Transport experiments with colloids and radionuclides in a shear zone were conducted during the Colloid and Radionuclide Retardation experiment (CRR) at Nagra's Grimsel Test Site. Breakthrough curves of bentonite colloids and uranine, a non-sorbing solute, were measured in an asymmetric dipole flow field. The colloid breakthrough is earlier than that of uranine. Both breakthrough curves show anomalously long late time tails and the slope of the late time tails for the colloids is slightly higher. Anomalous late time tails are commonly associated with matrix diffusion processes; the diffusive interaction of solutes transported in open channels with the adjacent porous rock matrix or zones of stagnant water. The breakthrough curves for different colloid size classes are very similar and show no signs of fractionation due to their (size-dependent) diffusivity. It is proposed that tailing of the colloids is mainly caused by the structure of the flow field and that for the colloid transport, matrix diffusion is of minor importance. This has consequences for the interpretation of the uranine breakthrough. Comparisons of experimental results with numerical studies and with the evaluation of the colloid breakthrough with continuous time random theory imply that the tailing in the conservative solute breakthrough in this shear zone is not only caused by matrix diffusion. Part of the tailing can be attributed to advective transport in fracture networks and advection in low velocity regions. Models based on the advection-dispersion equation and matrix diffusion do not properly describe the temporal and spatial evolution of colloid and solute transport in such systems with a consistent set of parameters.  相似文献

Objective and Background

PCB (polychlorinated biphenyle) are typical man-made environmental pollutants that cause a broad spectrum of effects in vertebrates. Although a lot has been studied about the toxic effects of xenobiotic substances on aquatic organisms, their subacute effects are not yet well known. Fish occupies a central position in freshwater food webs and therefore, carp was chosen to be our test organism. The aim of our study was the investigation of subacute effects of Aroclor 1254 (22μg l?1) on respiration, swimming activity, and biotransformation, allowing discussion of changed interaction between the available energy resources.

Methods

The respiration experiments under controlled laboratory conditions were run for 29 days (5 d before, 16 d with and 8 d after chemical exposure) with exposed carp compared to the non-exposed fish. Furthermore, the BehavioQuant system quantitatively monitors the positions of each individual fish before (8 d) and during (21 d) PCB exposure and swimming activity (number of horizontal turnings s?1) of animals was calculated. Thereafter, liver samples were taken from animals for analysis of the phase 2 enzyme activity (glutathione-S-transferase). In the respiration experiments were 6 exposure, 6 controll, and 3 solubilizer controll groups (12 animals each group); in the behavioral experiments were 9 exposure, 6 controll- und 3 solubilizer controll groups (6 animals each group), and in the enzyme activity measurements were 9 exposure, 6 controll and 3 solubilizer controll groups (6 animals each group) examined.

Results and Discussion

A clearly increased (33%) oxygen consumption due to the PCB-exposure is found to be reversible; after the exposure period the respiration of carp shows a level comparable to the oxygen consumption during non-exposure time. By evaluating quantitative behavioral parameters of animals, it becomes evident that the exposure to PCB also causes a reversible change in their swimming behavior. Chemical stress leads to a decrease of mean daily swimming activity combined with an increase of the mean number of turnings during the night. Thereafter, the swimming activity shifts to the circadian swimming behavior under control conditions. Furthermore, our measurement of soluble and microsomal glutathione-S-transferase activity of fish liver shows a significant elevation after exposure period.

Conclusion

Our results prove that it was feasible to detect sublethal effects of PCB-mixture on all parameters under these conditions successfully. Our findings imply that fish are able to cope with the chemicals and we purse the hypothesis that the higher physiological energy demand caused by increased activity of enzymes, which are involved in biotransformation of foregin substances, may be related to a higher respiration of fish stressed by xenobiotics.

Recommendation and Outlook

Further experiments should investigate the effects of PCB on ecophysiological parameters of carp fed with exposed natural food allowing discussion of ecological consequences.  相似文献

The study of the fetal platelet count and size can, according to the literature, be used for the prenatal diagnosis of the Wiskott-Aldrich syndrome (WAS). So far, no affected fetuses have been identified by this method. All pregnancies in which this method had been applied to resulted, as correctly predicted, in the birth of normal children. Here we report on a familial case of WAS where the haematological parameters failed to reveal the affected second child. Hence we assume that the platelet count and size of platelets remain normal in fetuses with WAS to the gestational age of 22 weeks and cannot be used for prenatal diagnosis.  相似文献

Protecting our water resources in terms of quality and quantity is considered one of the big challenges of the twenty-first century, which requires global and multidisciplinary solutions. A specific threat to water resources, in particular, is the increased occurrence and frequency of flood events due to climate change which has significant environmental and socioeconomic impacts. In addition to climate change, flooding (or subsequent erosion and run-off) may be exacerbated by, or result from, land use activities, obstruction of waterways, or urbanization of floodplains, as well as mining and other anthropogenic activities that alter natural flow regimes. Climate change and other anthropogenic induced flood events threaten the quantity of water as well as the quality of ecosystems and associated aquatic life. The quality of water can be significantly reduced through the unintentional distribution of pollutants, damage of infrastructure, and distribution of sediments and suspended materials during flood events. To understand and predict how flood events and associated distribution of pollutants may impact ecosystem and human health, as well as infrastructure, large-scale interdisciplinary collaborative efforts are required, which involve ecotoxicologists, hydrologists, chemists, geoscientists, water engineers, and socioeconomists. The research network “project house water” consists of a number of experts from a wide range of disciplines and was established to improve our current understanding of flood events and associated societal and environmental impacts. The concept of project house and similar seed fund and boost fund projects was established by the RWTH Aachen University within the framework of the German excellence initiative with support of the German research foundation (DFG) to promote and fund interdisciplinary research projects and provide a platform for scientists to collaborate on innovative, challenging research. Project house water consists of six proof-of-concept studies in very diverse and interdisciplinary areas of research (ecotoxicology, water, and chemical process engineering, geography, sociology, economy). The goal is to promote and foster high-quality research in the areas of water research and flood-risk assessments that combine and build off-laboratory experiments with modeling, monitoring, and surveys, as well as the use of applied methods and techniques across a variety of disciplines.  相似文献

A multi-component screening analysis method for polar to medium-polar water pollutants was developed. Sample clean-up and group separation are performed by sequential solid-phase extraction (SSPE) using automated SPE with C18 and polymeric sorbent materials. Analyses are performed by liquid chromatography electrospray ionization mass spectrometry (LC-ESI-MS) using a single-quadrupole instrument. More than 90 priority compounds of environmental interest--comprising the most important chemical and substance classes: phenols, carboxylic acids, aromatic sulfonates, aromatic amines, pharmaceuticals, surfactants, dyes, and pesticides--have been chosen for the experiments. The compounds are divided by the SSPE procedure into 3 different polarity classes. The extraction recoveries were determined in the 3 fractions for every single substance, and were for most of the analytes in the range of 50-100%. A mixture of hexane-dichloromethane was used for the elution of nonpolar compounds like alkylphenols from C18. Methanol and acetone are well suited for the elution of more polar substances. The limits of detection (LODs) were determined for all compounds. Effluents from municipal and industrial wastewater treatment plants (WWTPs) treating waste water from textile industries; and the corresponding receiving waters (rivers and lakes) have been analysed with the developed method. Urban and industrial pollution was observed in rivers and streams in the area north of Milan, Italy. In the water samples different phenols (nitrophenols, bisphenol A, nonylphenol), alkylphenol ethoxylate surfactants, their metabolites with endocrine disrupting potential, aromatic sulfonates, linear alkylbenzenesulfonate surfactants, dyes, pesticides, pharmaceuticals, and a dichlorobenzidine compound were identified.  相似文献

It has been generally accepted, during the last few decades, that the dissolved natural organic matter in water [DNOM] appearing as yellow brownish color, has become more and more "polluted" by inorganic and organic micro-pollutants. Due to the complexing properties of NOM, lipophilic organic micro-pollutants, such as, PCBs will be mobilized into water together with the DNOM. A mixture of eight PCBs, with Cl-content from tri-Cl to hepta-Cl, was added to solutions of ten different DNOMs. The DNOMs were aqua's solutions of RO-(reverse osmosis)-isolated material, having approximately the same concentration of carbon. After a contact time of three days, standard analytical PCB-method was used to determine the recovery of the added PCBs. The results show that the analytical availability of the added PCB was significantly reduced in the presence DNOM, compared to distilled water. The percentage loss in recovery of PCB increased with the content of Cl, in mean, from 3%/mg C for tri-Cl to 9%/mg C for hepta-Cl. The results also suggest that the analytical recovery of PCB was affected by the quality and the nature of the organic matter. For example the longer the DNOMs had been in the aquatic phase, the less efficient they are attached to the PCBs.  相似文献
