Inonotus hispidus is a kind of rare medicinal fungus, and its natural resources are very scarce. Currently, the artificial cultivation technology of I. hispidus is not completely developed, and this reflects on its extremely low biological conversion rate and long cultivation period. In order to improve the bioconversion rate and shorten the production cycle of I. hispidus, we first analyzed the mycelia culture conditions of the collected I. hispidus, and then we further explore the method of domesticated cultivation of its fruiting body in rice medium. During the process of mycelial culture, the suitable temperature, pH, carbon source, and nitrogen source for mycelial growth were selected using the mycelial growth rate as index. During the domesticated cultivation of the fruiting body, the suitable culture medium for its growth was selected using the bioconversion rate as index. Screening results of mycelial culture conditions showed that the optimal culture conditions for the growth of mycelium of the wild I. hispidus were: temperature of 25 °C, initial pH of 6.0, glucose as the carbon source, and yeast extract powder as the source of nitrogen. The results of the domesticated cultivation showed that the biotransformation rate of I. hispidus was higher when using rice as the main medium substrate. The optimal cultivation conditions were: a 0.2% yeast extract content in the nutrient solution, a 1:1.6 ratio of rice to nutrient solution, and a 4 mL inoculum of the liquid strain. Under these conditions, it took about 4 days for the mycelium to grow over the cultivation medium. The time required for the differentiation of the primordium to form fruit bodies was about 20 days, and the bioconversion rate reached 28.70% ± 5.05%. The results of this study indicate the feasibility of using rice as the main substrate for the cultivation of I. hispidus, and it also provide new insights for the finding of new cultivation substrates for other rare medicinal fungi. © 2018 Science Press. Based on field investigations in the Changjiang River mouth, rain sampling from the river's upper reaches to the mouth, historical data, and relevant literatures, the budget and major control factors of total phosphorus (TP) and dissolved inorganic phosphorus (DIP) in the river catchment were estimated and discussed from the catchment scale. It will supply a base for effective controlling P concentration in the Changjiang River and its mouth. Results show that the export fluxes of TP and DIP were mainly controlled by the river runoff. The TP and DIP in the Changjiang River came mainly from agricultural nonpoint source P losses from fertilizer and soil and point sources of industrial waste and residential sewage discharges. The export fluxes of TP and DIP in the river mouth were only 27.0% and 28.3% of the imports from the river catchment, respectively, suggesting that most of P were removed in transportation. A mass of freshwater marshes in the river catchment were the main areas to remove P. Significant increasing in utilization of fertilizer P and incessant increasing in water and soil loss areas were the primary reasons of agricultural nonpoint source P losses. How to decrease the agricultural nonpoint source P loss from the catchment scale is a key controlling P concentration in the Changjiang River and its mouth. The point source industrial waste P discharge has been primarily treated in the river catchment, but the residential sewage P discharge needs further control. 相似文献

Nitrogen removal from ammonium-containing wastewater was conducted using polylactic acid (PLA)/starch blends as carbon source and carrier for functional bacteria. The exclusive and interactive influences of operating parameters (i.e., temperature, pH, stirring rate, and PLA-to-starch ratio (PLA proportion)) on nitrification (Y1), denitrification (Y2), and COD release rates (Y3) were investigated through response surface methodology. Experimental results indicated that nitrogen removal could be successfully achieved in the PLA/starch blends through simultaneous nitrification and denitrification. The carbon release rate of the blends was controllable. The sensitivity of Y1, Y2, and Y3 to different operating parameters also differed. The sequence for each response was as follows: for Y1, pH>stirring rate>PLA proportion>temperature; for Y2, pH>PLA proportion>temperature>stirring rate; and for Y3, stirring rate>pH>PLA proportion>temperature. In this study, the following optimum conditions were observed: temperature, 32.0°C; pH 7.7; stirring rate, 200.0 r·min-1; and PLA proportion, 0.4. Under these conditions, Y1, Y2, and Y3 were 134.0 μg-N·g-blend-1·h-1, 160.9 μg-N·g-blend-1·h-1, and 7.6 × 103 μg-O·g-blend-1·h-1, respectively. Distribution behaviors of thifluzamide, fenoxanil, and tebuconazole applied to rice were investigated in South China. Analysis was by a modified QuEChERS method with gas chromatography (thifluzamide and fenoxanil) and liquid chromatography mass spectrometry (tebuconazole). Thifluzamide and tebuconazole partitioned mainly into the soil, with half-lives in the paddy soil of 12–14 and 5.3–7.8 days, respectively. Fenoxanil partitioned mainly into the rice plants, with half-lives of 3.3–4.4 days. The half-lives of thifluzamide, fenoxanil, and tebuconazole in paddy water were 0.17–0.89, 1.8–3.0, and 1.6–4.0 days, respectively. The residues in rice grains at the pre-harvest interval of 14 days were all below the established maximum limit values. Devils Lake is a terminal lake located in northeast North Dakota. Because of its glacial origin and accumulated salts from evaporation, the lake has a high concentration of sulfate compared to the surrounding water bodies. From 1993 to 2011, Devils Lake water levels rose by ~10 m, which flooded surrounding communities and increased the chance of an overspill to the Sheyenne River. To control the flooding, the State of North Dakota constructed two outlets to pump the lake water to the river. However, the pumped water has raised concerns about of water quality degradation and potential flooding risk of the Sheyenne River. To investigate these perceived impacts, a Soil and Water Assessment Tool (SWAT) model was developed for the Sheyenne River and it was linked to a coupled SWAT and CE‐QUAL‐W2 model that was developed for Devils Lake in a previous study. While the current outlet schedule has attempted to maintain the total river discharge within the confines of a two‐year flood (36 m3/s), our simulation from 2012 to 2018 revealed that the diversion increased the Sheyenne River sulfate concentration from an average of 125 to >750 mg/L. Furthermore, a conceptual optimization model was developed with a goal of better preserving the water quality of the Sheyenne River while effectively mitigating the flooding of Devils Lake. The optimal solution provides a “win–win” outlet management that maintains the efficiency of the outlets while reducing the Sheyenne River sulfate concentration to ≤600 mg/L. 相似文献

2008年汶川8.0级地震发生以来,极震区泥石流爆发频率有增无减且规模越来越大,破坏性越来越强,严重威胁灾区人民生命财产安全。对比采取现场勘查、遥感图像分析、历史资料对比分析等方法,分析了极震区泥石流动力特征受地形、地层、物源等因素的影响,特别是泥石流爆发过程中深侵蚀和溃决对泥石流动力特征的影响。针对四川安县高川乡流域泥石流沟的调查和分析,建立泥石流流量-冲刷模型,分析了常规地区泥石流峰值流量与极震区泥石流峰值流量差别,推导出极震区峰值流量及冲刷深度定量计算公式。通过实例分析,理论与实际具有较好的一致性,对极震区泥石流动力参数计算具有较好适用性和有效性。 相似文献
