A polyphasic approach based on cultivation and direct recovery of 16S rRNA gene sequences was utilized for microbial characterization of an aquifer contaminated with chlorinated ethenes. This work was conducted in order to support the evaluation of natural attenuation of chlorinated ethenes in groundwater at Area 6 at Dover Air Force Base (Dover, DE). Results from these studies demonstrated the aquifer contained relatively low biomass (e.g. direct microscopic counts of < 10(7) bacteria/g of sediment) comprised of a physiologically diverse group of microorganisms including iron reducers, acetogens, sulfate reducers, denitrifiers, aerobic and anaerobic heterotrophs. Laboratory microcosms prepared with authentic sediment and groundwater provided direct microbiological evidence that the mineralization of vinyl chloride and cis-dichloroethene as well as each step in the complete reductive dechlorination of tetracloroethene to ethene can occur in the Area 6 aquifer. Enrichment cultures capable of the oxidative degradation of cis-1,2-dichloroethene (cis-DCE) and vinyl chloride (VC) were obtained from groundwater across the aquifer demonstrating the possible importance of direct, non-cometabolic oxidation of cis-DCE and VC in natural attenuation. Culture-independent analyses based upon recovery of 16S rRNA gene sequences revealed the presence of anaerobic organisms distributed primarily between two major bacterial divisions: the delta subdivision of the Proteobacteria and low-G + C gram positive. Recovery of sequences affiliated with phylogenetic groups containing known anaerobic-halorespiring organisms such as Desulfitobacterium, Dehalobacter, and certain groups of iron reducers provided qualitative support for a role of reductive dechlorination processes in the aquifer. This molecular data is suggestive of a functional linkage between the microbiology of the site and the apparent natural attenuation process. The presence and distribution of microorganisms were found to be consistent with a microbially driven attenuation of chlorinated ethenes within the aquifer and in accord with a conceptual model of aquifer geochemistry which suggest that both reductive and oxidative mechanisms are involved in heterogeneous, spatially distributed processes across the aquifer. 相似文献

Coastal fisheries are a critical component of Pacific island food systems; they power village economies and provide nutritious aquatic foods. Many coastal women and men actively fishing in this region rely on multi-species fisheries, which given their extraordinary diversity are notoriously difficult to both characterize, and to manage. Understanding patterns of fishing, diversity of target species and drivers of these patterns can help define requirements for sustainable management and enhanced livelihoods. Here we use a 12-month data set of 8535 fishing trips undertaken by fishers across Malaita province, Solomon Islands, to create fisheries signatures for 13 communities based on the combination of two metrics; catch per unit effort (CPUE) and catch trophic levels. These signatures are in turn used as a framework for guiding suitable management recommendations in the context of community-based resource management. While a key proximate driver of these patterns was fishing gear (e.g. angling, nets or spearguns), market surveys and qualitative environmental information suggest that community fishing characteristics are coupled to local environmental features more than the market value of specific species they target. Changes in climate are occurring around the world and the effects on ecosystems will vary, depending on the extent and nature of these changes. In northern Europe, experts predict that annual rainfall will increase significantly, along with dramatic storm events and flooding in the next 50-100 years. Scotland is a stronghold of the endangered freshwater pearl mussel, Margaritifera margaritifera (L.), and a number of populations may be threatened. For example, large floods have been shown to adversely affect mussels, and although these stochastic events were historically rare, they may now be occurring more often as a result of climate change. Populations may also be affected by a number of other factors, including predicted changes in temperature, sea level, habitat availability, host fish stocks and human activity. In this paper, we explain how climate change may impact M. margaritifera and discuss the general implications for the conservation management of this species. 相似文献

There may be important biological sources of polycyclic aromatic hydrocarbons (PAHs) to the global environment, particularly of naphthalene, phenanthrene, and perylene, that originate in the tropics. We (i) studied the distribution of PAHs among different compartments of a typical Cerrado to locate their sources and (ii) quantified the PAH storage of this ecosystem. The sum of 20 PAH (sigma20PAHs) concentrations ranged from 25 to 666 microg kg(-1) in plant tissue, 7.4 to 32 microg kg(-1) in litterfall, 206 to 287 microg kg(-1) in organic soil, and 10 to 79 microg kg(-1) in mineral soil. Among the living biomass compartments, the bark had the highest mean PAH concentrations and coarse roots the lowest, indicating that PAHs in the plants originated mainly from aboveground sources. Naphthalene and phenanthrene were the most abundant individual PAHs, together contributing 33 to 96% to the sigma20PAHs concentrations. The total storage of the X20PAHs in Cerrado was 7.5 mg m(-2) to a 0.15-m soil depth and 49 mg m(-2) to a 2-m soil depth. If extrapolated to the entire Brazilian Cerrado region, roughly estimated storages of naphthalene and phenanthrene correspond to 7300 and 400 yr of the published annual emissions in the United Kingdom, respectively. The storage of benzo[a]pyrene, a typical marker for fossil fuel combustion, in the Cerrado only corresponds to 0.19 yr of UK emissions. These results indicate that the Brazilian savanna comprises a huge reservoir of naphthalene and phenanthrene originating most likely from the aboveground parts of the vegetation or associated organisms.
