The larvae and pupae of the ladybird Thalassa saginata develop inside colonies of the dolichoderine ant Dolichoderus bidens. This association is the first specific and obligatory relationship recorded between ants and ladybirds. The ants provide shelter and protection to the larvae but the diet of the latter remains unclear. The integration of T. saginata larvae into the ant colonies is achieved by mimicking the cuticular patterns of the ants brood. Human noroviruses are a major cause for gastroenteritis outbreaks. Filter-feeding bivalve molluscs, which accumulate noroviruses in their digestive tissues, are a typical vector for human infection. RT-qPCR, the established method for human norovirus detection in food, does not allow discrimination between infectious and non-infectious viruses and can overestimate potentially infectious viral loads. To develop a more accurate method of infectious norovirus load estimation, we combined intercalating agent propidium monoazide (PMAxx™)-pre-treatment with RT-qPCR assay using in vitro-cultivable murine norovirus. Three primer sets targeting different genome regions and diverse amplicon sizes were used to compare one-step amplification of a short genome fragment to three two-step long-range RT-qPCRs (7 kbp, 3.6 kbp and 2.3 kbp amplicons). Following initial assays performed on untreated infectious, heat-, or ultraviolet-inactivated murine noroviruses in PBS suspension, PMAxx™ RT-qPCRs were implemented to detect murine noroviruses subsequent to their extraction from mussel digestive tissues; virus extraction via anionic polymer-coated magnetic beads was compared with the proteinase K-dependent ISO norm. The long-range RT-qPCR process detecting fragments of more than 2.3 kbp allowed accurate estimation of the infectivity of UV-damaged murine noroviruses. While proteinase K extraction limited later estimation of PMAxx™ pre-treatment effects and was found to be unsuited to the assay, magnetic bead-captured murine noroviruses retained their infectivity. Genome copies of heat-inactivated murine noroviruses differed by 2.3 log10 between RT-qPCR and PMAxx™-RT-qPCR analysis in bivalve molluscs, the PMAxx™ pre-treatment allowing a closer approximation of infectious titres. The combination of bead-based virus extraction and PMAxx™ RT-qPCR thus provides a more accurate model for the estimation of noroviral bivalve mollusc contamination than the conjunction of proteinase K extraction and RT-qPCR and has the potential (once validated utilising infectious human norovirus) to provide an added measure of security to food safety authorities in the hazard assessment of potential bivalve mollusc contamination.
In many insect species, sperm-depleted males (SDMs, i.e. males that have exhausted their sperm after a given number of matings) remain sexually active and continue to mate females. Here, we investigated the behavioural modifications that occur in both sexes of the parasitoid Asobara tabida Nees (Hymenoptera: Braconidae), after matings by fertile males and sperm-depleted males. We show that (i) virgin females, mated females and females mated to a SDM exhibited different behaviours and that (ii) males responded differently to females depending on whether the females had previously mated with an SDM or not. Our findings demonstrate that SDM influenced the reproductive behaviour of both males and females, especially with regard to male responsiveness and female attractiveness. These findings are discussed in the context of adaptive behaviour and fitness maximization in both males and females. 相似文献
Environmental vulnerability analysis has been sparsely used in environmental performance evaluation (EPE) of technological innovations. The present paper proposes a methodological approach to carry out vulnerability analysis of watersheds and to integrate this analysis into methods of environmental performance evaluation of agro-industrial innovations. This approach is applied to the Ambitec-Life Cycle method, described in Part 1 (this issue) of this study. The case study of green coconut substrate compared to ripe coconut substrate, also described in Part 1 (this issue), is now presented considering the vulnerability analysis of the watersheds where the life cycle stages of these products occur. The integration of vulnerability analysis in Ambitec-Life Cycle contributes to a better understanding of the environmental aspects of agro-industrial technological innovations with potential to cause significant impacts in watersheds where these innovations are implemented. 相似文献
Much of our understanding of physiology and metabolism is derived from investigating mouse mutants and transgenic mice, and
open-access platforms for standardized mouse phenotyping such as the German Mouse Clinic (GMC) are currently viewed as one
powerful tool for identifying novel gene-function relationships. Phenotyping or phenotypic screening involves the comparison of wild-type control mice with their mutant or transgenic littermates. In our study, we explored
the extent to which standardized phenotyping will succeed in detecting biologically relevant phenotypic differences in mice
generated and provided by different collaborators. We analyzed quantitative metabolic data (body mass, energy intake, and
energy metabolized) collected at the GMC under the current workflow, and used them for statistical power considerations. Our
results demonstrate that there is substantial variability in these parameters among lines of wild-type C57BL/6 (B6) mice from
different sources. Given this variable background noise in mice that serve as controls, subtle phenotypes in mutant or transgenic
littermates may be overlooked. Furthermore, a phenotype observed in one cohort of a mutant line may not be reproducible (to
the same extent) in mice coming from a different environment or supplier. In the light of these constraints, we encourage
researchers to incorporate information on intrastrain variability into future study planning, or to perform advanced hierarchical
analyses. Both will ultimately improve the detectability of novel phenotypes by phenotypic screening.

This paper is part of a two-year study to investigate the feasibility of initiating a Clean Development Mechanism (CDM) project
in an indigenous community of Eastern Panamá, Ipetí-Emberá. We use participatory mapping and matrices as well as household
surveys to develop a land-use/land-cover baseline scenario and examine the role of local participation in assessing land-use
change. In Ipetí, land-use change has not occurred in a linear way over the last decades, and our data unveils socio-economic
factors as potential key drivers of change. The concordance that we observed between geographic information and individual
and collective perceptions of land-use change substantiates the possibility of using local knowledge in the establishment
of baseline data for CDM projects. Our calculations suggest that the total carbon (C) stocks in the Tierra Colectiva (TC) of Ipetí-Emberá in 2004 represents a 47% reduction from the estimated C stock at the onset of settlement in the early
1970’s. Results from the participatory assessments predict that, in 2024 and in absence of a CDM project, the C stocks will
decline from 301,859 t C in 2004 to 155,730 t C, which constitutes a reduction of 52%. The scenario with CDM estimates C stocks
of 305,853 t C for 2024, a value slightly superior to the 2004 value. In the TC there is ground to believe that cattle ranching
is likely to become an ever more important activity as the population is young and growing and cannot easily move elsewhere.
Forests tend to be cleared for cultivation while pastures are established on short fallows. Our baseline scenario underlines
the potential for a CDM project to make a significant difference in the future C stocks of this landscape. 相似文献
This paper investigates threats to farm management in the northern and central region of Côte d’Ivoire, with a particular focus on climate-related threats. To this end, farmers’ perception and adaptation strategies for climate change have been analyzed. The data were collected from 205 respondents by means of the Focus Groups method, and they were evaluated using a framework analysis. The main reported threats related to the implementation of farming activities are the high cost of inputs and the lack of technical support, which are followed by diseases, insects, and climate variations (scarcity of rains, strong winds, and high temperature). We find that most farmers have a strong perception of changes in climatic conditions. Their perceived impacts on the local environment through evidences like the disappearance of certain farming practices, occurrence of new insects, and the disruption of key time reference periods. Farmers mainly attempt to adapt by adjusting their agricultural calendar, adopting new short-season varieties, and using mixed cropping. We find that the most influential factors for farmers’ adaptation behavior is lack of contact with extension services and the scarcity of rainfall. Our suggestions for future agricultural policies for better adaptation to climate change are to take into account farmers’ perception, to provide suitable climate forecast, and to improve local technical support. 相似文献
A considerable number of bird species carry feathers to their nests. Feathers’ presence in the nests has traditionally been explained by their insulating properties. Recently, however, it has been suggested that feathers carried to the nests by females of the spotted starling (Sturnus unicolor L.) could have an ornamental function based on their ultraviolet (300–400 nm) and human-visible longer wavelength (400–700 nm) coloration. In our population, 95.7% of feathers found inside next-boxes occupied by nesting starlings were rock dove fly feathers. Of these feathers, 82.7% were naturally positioned with their reverse side oriented toward the entrance hole and 42.4% of all found feathers were situated within the nest-cup. Here we experimentally assess the signaling function of ultraviolet coloration of feathers in nests of spotless starlings by providing nests with a number of pigeon flight feathers that were respectively treated on their obverse, reverse, both, or neither side with a UV blocker. Starlings placed 42.5% of the experimental feathers in the nest-cup irrespective of the UV block treatment. Orientation of feathers toward the entrance hole was not related with their ultraviolet radiation. However, feathers placed within the nest-cup were more likely found with their reverse side oriented toward the entrance hole confirming our correlative findings.
