The choice of wastewater compliance methods used in the United States has been largely prescribed; however, in some cases, this has led to data of unknown or poor quality. This problem is further compounded by the relatively slow regulatory approval process to incorporate discharge-specific method modifications or flexibility to using alternate, potentially better technologies. In this study, a framework is presented, using a performance-based-system approach, which a discharger could use to verify proper use of an alternate or modified method. An example, using two chemical oxygen demand methods (a currently approved method and an alternate method that does not generate hazardous waste) demonstrates that the protocol is simple to use, yet scientifically defensible and effective and that this approach should be readily understandable to both regulators and the regulated community. Our results also suggest that the reference method approach, without associated measurement quality objectives, may yield a false sense of competency with an alternate method.  相似文献

Ethanolic extracts from Magnolia dealbata (Zucc.) (Magnoliaceae); leaves, bark, seeds, sarcotesta and flowers were evaluated for insecticidal activity against adults of the Mexican fruit fly, Anastrepha ludens (Loew) (Diptera: Tephritidae). Using feeding bioassays composed from sugar-extract mixtures, only the extract from sarcotesta indicated insecticidal activity against the flies. The extracts from the other four plant tissues (leaves, bark, seeds and flowers) did not manifest any biological activity. The most effective extract was obtained from oven-dried sarcotesta, whereas extracts from fresh sarcotesta were inactive. Our results suggest that M. dealbata sarcotesta contains secondary metabolites with insecticidal activity against A. ludens adults. These metabolites are as potent as natural pyrethins and represent a potential substance for controlling this type of pest.  相似文献

Ecological risk assessment (ERA) evaluates potential causal relationships between multiple sources and stressors and impacts on valued ecosystem components. ERAs applied at the watershed scale have many similarities to the place-based analyses that are undertaken to develop Total Maximum Daily Loads (TMDLs), in which linkages are established between stressors, sources, and water quality standards, including support of designated uses. TMDLs focus on achieving water quality standards associated with attainment of designated uses. In attempting to attain the water quality standard, many TMDLs focus on the stressor of concern rather than the ecological endpoint or indicators of the designated use that the standard is meant to protect. A watershed ecological risk assessment (WERA), at least in theory, examines effects of most likely stressors, as well as their probable sources in the watershed, to prioritize management options that will most likely result in meeting environmental goals or uses. Useful WERA principles that can be applied to TMDL development include: development and use of comprehensive conceptual models in the Problem Identification step of TMDLs; use of a transparent process for selecting Numeric Targets for TMDLs based on assessment endpoints derived from the management goal or designated use under consideration; analysis of co-occurring stressors likely to cause beneficial use impairment based on the conceptual model; use of explicit uncertainty analyses in the Linkage Analysis step of TMDL development; and frequent stakeholder interactions throughout the process. WERA principles are currently most applicable to those TMDLs in which there is no numeric standard and, therefore, indicators and targets need to be developed, such as many nutrient or sediment TMDLs. WERA methods can also be useful in determining TMDL targets in situations where simply targeting the water quality standard may re-attain the numeric criterion but not the broader designated use. Better incorporation of problem formulation principles from WERA into the TMDL development process would be helpful in improving the scientific rigor of TMDLs.  相似文献

Juvenile Pacific giant lions-paw scallops Nodipecten subnodosus were fed the toxic dinoflagellate Gymnodinium catenatum, a producer of paralytic shellfish poison (PSP), supplied with Isochrysis galbana (a nontoxic microalgae). Short-term (<24 h) experiments were performed to determine clearance and ingestion rates of G. catenatum. Kinetics of PSP was examined in longer-term experiments (>2 days). At high food concentrations, juvenile scallops showed production of pseudofeces, partial shell valve closure, and reduction in feeding. According to HPLC analysis, the only toxins present in the dinoflagellate G. catenatum and in the scallops were the gonyautoxins (GTXs), except in the labial palps and digestive gland, where trace amounts of saxitoxin (STX) were present in scallops. These tissues could play an important role in toxin biotransformation. The ranking of toxin concentration in tissues was: digestive gland > labial palps > intestine > gills > mantle > adductor muscle, where the total contribution of viscera was more than 80% of the total toxin body burden. Juvenile scallops exhibited no apparent detrimental physiological responses during the long-term feeding experiment. The dinoflagellate may contribute nutrients to the scallop, in addition to the microalgae I. galbana. The dinoflagellate may enhance cell uptake and byssus production. Once PSP accumulated during the first 12 days, it was slowly eliminated. The limited capacity for accumulating toxins in the adductor muscle favors domestic marketing of scallops.  相似文献

Atmospheric deposition of metals emitted from mining operations has raised metal concentrations in the surrounding soils. This repository may be remobilized and act as a source of metals to nearby surface aquatic systems. It is important to understand metal dynamics and the impact of various chemistry and fate parameters on metal movement in the soil environment in order to evaluate risk associated with metals in terrestrial ecosystems and accurately establish critical discharge limits that are protective of aquatic biota. Here we extend our previously developed coupled multispecies metal fate-TRANsport and SPECiation/complexation (TRANSPEC) model, which was applicable to surface aquatic systems. The extended TRANSPEC, termed TRANSPEC-II, estimates the partition coefficient, K(d), between the soil-solid and -soluble phases using site-specific data and a semi-empirical regression model obtained from literature. A geochemical model calculates metal and species fractions in the dissolved and colloidal phases of the soil solution. The multispecies fugacity/aquivalence based fate-transport model then estimates inter-media transport rates such as leaching from soil, soil runoff, and water-sediment exchanges of each metal species. The model is illustratively applied to Ni in the Kelly Lake watershed (Sudbury, Ontario, Canada), where several mining operations are located. The model results suggest that the current atmospheric fallout supplies only 4% of Ni removed from soil through soil runoff and leaching. Soil runoff contributes about 20% of Ni entering into Kelly Lake with the rest coming from other sources. Leaching to groundwater, apart from runoff, is also a major loss process for Ni in the soil. A sensitivity analysis indicates that raising soil pH to above 6 may substantially reduce metal runoff and improve water quality of nearby water bodies that are impacted by runoff.  相似文献

We used an existing conservation opportunity area (OA) data layer for four contiguous ecological subsections within the Ozark Highlands to quantitatively evaluate the influence of conservation targets and assessment region size on conservation priorities. OAs are natural and seminatural land-cover patches that are away from roads and away from patch edges. To evaluate the influence of targets, we assigned a priority score to each OA polygon for each of five different conservation targets, including land-cover patch size, landform representation, target vertebrate richness, target breeding bird richness, and target land cover. The top-scoring OAs for each target were added to an OA selection set for that target until 50% of the study area was chosen. These five OA selection sets were overlain to quantify overlap in priorities. Only 1.6% of the study area, or 2.1% of all OA polygons, was selected by all five targets. To evaluate the influence of assessment region size, we compared results of priority ranking of OAs relative to the entire study area against a merged set of priority rankings established separately relative to each of the four subsections within the study area. When high-priority OAs were added until 25% of the region was within the selection set for each of the five targets, the sets based on the whole study area versus each subsection evaluated separately overlapped from 45.4% to 81.9%. Thus, perceived priorities of conservation assessments are strongly influenced both by the targets that are evaluated and by the size of the assessment region.  相似文献

Abstract

The effects of suspensions of zinc oxide nanoparticles at concentrations between 100 and 500?mg L?1 on germination of Capsicum chinense seeds and vigor of plants were evaluated using a randomized complete design with four replications, resulting in increased germination and improved development of lengths of plumule and radicle. The activities of peroxidase, catalase and ascorbate peroxidase were determined, showing that at all concentrations the activities of the former two enzymes were increased, whereas the activity of the latter was reduced at 500?mg L?1. Pretreatments of seeds with ZnO nanoparticles can improve germination and development of seedlings and the activities of antioxidant enzymes.  相似文献

Many organizations in the USA collect aquatic bioassessment data using different sampling and analysis methods, most of which have unknown performance in terms of data quality produced. Thus, the comparability of bioassessments produced by different organizations is often unknown, ultimately affecting our ability to make comprehensive assessments on large spatial scales. We evaluated a pilot approach for determining bioassessment performance using macroinvertebrate data obtained from several states in the Southeastern USA. Performance measures evaluated included precision, sensitivity, and responsiveness to a human disturbance gradient, defined in terms of a land disturbance index value for each site, combined with a value for specific conductance, and instream habitat quality. A key finding of this study is the need to harmonize ecoregional reference conditions among states so as to yield more comparable and consistent bioassessment results. Our approach was also capable of identifying potential areas for refinement such as reevaluation of less precise, sensitive, or responsive metrics that may result in suboptimal index performance. Higher performing bioassessments can yield information beyond “impaired” versus “unimpaired” condition. Acknowledging the limitations of this pilot study, we would recommend that performance evaluations use at least 50 sites, 10 of which are ecoregional reference sites. Efforts should be made to obtain data from the entire human disturbance gradient in an ecoregion to improve statistical confidence in performance measures. Having too few sites will result in an under-representation of certain parts of the disturbance gradient (e.g., too few poor quality sites), which may bias sensitivity and responsiveness estimates.  相似文献

In Mexican waters, there is no a formal and well-established monitoring program of harmful algal blooms (HAB) events. Until now, most of the work has been focused on the characterization of organisms present in certain communities. Therefore, the development of new techniques for the rapid detection of HAB species is necessary. Capillary electrophoresis finger print technique (CE-SSCP) is a fingerprinting technique based on the identification of different conformers dependent of its base composition. This technique, coupled with capillary electrophoresis, has been used to compare and identify different conformers. The aim of this study was to determine if CE-SSCP analysis of ribosomal RNA (rRNA) gene fragments could be used for a rapid identification of toxic and harmful HAB species to improve monitoring activities along the coasts of Baja California Sur, Mexico.Three different highly variable regions of the 18S and 28S rRNA genes were chosen and their suitability for the discrimination of different dinoflagellate species was assessed by CE-SSCP.The CE-SSCP results obtained for the LSU D7 fragment has demonstrated that this technique with this gene region could be useful for the identification of the ten dinoflagellates species of different genera.We have shown that this method can be used to discriminate species and the next step will be to apply it to natural samples to achieve our goal of molecular monitoring for toxic algae in Mexican waters. This strategy will offer an option to improve an early warning system of HAB events for coastal BCS, allowing the possible implementation of mitigation strategies. A monitoring program of HAB species using molecular methods will permit the analysis of several samples in a short period of time, without the pressure of counting with a taxonomic expert in phytoplankton taxonomy.  相似文献
