fire disturbance. The model is used to assess the effects of a prescribed fire on P dynamics and cattail growth through dynamic interaction among four modules: fire, water chemistry, soil, and vegetation. The simulation results are in agreement with observed data including cattail above- and belowground biomass and dead mass, P concentration in surface-water, pore-water, and soil, and soil and water temperature. Cattail aboveground biomass reached the unburned level one year after burn; belowground biomass recovered to unburned level one and half years after the fire, however, dead mass did not completely reach unburned level two years after fires. The fire increased water and soil temperatures in the short term, while indirectly increasing the sensitivity of water and soil temperature post-fire response to air temperature by altering the energy exchange between air and water through a canopy gap created by fire. The fire also altered the P dynamics in surface-water and pore-water. A post-fire P pulse that lasted for less than one month was observed in surface-water. A similar P pulse, but in a small magnitude and a longer duration, was also observed in the pore-water total phosphorus (TP), and then came back to normal level after approximately three months. No significant changes in soil TP was observed during the study period. Meanwhile, no significant changes in water nutrients were observed downstream of the study plot. This finding indicated that the P-enriched wetlands in Everglades act as a buffer in regulating the P concentration in surface-water. Our study showed that the distance of fire effects on a 300 m × 300 m plot was less than 300 m downstream. Sensitivity analysis identified that the air temperature and hydrological conditions are two important driving factors which may alter the cattail community dynamics in response to prescribed fires. This paper discusses the need for critically evaluating regional-scale (~200–2,000 km) three-dimensional numerical photochemical air quality modeling systems to establish a model’s credibility in simulating the spatio-temporal features embedded in the observations. Because of limitations of currently used approaches for evaluating regional air quality models, a framework for model evaluation is introduced here for determining the suitability of a modeling system for a given application, distinguishing the performance between different models through confidence-testing of model results, guiding model development and analyzing the impacts of regulatory policy options. The framework identifies operational, diagnostic, dynamic, and probabilistic types of model evaluation. Operational evaluation techniques include statistical and graphical analyses aimed at determining whether model estimates are in agreement with the observations in an overall sense. Diagnostic evaluation focuses on process-oriented analyses to determine whether the individual processes and components of the model system are working correctly, both independently and in combination. Dynamic evaluation assesses the ability of the air quality model to simulate changes in air quality stemming from changes in source emissions and/or meteorology, the principal forces that drive the air quality model. Probabilistic evaluation attempts to assess the confidence that can be placed in model predictions using techniques such as ensemble modeling and Bayesian model averaging. The advantages of these types of model evaluation approaches are discussed in this paper. 相似文献

Chambers are commonly used to measure the emission of many trace gases and chemicals from soil. An aerodynamic (flow through) chamber was designed and fabricated to accurately measure the surface flux of trace gases. Flow through the chamber was controlled with a small vacuum at the outlet. Due to the design using fans, a partition plate, and aerodynamic ends, air is forced to sweep parallel and uniform over the entire soil surface. A fraction of the air flowing inside the chamber is sampled in the outlet. The air velocity inside the chamber is controlled by fan speed and outlet suction flow rate. The chamber design resulted in a uniform distribution of air velocity at the soil surface. Steady state flux was attained within 5 min when the outlet air suction rate was 20 L/min or higher. For expected flux rates, the presence of the chamber did not affect the measured fluxes at outlet suction rates of around 20 L/min, except that the chamber caused some cooling of the surface in field experiments. Sensitive measurements of the pressure deficit across the soil layer in conjunction with measured fluxes in the source box and chamber outlet show that the outflow rate must be controlled carefully to minimize errors in the flux measurements. Both over- and underestimation of the fluxes are possible if the outlet flow rate is not controlled carefully. For this design, the chamber accurately measured steady flux at outlet air suction rates of approximately 20 L/min when the pressure deficit within the chamber with respect to the ambient atmosphere ranged between 0.46 and 0.79 Pa. 相似文献

The surface chemical characteristics of activated carbon treated by ozone and alkaline are determined in terms of surface functional groups and surface acidity. Surface functional groups are analyzed by the IR spectroscopic method and Boehm's titration technique. The surface acidity of activated carbon is determined by electrophoretic mobility measurements. The oxygen concentration of activated carbon increases upon ozone and NaOH treatment. South Australian freshwaters from a wide variety of environments were analysed for bromide and the results correlated with both chloride and total dissolved solids (TDS) concentrations. A linear relationship was obtained which allows chloride data to be successfully used to estimate bromide concentrations. Bromide displayed a slightly better correlation with TDS indicating that an improved estimate of bromide could be made by reference to TDS data which is more easily and commonly obtained, and generally available extensively as historical data. The bromide content in relation to its ratio with both chloride and TDS contents was around seventy percent of the corresponding ratios found in seawater, a finding reported in other published data. The ability to estimate bromide concentrations is a potentially useful tool in the drinking water industry as it enables the assessment of the extent of bromate formation (predicted through the use of mathematical models and other water quality data) which is an important factor when ozonation is being considered as a treatment option, particularly as many water industry regulatory bodies have imposed stringent limits on the levels of acceptable bromate. 相似文献

This study investigates the distribution of three trace elements (Cd, Cu and Zn) in tissues (exoskeleton, abdominal muscle and hepatopancreas) of two shrimp species (Penaeus merguiensis and Penaeus semisulcatus). Special emphasis in this study was placed on evaluating the effects of storage conditions on redistribution of the elements and moreover the potential roles of metallothionein in tissue redistribution. The specimens were collected from the northern part of Persian Gulf during September 2001 and January 2002. The concentrations of metals in the tissues were measured using an Inductively Coupled Plasma-Atomic Emission Spectrophotometer (ICP-AES) and an ICP-Mass Spectrometer (ICP-MS). Metallothionein levels were determined using Differential pulse polarography. Results show a similar hierarchy for Cd and Cu concentrations among different tissues as follows: hepatopancreas>exoskeleton>muscle; and for Zn, the hepatopancreas again contained the highest levels, though there were not any significant differences in Zn concentrations between muscle and exoskeleton. Significant differences were found between the species in the case of Cd and Zn concentrations. Cd and Zn levels in P. semisulcatus were markedly higher than in P. merguiensis. In general, there were highly significant differences among the storage conditions from accumulation the selected trace elements and metallothionein in both species point of view, but in most cases, no significant differences could be observed among the fresh, refrigerated and frozen specimens of P. semisulcatus. In regards to Zn and Cu accumulation in edible (muscle) tissues of P. merguiensis, optimum conditions for frozen storage is -30 degrees C as ice-frozen, and in terms of short-term storage, 1-day refrigeration is recommended. The redistribution of Zn and Cu during the storage period can be attributed to the role of metallothionein in their transport from organ(s) to muscle during storage. The variations in pH values measured in the muscle of P. merguiensis in different storage conditions showed a similar trend with metallothionein, Zn and Cu. In contrast, it appears that cadmium is transported from muscle to the other tissues. Exoskeleton is probably one of the recipient tissues of cadmium. The inter-elemental relationships were assessed for each species, separately. A significant positive relationship was observed between the concentrations of Zn and metallothionein in P. semisulcatus, whereas in P. merguiensis, Cd concentrations decreased significantly with increasing metallothionein. Highly significant differences between the sexes in Cu and Zn contents could be detected. The differences between the sexes in the levels of Zn and Cu were significantly greater in P. semisulcatus. Highly significant size-related differences were found in the levels of Cd, Cu and metallothionein in the selected tissues of the shrimp species. Comparison between the mean concentrations of the elements in muscle tissue and existing guidelines indicate that in nearly all cases, the concentrations were well below permissible levels for human consumption. The objectives of this study were to estimate the prevalence of children's exposure to passenger air bag (PAB) deployments and to determine the relative risk of both minor and more serious nonfatal injuries to restrained children exposed to PABs in frontal impact collisions. Data were collected from 1 December 1998 to 30 November 2001 from a large-scale, child-specific crash surveillance system based on insurance claims, a telephone survey, and on-site crash investigations. Vehicles qualifying for inclusion were State Farm-insured, model year 1990 or newer, and involved in a crash with at least one child occupant < or =15 years of age. Qualifying crashes were limited to those that occurred in 15 states and the District of Columbia. A stratified cluster sample was designed in order to select vehicles (the unit of sampling) for the conduction of a telephone survey with the driver. For cases in which child occupants were seriously injured or killed, in-depth crash investigations were performed. The prevalence of exposure to PABs was calculated as the number of children occupying the right front seat in a PAB deployment crash among all children occupying the right front seat in vehicles equipped with PABs. Complete interview data were obtained on 9,779 vehicles involving 15,341 children. Among PAB-exposed children, 175 (14%) suffered serious injuries versus 41 (7.5%) of those in the comparison group (OR 2.0; 95% CI, 1.1-3.7). The overall risk of any injury (both minor and serious) was 86% among children exposed to PABs, compared to 55% among the comparison group (OR 5.3; 95% CI, 2.1-13.4). Exposure to PABs increased the risk of both minor injuries, including facial and chest abrasions, and more serious injuries, particularly upper extremity fractures. 相似文献
