- DOI: http://dx.doi.org/10.1065/espr2006.01.006 Background The use of vegetal organisms as indicators of contamination of the environment is partially replacing traditional monitoring techniques. Amongst the vegetal organisms available, mosses appear to be good bioindicators and are used for monitoring anthropogenic and natural fall-out on soils. This study has two objectives: the evaluation of the concentrations of heavy metals in soils and mosses of the Sicily Region, in Italy and the identification of the origin of fall-out of heavy metals. Methods Mosses and the surface soil were sampled at 28 sites, only the youngest segments of Hylocomium splendens and Hypnum cupressiforme, corresponding to the plant tissues produced during the last 3 years, were taken. The elements Cd, Cu, Ni, Pb and Zn were analysed by ICP-MS and Hg by AAS. Statistical analysis was by PCA and spatial representation by GIS. Results and Discussion In the mosses sampled in Sicily, the highest concentrations of Cd were found around the cities of Palermo and Messina. The highest concentrations of Hg were recorded in the northern part of the island between Trapani and Messina, similar to the distribution of Cu. Different areas with the highest concentrations of Ni were found near the south coast, in the vicinity of Palermo and around the Volcano Etna. The highest concentrations of Pb were found in the south-west coast near Agrigento, where important chemical plants and petroleum refineries are located. Except for a few locations, Zn fall-out was found to be evenly distributed throughout Sicily. Conclusion The sites where the concentrations of heavy metals cause greatest concern have been revealed by the PCA analysis and portrayed using GIS. Also of some concern is the diffuse and anthropogenic origin of Hg and Cd. The combined approach of using soil and mosses, together with pedological interpretation and application of multivariate statistical techniques has provided valuable insight into the environmental aspects of heavy metal deposition in a region of southern Europe. Recommendations and Outlook Further insight into the deposition of heavy metals will require more detailed sampling of soils and mosses in both new and previous study areas. This needs to be complemented by detailed pedological investigations in the study areas. Future research programmes will address these issues.  相似文献

The development of approaches to estimate the vulnerability of biological communities and ecosystems to extirpations and reductions of species is a central challenge of conservation biology. One key aim of this challenge is to develop quantitative approaches to estimate and rank interaction strengths and keystoneness of species and functional groups, i.e. to quantify the relative importance of species. Network analysis can be a powerful tool for this because certain structural aspects of ecological networks are good indicators of the mechanisms that maintain co-evolved, biotic interactions. A static view of ecological networks would lead us to focus research on highly-central species in food webs (topological key players in ecosystems). There are a variety of centrality indices, developed for several types of ecological networks (e.g. for weighted and un-weighted webs). However, truly understanding extinction and its community-wide effects requires the use of dynamic models. Deterministic dynamic models are feasible when population sizes are sufficiently large to minimize noise in the overall system. In models with small population sizes, stochasticity can be modelled explicitly. We present a stochastic simulation-based ecosystem model for identification of “dynamic key species” in situations where stochastic models are appropriate. To demonstrate this approach, we simulated ecosystem dynamics and performed sensitivity analysis using data from the Prince William Sound, Alaska ecosystem model. We then compare these results to those of purely topological analyses and deterministic dynamic (Ecosim) studies. We present the relationships between various topological and dynamic indices and discuss their biological relevance. The trophic group with the largest effect on others is nearshore demersals, the species mostly sensitive to others is halibut, and the group of both considerable effect on and sensitivity to others is juvenile herring. The most important trophic groups in our dynamical simulations appear to have intermediate trophic levels.  相似文献

Objective

To develop a multi-step workflow for the isolation of circulating extravillous trophoblasts (cEVTs) by describing the key steps enabling a semi-automated process, including a proprietary algorithm for fetal cell origin genetic confirmation and copy number variant (CNV) detection.

Methods

Determination of the limit of detection (LoD) for submicroscopic CNV was performed by serial experiments with genomic DNA and single cells from Coriell cell line biobank with known imbalances of different sizes. A pregnancy population of 372 women was prospectively enrolled and blindly analyzed to evaluate the current workflow.

Results

An LoD of 800 Kb was demonstrated with Coriell cell lines. This level of resolution was confirmed in the clinical cohort with the identification of a pathogenic CNV of 800 Kb, also detected by chromosomal microarray. The mean number of recovered cEVTs was 3.5 cells per sample with a significant reverse linear trend between gestational age and cEVT recovery rate and number of recovered cEVTs. In twin pregnanices, evaluation of zygosity, fetal sex and copy number profiling was performed in each individual cell.

Conclusion

Our semi-automated methodology for the isolation and single-cell analysis of cEVTS supports the feasibility of a cell-based noninvasive prenatal test for fetal genomic profiling.  相似文献

The relationship between fetal renal function (FRF) and fetal serum beta2-microglobulin (B2MG) was investigated by comparing its value in 112 unaffected fetuses with that of 23 fetuses presenting with urinary tract malformations (UTM). Fetal serum level of B2MG was totally unrelated to gestational age; its value increased in cases of severe impairment of FRF but was similar to controls in all mild uropathies (p<0.05). Evaluating serum B2MG could be beneficial in fetuses with severe renal damage, but is of no use in unilateral UTM since only the global FRF is tested and not the function of each single kidney. Nine polycyclic aromatic hydrocarbons (PAHs) were analysed in pine needles of different ages (from 6 to 30 months) collected from two species, Pinus nigra and Pinus pinaster, in seven sites located along a transect from a suburban to a rural area of Genoa (Italy). In all sites and for both species, concentrations of more volatile PAHs (phenanthrene, anthracene, fluoranthene, pyrene) were higher than those for other less volatile PAHs, which are preferentially sorbed to airborne particulates (benzo[a]anthracene, chrysene, benzofluoranthenes, benzo[a]pyrene). Concentrations of total PAHs found in P. nigra in the rural sites were, on the average, 2.3 times higher than those in P. pinaster growing nearby. In both pine species, concentrations of volatile PAHs increased according to needle age. Annual trends of other PAHs were more variable, with a general decrease in older needles. P. pinaster needles are shown to be more reliable passive samplers, since they are more resistant to plant diseases, and considerable variation in PAH concentration was observed in P. nigra needles with moulds and fungi.  相似文献

A study was done to evaluate the effects of diagnostic ultrasound on sister chromatid exchange (SCE) in first-trimester chorionic villi under controlled technical conditions. Chromosome analysis was performed by the direct method using spontaneous mitoses from the cytotrophoblast layer, and SCE visualization was accomplished by a 72 h treatment with 5-bromodeoxyuridine (BrdU) at a concentration of 10μg/ml. The slides were stained with ac ridine orange. Immediately before first-trimester chorionic villus sampling, a group of ten pregnant women was exposed to diagnostic ultrasound for 20min (in vivo exposure). This group of patients was compared with a control group who were not exposed. A mean value of SCE/cell frequency of 4·2 ± 0·2 was found in the exposed pregnancies, while a value of 3·7 ± 0·2 was observed in the control group. After in vitro exposure of chorionic villi obtained from elective abortions, the frequency of SCE/cell did not differ significantly among samples with different exposures (1,2, and 3 h) and controls. The positive control (mitomycin C) yielded a significant increase in SCE frequency.  相似文献
