Ammonia oxidation, the first and rate-limiting step of nitrification, is mainly performed by ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB). However, the activities of AOA and AOB in soil and their relative contribution to ammonia oxidation are unclear, and whether there is a significant correlation between the quantity of AOA and AOB and the ammonia oxidation rate is also controversial. In this study, quantitative PCR combined with acetylene (C2H2) and 1-octyne inhibition methods were used to determine the quantity and activity of AOA and AOB in wheat, highland barley, and oilseed rape soils in Nyingchi, Lhatse, Sangzhuzi, and Sangri counties on the Qinghai-Tibet Plateau. The results showed that the quantity of AOB ((2.34 ± 0.84) ×105 - (2.65 ± 1.07) ×106 copies g-1 dry soil) was significantly higher than that of AOA ((0.20 ± 0.10) ×104 - (4.02 ± 0.39) ×104 copies g-1 dry soil) in all the soil samples. Soil pH was the key factor affecting the quantity of AOB, and the total phosphorus and ammonium nitrogen in soil were the key factors affecting the quantity of AOA. The rates of ammonia oxidation in the farmland soils of Lhatse (2.42 ± 0.73 mg kg-1 d-1) and Sangzhuzi (3.24 ± 1.15 mg kg-1 d-1) were significantly higher than those in the soils of Nyingchi (1.17 ± 0.43 mg kg-1 d-1) and Sangri counties (0.88 ± 0.57 mg kg-1 d-1). The rates of ammonia oxidation in the farmland soils of Lhatse and Sangzhuzi were dominated by AOB, while those in the farmland soils of Nyingchi and Sangri counties were dominated by AOA. For crops, the ammonia oxidation rates of wheat and oilseed rape soils in all four regions were significantly higher than those of highland barley soil, whereas the activity of AOA and AOB was not influenced by crops. The ratio of nitrogen to phosphorus was the key factor influencing AOA activity, whereas soil pH and total carbon were the main factors influencing AOB activity. Additionally, the quantities of AOA and AOB were not significantly correlated with the total ammonia oxidation rates and AOA and AOB activity. Overall, our study suggests that both AOA and AOB play important roles in ammonia oxidation in farmland soils of the Qinghai-Tibet Plateau. Moreover, it is unreliable to predict the activity of AOA and AOB and their relative contribution to ammonia oxidation directly based on their number of amoA genes, and the activity of AOA and AOB should be directly and accurately measured. These results are important for understanding ammonia nitrogen removal processes, slowing nitrate loss, and reducing the emission of the greenhouse gas nitrous oxide in the farmland ecosystem of the Qinghai-Tibet Plateau. © 2022 Science Press. ● Converting xylose to caproate under a low temperature of 20 °C by MCF was verified. ● Final concentration of caproate from xylose in a batch reactor reached 1.6 g/L. ● Changing the substrate to ethanol did not notably increase the caproate production. ● Four genera, including Bifidobacterium , were revealed as caproate producers. ● The FAB pathway and incomplete RBO pathway were revealed via metagenomic analysis. Mixed culture fermentation (MCF) is challenged by the unqualified activity of enriched bacteria and unwanted methane dissolution under low temperatures. In this work, caproate production from xylose was investigated by MCF at a low temperature (20 °C). The results showed that a 9 d long hydraulic retention time (HRT) in a continuously stirred tank reactor was necessary for caproate production (~0.3 g/L, equal to 0.6 g COD/L) from xylose (10 g/L). The caproate concentration in the batch mode was further increased to 1.6 g/L. However, changing the substrate to ethanol did not promote caproate production, resulting in ~1.0 g/L after 45 d of operation. Four genera, Bifidobacterium, Caproiciproducens, Actinomyces, and Clostridium_sensu_stricto_12, were identified as the enriched caproate-producing bacteria. The enzymes in the fatty acid biosynthesis (FAB) pathway for caproate production were identified via metagenomic analysis. The enzymes for the conversion of (Cn+2)-2,3-Dehydroxyacyl-CoA to (Cn+2)-Acyl-CoA (i.e., EC 1.3.1.8 and EC 1.3.1.38) in the reverse β-oxidation (RBO) pathway were not identified. These results could extend the understanding of low-temperature caproate production.  相似文献

广东鼎湖山自然保护区的土壤主要为山地黄壤和赤红壤。本文用差热-热重-微商热重(DTA-TG-DTG)联合热分析法,对两种土壤的粘粒矿物进行了初步研究,确认两种土壤的粘粒矿物组成主要为伊利石、高岭石、三水铝石、蛭石、针铁矿等。伊利石、三水铝石、蛭石的含量山地黄壤多于赤红壤,而高岭石则相反。两种土壤的风化程度不高,土壤矿物风化主要处于幼年阶段,山地黄壤尤其明显。另外,对不同林型下的赤红壤也进行了对比研究,其粘粒矿物组成和数量差异不大。  相似文献

Providing an accurate estimate of the dry component of N deposition to low N background, semi-natural habitats, such as bogs and upland moors dominated by Calluna vulgaris is difficult, but essential to relate nitrogen deposition to effects in these communities. To quantify the effects of NH3 inputs to moorland vegetation growing on a bog at a field scale, a field release NH3 fumigation system was established at Whim Moss (Scottish Borders) in 2002. Gaseous NH3 from a line source was released along of a 60 m transect, when meteorological conditions (wind speed >2.5 m s?1 and wind direction in the sector 180–215°) were met, thereby providing a profile of decreasing NH3 concentration with distance from the source. In a complementary study, using a NH3 flux chamber system, the relationships between NH3 concentrations and cuticular resistances were quantified for a range of NH3 concentrations and micrometeorological conditions for moorland vegetation. Cuticular resistances increased with NH3 concentration from 11 s m?1 at 3.0 μg m?3 to 30 s m?1 at 30 μg m?3. The NH3 concentration data and the concentration-dependent canopy resistance are used to calculate NH3 deposition taking into account leaf surface wetness. The implications of using an NH3 concentration-dependent cuticular resistance and the importance for refining critical loads are discussed.  相似文献

采用AM-1吸附树脂和ND-900络合树脂对硝基苯乙酮生产废水进行吸附处理,试验结果表明,使用2种树脂串联的处理效果优于单独使用某一种树脂,而且ND-900络合树脂在前,AM-1吸附树脂在后串联处理的效果最好,废水体积为30BV时,COD去除率为92.8%;脱色率为100%,出水由原废水的橙红色变为无色,采用质量分数为8%的NaOH溶液作为脱附剂,脱附效果良好。  相似文献
