Bacterial strains with poly-3-hydroxybutyrate (PHB) degradation potential were isolated from waste yard soil samples of selected industrial sites in Uttarakhand, India, and two microbial consortia were developed, i.e. Consortium I comprises Pseudomonas sp. strain Rb10, Pseudomonas sp. strain Rb11 and Bacillus sp. strain Rb18, and Consortium II is composed of Lysinibacillus sp. strain Rb1, Pseudomonas sp. strain Rb13 and Pseudomonas sp. strain Rb19. The current study involved enrichment selection via liquid and semi-solid media, followed by isolation and screening of bacterial strains using PHB pellets and films. Furthermore, the identification and characterisation was done by triphasic approach. The utilisation of PHB by the characterised strains was confirmed by Fourier transform infrared spectroscopy and scanning electron microscopy. Moreover, the minimum inhibitory concentration of solubilised PHB was found to be 2.5?mg/mL, which was detected through ‘clear zone assay’. Further, the selection and biocompatibility testing of potential isolates were performed for the formation of bacterial consortia. Thus, the present work would provide direct and standardised protocol for screening and selection of potential microbiomes for biodegradation of polymers by overcoming the negative effect of organic solvents. Traditional Central Himalaya societies face a range of socio-economic and environmental problems. The potential of science and technology has not yet been adequately and appropriately harnessed to overcome the development constraints posed by the fragile Himalayan environment. Thus there is a need for large-scale establishment of technology resource centers. The Garhwal Unit of GB Pant Institute of Himalayan Environment and Development established rural technology demonstration and training centers in three different agroecological zones between 550 and 2200 m asl and in 13–15 locations, with suitable technologies based on appropriate use and management of locally available bio-resources that ensure people participation. About 35 on-site training courses were organised for about 2329 participants. The participants were given on-site demonstrations, training and technical know-how on various technologies, to enable farmers to better understand problems faced during implementation of new technologies for sustainable management of natural resources. An action research framework and training manuals were also developed in consideration of local socio-economic condition. The documentation and analysis of research (quantitative and qualitative) and data related to cost–benefit analysis of the technologies adopted by farmers generated through this study has created wider sharing of farmer training outputs, at farm level, amongst the scientific communities and with policy planners. 相似文献

Sprague Dawley strain of male rats weighing 200 +/- 10.0 g, were exposed intramuscularly to non-lethal dose of mercury for short acute duration of 24 and 48 hr. Mercury treatment increased thio-barbituric acid reactive substance (TBARS) and conjugated diene (CD) content with increase in duration when compared with control. This reflects possible increase in lipid peroxidation, revealing that sufficient intoxication was generated by non-lethal dose of mercury. Furthermore, mercury treatment decreased tissue glutathione (GSH) content to 2.07 and 1.49 microg GSH mg protein(-1) with concomitant decrease in glutathione-S-transferase (GST) activity by 26.06 and 36.40% after 24 and 48 hr of exposure respectively. The elevations of aspartate transaminase (AST) and alanine transaminase (ALT) levels measured exhibited increase of 287.5 and 214.5% after 48 hr of exposure respectively which were found to be highly significant compared with control. Western blot analysis indicated upregulation of caspase-9 and upsurge in effect or caspase-3 activity leading to apoptosis. The concluded findings of the present investigation suggests possible role of early mercury exposure in inducing oxidative stress mediated apoptosis in mammalian model systems as an indicator component of environmental toxicology. 相似文献

Ambient concentrations of carbon monoxide (CO), nitrogen oxides (NOx), sulfur dioxide (SO2), and total suspended particulates (TSP) were measured from January 1997 to November 1998 in the center of downtown [the Income Tax Office (ITO) located on B.S.G. Marg] New Delhi, India. The data consist of 24-h averages of SO2, NOx, and TSP as well as 8 and 24-h averages of CO. The measurements were made in an effort to characterize air pollution in the urban environment of New Delhi and assist in the development of an air quality index. The yearly average CO, NOx, SO2, and TSP concentrations for 1997 and 1998 were found to be 4810+/-2287 and 5772+/-2116 microg/m3, 83+/-35 and 64+/-22 microg/m3, 20+/-8 and 23+/-7 microg/m3, and 409+/-110 and 365+/-100 microg/m3, respectively. In general, the maximum CO, SO2, NOx, and TSP values occurred during the winter with minimum values occurring during the summer, which can be attributed to a combination of meteorological conditions and photochemical activity in the region. The ratio of CO/NOx (approximately 50) indicates that mobile sources are the predominant contributors for these two compounds in the urban air pollution problem in New Delhi. The ratio of SO2/NOx (approximately 0.6) indicates that point sources are contributing to SO2 pollution in the city. The averaged background CO concentrations in New Delhi were also calculated (approximately 1939 microg/m3) which exceed those for Eastern USA (approximately 500 microg/m3). Further, all measured concentrations exceeded the US National Ambient Air Quality Standards (NAAQS) except for SO2. TSP was identified as exceeding the standard on the most frequent basis. 相似文献

High performance liquid chromatographic (HPLC) method was developed for analysis of seven gibberellins, i.e., GA3, GA4, GA7, GA3 methyl ester, GA7 methyl ester 3,13 diacetate, GA7 methyl ester, and fusaric acid, using an isocratic system. Method was used for estimation of gibberellins from different Fusarium strains. Gibberellins were extracted from 28 strains of Fusarium, out of which six strains of Fusarium were isolated from soil of different parts of India and 22 strains were procured from the Indian Type Culture Collection, Indian Agricultural Research Institute, New Delhi. Extracts were analyzed for qualitative and quantitative estimation of gibberellins by thin layer chromatography and HPLC, respectively. On the basis of quantitative analysis of produced gibberellins by HPLC, they were categorized as low, moderate, and high gibberellin producing strain. Dynamics of toxic heavy metals in different compartments of a highly urbanized closed aquatic system
This paper deals with the dynamics of chromium, nickel, copper and lead among the different components namely water, surface sediments, submerged and free floating macrophytes and fish of the twin manmade lakes, Upper and Lower lakes, of Bhopal (M.P., India). Some basic parameters of water and sediment have also been studied. The basin of the lake system is densely populated and the water is used for various purposes including drinking. Ni and Pb along with nitrate in both lakes are significantly higher than the drinking water quality criteria of USEPA. The concentration of the metals in the sediments is noticeably higher than that present in the adjoining rock, particularly Ni and Pb. There is a significant uptake of metals by the macrophytes (Eichhornia crassipes and Hydrilla verticillata) and fish (Labeo rohita and Oreochromis niloticus) mainly in summer. The fish of Lower lake (O. niloticus) is unfit for human consumption. The data have been statistically treated. Principle component analysis and cluster analysis were performed to define the origin of metals and to assess the relationship among the sites. Overall the Lower lake is more polluted than the Upper lake. In aggregate, the lake system is under an environmental stress due to certain practices. 相似文献

Aerosol (total suspended particulate) samples collected at three diverse locations (urban-commercial, semi-urban and rural-agricultural) in Patiala, India were analyzed for loss on ignition (LOI) and organic tarry matter (OTM) content in ambient air during crop residue burning (CRB) episodes and non-crop residue burning (NCRB) months in 2006–2007. Results showed high levels of LOI and OTM during wheat and rice crop residue-burning periods at all the sites. Higher levels were obtained during rice crop residue-burning period as compared to the wheat residue-burning period. At semi-urban site, LOI varied between 53 ± 36 μg m?3 and 257 ± 14 μg m?3 constituting 38–78% (w/w) part of the aerosols whereas levels of OTM varied between 0.98 ± 0.11 μg m?3 and 7.93 ± 2.76 μg m?3 comprising 0.42–3.28% (w/w) fraction. At rural-agricultural area site, levels of LOI varied between 86 ± 40 μg m?3 and 293 ± 70 μg m?3 comprising 27–84% (w/w), whereas OTM levels varied between 1.31 ± 0.64 μg m?3 and 10.09 ± 6.56 μg m?3 constituting 0.83–2.42% (w/w) fraction of the aerosols. At urban-cum-commercial site, levels of LOI and OTM varied between 48 ± 23 μg m?3 and 281 ± 152 μg m?3 and 2.53 ± 1.23 μg m?3 and 17.40 ± 8.50 μg m?3, constituting 24–62% (w/w) part of the aerosols, respectively. Results also indicated that OTM and LOI were integral parts of aerosols and their concentrations were influenced by the crop residue burning practices with incorporated effect of vehicular activities in Patiala. 相似文献

The utilization of sustainable and biodegradable lignocellulosic fiber to detoxify the noxious Cr(VI) from wastewater is considered a versatile approach to clean up a contaminated aquatic environment. The aim of the present research is to assess the proficiency and mechanism of biosorption on Ficus carica bast fiber via isotherm models (Langmuir, Freundlich, Temkin, Harkin’s–Jura, and Dubinin–Radushkevich), kinetic models, and thermodynamic parameters. The biomass extracted from fig plant was characterized by scanning electron microscopy and Fourier-transform infrared spectroscopy. To optimize the maximum removal efficiency, different parameters like effect of initial concentration, effect of temperature, pH, and contact time were studied by batch method. The equilibrium data were best represented by the Langmuir isotherm model, and the maximum adsorption capacity of Cr(VI) onto biosorbent was found to be 19.68 mg/g. The pseudo-second-order kinetic model adequately described the kinetic data. The calculated values of thermodynamic parameters such as enthalpy change (?H 0), entropy change (?S 0), and free energy change (?G 0) were 21.55 kJ/mol, 76.24 J/mol?K, and ?1.55 kJ/mol, respectively, at 30 °C which accounted for spontaneous and endothermic processes. The study of adsorbent capacity for Cr(VI) removal in the presence of Na+, Mg2+, Ca2+, SO 4 2? , HCO 3 ? and Cl? illustrated that the removal of Cr(VI) increased in the presence of HCO3? ions; the presence of Na+, SO 4 2? or Cl? showed no significant influence on Cr(VI) adsorption, while Ca2+ and Mg2+ ions led to an insignificant decrease in Cr(VI) adsorption. Further, the desorption studies illustrated that 31.10 % of metal ions can be removed from an aqueous system, out of which 26.63 % of metal ions can be recovered by desorption in first cycle and the adsorbent can be reused. The results of the scale-up study show that the ecofriendly detoxification of Cr(VI) from aqueous systems was technologically feasible. 相似文献
