Until now, little is known about the influence of food additives on heat inactivation of noroviruses. Only a few studies have shown a protective or inhibiting effect on virus infectivity caused by the food matrix. Therefore, the aim of this study was to examine the influence of sodium chloride, sucrose and milk on heat stability of the surrogates murine norovirus (MNV) and MS2 phage at 60 °C for 1–5 min in PBS for MNV and for 5–120 min in suspension medium buffer for MS2 phage. Different concentrations of sodium chloride (5, 10 %) and sucrose (5, 50 %) were added to the respective buffers. In addition, commercially available milk with different fat concentrations (0.3, 1.5, 3.5 %) was investigated in this study. In general, a linear titre reduction for MNV and MS2 phage could be observed, except for the heat treatment of MNV in PBS with 50 % sucrose. A protective effect of PBS with 50 % sucrose and of the matrix milk on MNV could be concluded. All other tested conditions did not show any influence on virus inactivation. However, MS2 phage did show a higher heat resistance throughout the experiments compared to MNV. In future investigations, it should be tested, whether the achieved data may be considered in risk assessments of heat-treated food products with high concentrations of sugar. Furthermore, it should be clarified, whether these results can also be referred to complex food matrices.  相似文献

X-ray crystallography and NMR provide insight into the three-dimensional structure of proteins at high resolution yielding a relatively small number of stable "topologies". Their free energies of stabilization are minute compared to the total molecular energy. Molecular adaptation to extremes of physical conditions at the protein level refers to marginal alterations of the intramolecular interactions; clearcut predictions with respect to the correlation of protein structure and protein stability are at present not possible. The acquisition of the native structure of proteins is determined by the kinetically accessible minimum of potential energy. The overall mechanism may be described as a hierarchical condensation with elements of secondary/super-secondary structure and subdomains/domains as intermediate stages. In the case of oligomeric and multimeric proteins, folding and association of subunits follow a consecutive uni-bimolecular mechanism, the kinetics of which can be simulated with high precision. Nucleic acid hybridization techniques (e.g., Southern blotting and in situ hybridization) gain increasing importance in laboratory medicine, as demonstrated by examples in this paper. The molecular basis of this approach to detect nucleic acids homologous to known DNA probes is explained. It is concluded that this technique has advantages in the detection of pathogens compared to other methods used in laboratory medicine.  相似文献

A fetus with mucopolysaccharidosis type IV A (Morquio type A) is described. The family had one affected child exhibiting symptoms of classical Morquio A disease, and late in the subsequent pregnancy prenatal diagnosis was requested. At 23 weeks' gestation, moderate ascites was detected by detailed ultrasound scan and keratan sulphate was found in the amniotic fluid. The pregnancy was terminated by prostaglandin induction and the diagnosis of mucopolysaccharidosis type IV A was confirmed by demonstration of a deficiency of N-acetylgalactosamine-6-sulphate (GalNac-6-S) sulphatase in cultured amniotic cells and in post-mortem fibroblast cultures. The activities of β-galactosidase and arylsulphatase A were normal, ruling out Morquio disease type B and multiple sulphatase deficiency. These results indicate that mucopolysaccharidosis IV A (a disease that predominantly affects the skeletal system) may produce ascites in the fetus to such an extent that it can be detected by ultrasound.  相似文献

Using four dynamic criteria, the following environmental changes were evaluated: Global warming; destruction, of the stratospheric ozone layer; eutrophication and acidification of aquatic ecosystems; photochemical smog; reduction of the available area of agricultural soils; loss of species diversity; consumption of resources; reduction of crop yield; forest decline; noise, and smell. These ecosystem changes, or negative effects, were characterized by four dynamic criteria: 1) Regional importance, 2) extent of alteration, 3) resilience and 4) irreversibility. A multi-criteria assessment, accounting simultaneously for all four criteria, requires a generalized concept of order. Here, the concept of partially ordered sets, visualized by Hasse diagrams, was used to derive a priority list of environmental changes. Highest priority was equally assigned to the loss of species diversity, the consumption of resources and the destruction of the ozone layer. However, these ecosystem changes are incomparable to each other concerning the four criteria used and thus require different management strategies.  相似文献
