Air sampling for (1 → 3)-β-D-glucan may be a good method for assessing inhalation exposure to airborne fungi. Pre-sampling contamination of filter media used for sampling (1 → 3)-β-D-glucan may lead to substantial exposure measurement errors. Using the Limulus amebocyte lysate assay, we tested for pre-sampling levels of (1 → 3)-β-D-glucan on three types of filters-mixed cellulose ester (MCE)[1 brand], glass fiber (GF)[1 brand], and polycarbonate (PC)[5 brands]. Levels of (1 → 3)-β-D-glucan on MCE filters exceeded 4586.1 pg per filter. Levels on GF filters averaged 135.3 (± 28.9) pg per filter (range = 94.8-160.4 pg per filter) and levels on PC filters averaged 152.4 (± 236.1) pg per filter (range = non-detectable-1760.7 pg per filter). Efforts to clean MCE and GF filters were unfeasible or unsuccessful. Sonicating PC filters for two hours in ethanol, followed by a wash in pyrogen-free water, effectively eliminated measured levels of (1 → 3)-β-D-glucan on four brands of PC filters, as compared to untreated PC filters. This pretreatment process did not appear to physically damage the PC filters. Air sampling results highlighted the potentially problematic contamination of untreated PC filters. Ensuring that sampling media are free of (1 → 3)-β-D-glucan before sampling is crucial to accurately measure levels of (1 → 3)-β-D-glucan exposure, especially in environments where levels of (1 → 3)-β-D-glucan are low.  相似文献

Analytical methods for the speciation of mercury, based on microwave extraction and sonication extraction, have been tested to determine the inorganic mercury and methyl mercury contents in two standard soil reference materials: SRM 2710 Montana Soil and BCR 580 estuarine sediment. Prior to applying the speciation extraction methods, the mineral compositions were analyzed via XRD analysis, with SRM 2710 shown to be composed mostly of aluminum silicate minerals, while carbonate minerals were the major constituent in BCR 580. Two extraction methods, microwave and sonication, were tested for the analysis and recovery efficiency of total mercury. The accuracy and efficiency of each extraction method was also compared. In the analysis of total mercury, the microwave extraction method, with using methanol and HCl as extractants, was better for SRM2710, while the application of the sonication extraction method was more efficient for the calcite-based BCR 580. The results showed good separation and recovery efficiencies, with values reaching 100% of those estimated. The sonication method was selected for the speciation of mercury, especially in BCR 580. An extraction solution comprising of a 1:1 mixture of methanol and HCl was used for the sonication extraction of BCR 580, with the resulting extractants analyzed by IC-HG-ICP-MS for methyl mercury and inorganic mercury. As a simple, rapid, sensitive, and accurate method, sonication extraction was found to be satisfactory.  相似文献

We report one case of a familial complex chromosomal rearrangement (CCR) involving four different chromosomes 5, 10, 16 and 18. The CCR was detected prenatally at 20 weeks' gestation because of advanced maternal age and history of recurrent miscarriages. Cytogenetic analysis of cultured amniotic fluid cells with GTG banding showed a 46,XX,t(5;16;10;18)(q13;q22;q11.2;q21) karyotype. Parental cytogenetic study revealed that the mother has the same CCR. RBG banding, high-resolution banding and fluorescence in situ hybridization (FISH) were used to characterize further and confirm the conventional banding data. No physical abnormalities were shown in the targeted fetal ultrasonography examination. The parents decided to continue the pregnancy. The child is now 2 years old and has neither congenital anomalies nor evidence of delayed psychomotor development. The fetal targeted ultrasound and FISH analysis helped us reassure fetal status. This study evaluated the reduction e ect of non-point source pollution by applying best management practices (BMPs) to a 1.21 km2 small agricultural watershed using a SWAT (Soil andWater Assessment Tool) model. Two meter QuickBird land use data were prepared for the watershed. The SWAT was calibrated and validated using daily streamflow and monthly water quality (total phosphorus (TP), total nitrogen (TN), and suspended solids (SS)) records from 1999 to 2000 and from 2001 to 2002. The average Nash and Sutcli e model e ciency was 0.63 for the streamflow and the coe cients of determination were 0.88, 0.72, and 0.68 for SS, TN, and TP, respectively. Four BMP scenarios viz. the application of vegetation filter strip and riparian bu er system, the regulation of Universal Soil Loss Equation P factor, and the fertilizing control amount for crops were applied and analyzed.  相似文献

The formation of a dynamic membrane (DM) was investigated using polyethylene glycol (PEG) (molecular weight of 35000 g/mol, concentration of 1 g/L). Two natural organic matters (NOM), Dongbok Lake NOM (DLNOM) and Suwannee River NOM (SRNOM) were used in the ultrafiltration experiments along with PEG. To evaluate the effects of the DM with PEG on ultrafiltration, various transport experiments were conducted, and the analyses of the NOM in the membrane feed and permeate were performed using high performance size exclusion chromatography, and the effective pore size distribution (effective PSD) and effective molecular weight cut off (effective MWCO) were determined. The advantages of DM formed with PEG can be summarized as follows: (1) PEG interferes with NOM transmission through the ultrafiltration membrane pores by increasing the retention coefficient of NOM in UF membranes, and (2) low removal of NOM by the DM is affected by external factors, such as pressure increases during UF membrane filtration, which decreases the effective PSD and effective MWCO of UF membranes. However, a disadvantage of the DM with PEG was severe flux decline; thus, one must be mindful of both the positive and negative influences of the DM when optimizing the UF performance of the membrane.  相似文献

For the screening of the risk from environmental contamination, the cytotoxic/genotoxic effects of various model pollutants were determined using an in vivo system comprised of human HeLa cells; the ecotoxicity was also determined using the acute and genotoxicity tests on two aquatic sentinel species widely used in biomonitoring, namely, freshwater crustacean, Daphnia magna and larva of aquatic midge, Chironomus tentans. Nonylphenol (NP), bisphenol A (BPA), bis(2-ethylhexyl) phthalate (DEPH) and paraquat dichloride (PQ) were used as the model pollutants. The results showed that exposure of HeLa cells to NP, BPA and DEHP was sufficient for the expression of noticeable genotoxic and cytotoxic effects. Ecotoxicity results showed that, as expected, D. magna was more sensitive than C. tentans to chemical exposure. BPA may exert a genotoxic effect on D. magna and C. tentans, given that DNA strand breaks increased in both species exposed to this compound, whereas NP-induced DNA damage occurred only in C. tentans. In vivo genotoxic data obtained in aquatic sentinel species could provide valuable information for freshwater quality monitoring. From the results of the present study, the use of cytotoxic, genotoxic and ecotoxic tests using human cell system, as well as, biomonitoring species, seems to be relevant for preliminary evaluation of the human health and ecological effects of pollutants and thus, a promising screening tool for environmental monitoring and risk assessment.  相似文献

The transfer of 90Sr to rice plants following its acute ground deposition was examined experimentally in a greenhouse. Lysimeters were flooded after being filled with the soil monoliths from 12 paddy fields. A solution of 90Sr was applied to the standing water in the flooded lysimeters at the pre-transplanting stage or booting stage. Applied 90Sr was mixed with the topsoil only after the pre-transplanting application (PTA). The transfer was quantified with the areal transfer factor (TF(a), m2 kg(-1)-dry) defined as the ratio of the plant concentration to the initial ground deposition. In the PTA, the first-year TF(a) values in the 12 soils were in the range of 8.2 x 10(-3) -2.1 x 10(-2) and 1.7 x 10(-4) -3.6 x 10(-4) for the straws and hulled seeds, respectively. The TF(a) values from the booting-stage application (BSA) were higher than those from the PTA by a factor of up to four. The ratios of the seed TF(a) to the straw TF(a) were, on the whole, higher in the BSA. The 90Sr TF(a) in the PTA was negatively correlated with the soil pH and, to a lesser degree, the exchangeable Ca content. In the second year, the TF(a) in the PTA reduced to 53-90% of that in the first year. A more significant reduction, in general, occurred in a sandier soil. Based on the four consecutive years' transfer data, an overall half-time of the 90Sr TF(a) was estimated to be 2.2 years.  相似文献

The removal of As(III) and As(V) from aqueous solution was investigated using waste cast iron, which is a byproduct of the iron casting process in foundries. Two types of waste cast iron were used in the experiment: grind precipitate dust (GPD) and cast iron shot (CIS). The X-ray diffraction analysis indicated the presence of Fe0 on GPD and CIS. Batch experiments were performed under different concentrations of As(III) and As(V) and at various initial pH levels. Results showed that waste cast iron was effective in the removal of arsenic. The adsorption isotherm study indicated that the Langmuir isotherm was better than the Freundlich isotherm at describing the experimental result. In the adsorption of both As(III) and As(V), the adsorption capacity of GPD was greater than CIS, mainly due to the fact that GPD had higher surface area and weight percent of Fe than CIS. Results also indicated the removal of As(III) and As(V) by GPD and CIS was influenced by the initial solution pH, generally decreasing with increasing pH from 3.0 to 10.5. In addition, both GPD and CIS were more effective at the removal of As(III) than As(V) under given experimental conditions. This study demonstrates that waste cast iron has potential as a reactive material to treat wastewater and groundwater containing arsenic.  相似文献
