Three projects involving point velocity probes (PVPs) illustrate the advantages of direct groundwater velocity measurements. In the first, a glacial till and outwash aquifer was characterized using conventional methods and multilevel PVPs for designing a bioremediation program. The PVPs revealed a highly conductive zone that dominated the transport of injected substances. These findings were later confirmed with a natural gradient tracer test. In the second, PVPs were used to map a groundwater velocity field around a dipole recirculation well. The PVPs showed higher than expected velocities near the well, assuming homogeneity in the aquifer, leading to improved representations of the aquifer heterogeneity in a 3D flow model, and an improved match between the modelled and experimental tracer breakthrough curves. In the third study, PVPs detected subtle changes in aquifer permeability downgradient of a biostimulation experiment. The changes were apparently reversible once the oxygen source was depleted, but in locations where the oxygen source lingered, velocities remained low. PVPs can be a useful addition to the hydrogeologist's toolbox, because they can be constructed inexpensively, they provide data in support of models, and they can provide information on flow in unprecedented detail.  相似文献

Acetylcholinesterase (AChE) activity was analyzed as a molecular marker indicative of exposure to organophosphorus pesticide residues in individuals of the endemic clam species Semele solida in selected coastal locations of Chile's VIII Region. AChE activity was assayed in clams from (i) Penco Beach near the mouth of the Andalién River (Concepción Bay), (ii) Lenga Beach, near the mouth of the Lenga Estuary (San Vicente Bay), and (iii) Coliumo Beach, near the mouth of the Coliumo Estuary (Coliumo Bay). We also analyzed variations in protein content of clam hemolymph, and variability in the activity of AChE in relation to the sizes of the individual clams sampled. Collection of the clams was done using routine methods, during the spring of 2005, the period during which the use of pesticides is typically intensified in the surrounding forestry and agriculture. The results showed no significant correlation of AChE activity with either the size of the clam, or with the concentration of proteins in the hemolymph. The lowest AChE activity was observed in clams from mouth of the Andalién River (187.5 +/- 34.9 Umin(-1)) which was significantly less than that measured in specimens collected near the mouth of the Coliumo Estuary and the mouth of the Lenga Estuary. A very close relation was observed between the degree of anthropogenic inputs, ocean dynamics, and alterations in AChE activity in S. solida. This clam appears to be a useful indicator species, and AChE activity a sensitive marker for the presence of xenobiotics.  相似文献

DOI: 10.1111/j.1752-1688.2009.00407.x Abstract: Wetlands provide many critical functions in urban ecosystems, including habitat for wetland-dependent fauna and enhancement of water quality. Interest in restoring or creating wetlands to enhance these functions is increasing due to the scale and extent of wetland loss and water quality problems associated with urbanization. One of the most pressing questions associated with urban wetland restoration is the extent to which urban wetlands tend to concentrate contaminants, and if so, whether an associated risk to wildlife exists. The goal of this study was to better understand these potential risks, and the associated tradeoffs with using wetlands to treat urban runoff. Sediment toxicity, contaminant chemistry, and macroinvertebrate (MI) community metrics were measured in 21 southern California wetlands that receive urban runoff as their primary water source. MI organisms in 18 of the 21 urban wetlands examined were considered to be at risk due to sediment contaminant concentrations and toxicity. Most of the sites were either toxic to the amphipod Hyalella azteca, exceeded a sediment quality guideline, or both. Sediment chemistry and toxicity identification evaluation studies suggest that pyrethroid pesticides may have been responsible for much of the toxicity documented in this study. The mean Probable Effects Concentration quotient (an index of degree of sediment contamination) was found to negatively correlate with MI diversity in these wetlands suggesting that toxicity was affecting organisms at the base of the food chain in these wetlands.  相似文献

Background, Goals and Scope In response to concerns that have been raised about chemical substances that may alter the function of endocrine systems and result in adverse effects on human health, an OECD initiative was undertaken to develop and validate in vitro and in vivo assays to identify chemicals that may interfere with endocrine systems of vertebrates. Here we report on studies that were conducted to develop and standardize a cell-based screening assay using the H295R cell line to prioritize chemicals that may act on steroidogenic processes in humans and wildlife. These studies are currently ongoing as part of the ‘Special Activity on the Testing and Assessment of Endocrine Disruptors’ within the OECD Test Guidelines Program to review, develop, standardize, and validate a number of in vitro and in vivo toxicological assays for testing and assessment of chemicals concerning their potential to interact with the endocrine system of vertebrates. Study Design Six laboratories from five countries participated in the pre-validation studies. Each laboratory tested the effects of three model chemicals on the production of testosterone (T) and estradiol (E2) using the H295R Steroidogenesis Assay. Chemicals tested were well described inducers or inhibitors of steroidogenic pathways (forskolin, prochloraz and fadrozole). All experiments were conducted in 24 well plates following standard protocols. Six different doses per compound were analyzed in triplicate per plate. A quality control (QC) plate was run in conjunction with the chemical exposure plate to account for inter-assay variation. Each chemical exposure was conducted two or three times. Results All laboratories successfully detected increases and/or decreases in hormone production by H295R cells after exposure to the different model compounds and there was good agreement in the pattern of response for all groups. Forskolin increased both T and E2 while fadrozole and prochloraz decreased production of both hormones. All chemicals affected hormone production in a dose-dependent manner with the exception of fadrozole which caused maximum inhibition of E2 at the two least concentrations tested. Some inter-laboratory differences were noted in the alteration of hormone production measured in chemically exposed cells. However, with the exception of the production of T measured at one laboratory in cells exposed to forskolin, the EC50s calculated were comparable (coefficients of variation 34–49%) for all hormones. Discussion and Perspectives The results indicated that the H295R Steroidogenesis Assay protocol was robust, transferable and reproducible among all laboratories. However, in several instances that were primarily related to one laboratory there were unexplained minor uncertainties related to the inter-laboratory hormone production variation. Based on the findings from this Phase 2 prevalidation study, the H295R Steroidogenesis Assay protocol is currently being refined. The next phase of the OECD validation program will test the refined protocol among the same group of laboratories using an extended set of chemicals (∼30) that will include positive and negative chemical controls as well as a broad spectrum of different potential inducers and inhibitors of steroidogenic pathways. Three Lagrangian experiments were conducted during IGAC's second aerosol characterization experiment (ACE-2) in the area between Portugal, Tenerife and Madeira in June/July 1997. During each Lagrangian experiment, a boundary layer air mass was followed for about 30 h, and the temporal evolution of its chemical and aerosol composition was documented by a series of vertical profiles and horizontal box pattern flown by the Meteorological Research Flight research aircraft Hercules C130. The wealth of observational data that has been collected during these three Lagrangian experiments is the basis for the development and testing of a one-dimensional Lagrangian boundary layer model with coupled gas, aqueous, and aerosol phase chemistry. The focus of this paper is on current model limitations and strengths. We show that the model is able to represent the dynamical and chemical evolution of the marine boundary layer, in some cases requiring adjustments of the subsidence velocity and of the surface heat fluxes. Entrainment of a layer rich in ozone and carbon monoxide from a residual continental boundary layer into the marine boundary layer as well as in-cloud oxidation of sulphur dioxide by hydrogen peroxide are simulated, and coherent results are obtained, concerning the evolution of the small, presumably sulphate–ammonia aerosol mode.  相似文献

Given its geographic characteristics, the southern Chilean fjord area is subjected to growing environmental pressure from the development of diverse forms of aquaculture (i.e., fish, algae, shellfish). The sediments accumulate substances as a natural sink, and ecotoxicology assays offer a reliable and robust proxy for sediment quality analyses. This study's objective was to establish a mid-range toxicity base line for the sediments in the region by applying a battery of non-specific ecotoxicological assays. Sediment samples (28) were collected in the channels and fjords studied during the CIMAR-Fiordos 11 cruise (July 2005). The sediments were evaluated using different species endemic to the eastern Pacific as targets: Ampelisca araucana, Tisbe longicornis, Arbacia spatuligera, and Dunaliella tertiolecta. The conditions for each assay were reported previously. Of the four species used as ecotoxicological tools, only D. tertiolecta differed significantly from the control group (negative) in terms of its growth. This difference could be attributed to nutrient enrichment. In general, we concluded that, although local changes occurred in the sediments, the mesoscale magnitude of the ecotoxicological alterations was small. Nonetheless, a surveillance program should be implemented that would allow us to follow-up and analyze the changes that are taking place in the systems on broader scales of time and space.  相似文献
