Seaweeds have been used as a source of traditional medicine worldwide for the treatment of various ailments, mainly due to their ability to quench the free radicals. The present study aims at evaluating the protective effect of methanolic extract of Gelidiella acerosa, an edible red seaweed against 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)-induced toxicity in peripheral blood mononuclear cells (PBMC). For evaluating the protective effect of G. acerosa, PBMC were divided into four groups: vehicle control, TCDD (10 nM), TCDD?+?G. acerosa (300 μg/ml), and G. acerosa alone treated. Scavenging of intracellular reactive oxygen species (ROS) induced by TCDD was assessed by the dichloro-dihydro-fluorescein diacetate (DCFH-DA) method. Alterations at macromolecular level were quantified through lipid peroxidation (LPO) level, protein carbonyl content (PCC) level, and comet assay. The cellular morphology upon TCDD toxicity and G. acerosa treatment was obtained by light microscopy and histopathological studies. The chemical composition present in the methanolic extract of G. acerosa was determined by gas chromatography-mass spectrometry (GC-MS) analysis. The results reveal that 10 nM TCDD caused significant (P?<?0.05) reduction in cell viability (94.10?±?0.99), and treatment with 300 μg/ml extract increased the cell viability (99.24?±?0.69). TCDD treatment resulted in a significant increase in the production of ROS, LPO (114?±?0.09), and PCC (15.13?±?1.53) compared to the control, whereas co-treatment with G. acerosa significantly (P?<?0.05) mitigated the effects. Further, G. acerosa significantly (P?<?0.05) prevented TCDD-induced genotoxicity and cell damage. GC-MS analysis showed the presence of n-hexadecanoic acid (retention time (RT) 13.15), cholesterol (RT 28.80), α-d-glucopyranose, 4-O-α-d-galactopyranosyl (RT 20.01), and azulene (RT 4.20). The findings suggest that G. acerosa has a strong protective ability against TCDD-induced cytotoxicity, oxidative stress, and DNA damage. 相似文献

The present study was carried out to determine the mangrove area water quality, mangrove diversity and density at Minnie Bay, Lalaji Bay and Aerial Bay (Andaman Islands, India). The physicochemical parameters such as salinity, nitrate, inorganic phosphate and total phosphorus varied significantly. Mangrove diversity was high at Minnie Bay while density at Aerial Bay. Rhizophora mucronata dominated at all three sites during the study period. Shannon-Wiener diversity index (H′?=?2.261) and Pielou’s evenness (J′?=?0.8348) were high at Minnie Bay. The higher species density cover (5.679 Nos./m2) was recorded at Aerial Bay. Bray-Curtis cluster analysis showed 63.35 % similarity and formed two clusters. Though uplift and subsidence of coastal land due to the Mw 9.3 Sumatra earthquake in 2004 has affected the mangrove vegetation, its rejuvenation was observed in the newly formed inter-tidal areas due to changes in coastal geomorphology. Eighty-two surface soil samples were collected from forest, grassland, tea estate, wildlife sanctuary, wetland, and roadside areas from the northeastern states of India, viz., Tripura, Manipur, and Assam. Thirteen different organochlorine pesticides (OCPs) were detected from background soils using gas chromatography electron capture detector. Manipur soils were found to be with higher concentration of dichlorodiphenyltrichloroethanes (DDTs), hexachlorocyclohexanes (HCHs), and endosulfan followed by Tripura and Assam. The spearman correlation coefficient shows significant correlation between HCHs, DDTs, and endosulfan isomers (r 2?>?0.5 and p?<?0.05). Additionally, α-HCH, δ-HCH, o,p′-DDE, and endosulfan-sulfate shows good correlation with total organic carbon in soil (r 2?=?0.5, p?=?0.05), indicating that the soil organic matter could enhance adsorption of these compounds, also demonstrating that the present OCPs in the background soil were from similar source. Further principal component analysis evaluates that most of the higher volatile compounds where clustered together in soil. However, after comparing with different states of Indian soil samples, the concentrations of OCPs in the present study areas are much lower and comparable with background soil across the globe. 相似文献

The study was carried out to understand the variability in phytoplankton production (Chlorophyll a) and mesozooplankton diversity from two different shallow coastal regions of south Andaman viz. Port Blair Bay (PBB), the only real urban area among the islands and Mahatma Gandhi Marine National Park, a Marine Protected Area (MPA) at Wandoor. Seasonal sampling was carried out during the Northeast monsoon (NEM—November 2005), Intermonsoon (IM—April 2006), and Southwest monsoon (SWM—August 2006). Significant (P?<?0.05) seasonal variation was observed in the environmental variables at both the regions. Higher average chlorophyll a (Chl. a) and mesozooplankton standing stock were observed at PBB compared to MPA, but the seasonal variation observed was marginal at both the study areas. Chl. a showed a steep increasing gradient from outer to the inner regions of the PBB. The number of zooplankton taxa recorded at both areas was quite similar, but marked differences were noticed in their relative contribution to the total abundance. Eventhough the Copepoda dominated at both the areas, the non-copepod taxa differed significantly between the regions. Dominance of carnivores such as siphonophores and chaetognaths were noticed at PBB, while filter feeders such as appendicularians and decapod larvae were more abundant at MPA. A total of 20 and 21 copepod families was recorded from PBB and MPA, respectively. Eleven species of chaetognaths were observed as common at both areas. Larval decapods were found to be predominant at MPA with 20 families; whereas, at PBB, only 12 families were recorded. In the light of the recent reports on various changes occurring in the coastal waters of the Andaman Islands, it is suspected that the difference in Chl. a as well as the mesozooplankton standing stock and community structure observed between the two study areas may be related to the various anthropogenic events influencing the coastal waters. 相似文献

The protective effect of hydroxytyrosol (HT), a strong antioxidant compound from extra virgin olive oil, against TCDD induced toxicity was investigated in human peripheral blood mononuclear cells (PBMC). PBMC (1 × 106 cells mL−1) were divided into four groups and were incubated in a CO2 incubator (5% CO2) for 12 h with vehicle, TCDD (10 nM), TCDD + HT (10 nM + 100 μM) and HT alone (100 μM) respectively. To clarify the role of HT against TCDD induced cytotoxicity, oxidative stress and the levels of antioxidant enzymes were assessed. Incubation of PBMC with TCDD significantly decreased cell viability, catalase (CAT) and glutathione peroxidase (GPx) and increased the levels of superoxide dismutase (SOD), glutathione reductase (GR) and oxidative stress markers such as lipid peroxidation products (LPO), protein carbonyl content (PCC) and reactive oxygen species (ROS). Whereas, HT had an effective antioxidant property as observed by the increased cell viability, normalization of antioxidant enzymes and decreased levels of LPO, PCC and ROS in PBMC co-treated with HT and TCDD. Apoptosis detection and comet assay results shows that HT, by acting as an antioxidant, prevents the damage to DNA induced by TCDD. In addition light microscopic and histopathological observations revealed that the cells are apoptotic and degenerated during TCDD treatment, whereas cells showed intact morphology during co-treatment with HT. On the whole, the results reveal that HT exerts a promising antioxidant potential in protecting the PBMC against TCDD induced oxidative stress, which might be due to the presence of catechol moiety in its structure. 相似文献

Purpose
??-Hexachlorocyclohexane (HCH), ??-HCH, and lindane (??-HCH) were listed as persistent organic pollutants by the Stockholm Convention in 2009 and hence must be phased out and their wastes/stockpiles eliminated. At the last operating lindane manufacturing unit, we conducted a preliminary evaluation of HCH contamination levels in soil and water samples collected around the production area and the vicinity of a major dumpsite to inform the design of processes for an appropriate implementation of the Convention.Methods
Soil and water samples on and around the production site and a major waste dumpsite were measured for HCH levels.Results
All soil samples taken at the lindane production facility and dumpsite and in their vicinity were contaminated with an isomer pattern characteristic of HCH production waste. At the dumpsite surface samples contained up to 450?g?kg?1 ?? HCH suggesting that the waste HCH isomers were simply dumped at this location. Ground water in the vicinity and river water was found to be contaminated with 0.2 to 0.4?mg?l?1 of HCH waste isomers. The total quantity of deposited HCH wastes from the lindane production unit was estimated at between 36,000 and 54,000?t.Conclusions
The contamination levels in ground and river water suggest significant run-off from the dumped HCH wastes and contamination of drinking water resources. The extent of dumping urgently needs to be assessed regarding the risks to human and ecosystem health. A plan for securing the waste isomers needs to be developed and implemented together with a plan for their final elimination. As part of the assessment, any polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/PCDF) generated during HCH recycling operations need to be monitored. 相似文献9.

The provision of sheltered housing for sale in Scotland has experienced rapid growth since the opening of the first scheme in 1982. Development has been geographically concentrated in a small number of inner city and rural (coastal) areas. The expectation of continued rapid expansion is unlikely if recent trends of slower growth continue. This paper uses evidence from interviews with developers and owners of sheltered and mainstream housing, to examine the possible future contribution of the private market.
