It has been previously reported that a low or absent maternal serum unconjugated estriol (uE3) level is associated with placental steroid sulfatase (STS) deficiency. Here we report a correlation between patients who present with a very low or absent maternal serum uE3 and a deletion of the STS gene as assessed by fluorescence in situ hybridization (FISH). We studied nine prenatal cases that presented to the clinical laboratory with an abnormal triple screen, specifically low or absent maternal serum uE3 and a 46,XY karyotype. FISH analysis showed complete deletion of a probe containing the STS gene in six cases and one case had a partial deletion (reduced but not absent signal). The remaining two cases were not deleted for the STS probe. All mothers tested whose fetus showed a deletion were shown to be STS deletion carriers using FISH. Biochemical analysis was performed on 7/9 prenatal specimens. All fetuses deleted for the STS probe were also found to be deficient for STS by biochemical analysis of cultured amniotic fluid (5/5). Of the two fetuses not deleted for the STS probe, one was deficient for STS activity, while the other had a normal result. The abnormal result of enzyme deficiency by biochemical analysis in a non-deletion case likely represents a mutation in the STS gene, not detectable by this FISH assay. While new environmental policies and procedures often are developed incrementally, they can also result from crises or other significant events. In situations where policies and procedures are introduced in response to a crisis, questions about the strengths and weaknesses of existing mechanisms, and the extent to which they can be used to address concerns, may be ignored. This paper explores the complexities of introducing new policies and processes where planning systems and procedures already exist. Drinking water source protection policies that are being developed in response to the tragic events in Walkerton, Ontario, Canada serve as the context for the inquiry. Three case study watersheds were selected to reflect the diversity of municipal jurisdictions and water supply systems in Ontario. A content analysis was undertaken on regulatory and non-regulatory policy documents to determine the extent to which they addressed elements of the multi-barrier approach for drinking water safety. Findings from the research reveal considerable evidence of the multi-barrier approach in the policy and guiding documents analyzed. Policy development in response to a crisis can advance progress on the issue of drinking water safety and coincide with emerging governance strategies. Policy effectiveness may be enhanced by considering existing policies as well as contextual and jurisdictional differences. 相似文献

Due to their biological, chemical and physical actions, earthworms can be directly employed within bioremediation strategies to promote biodegradation of organic contaminants. Earthworms have been shown to aerate and bioturbate soils and improve their nutritional status and fertility, which are variables known to limit bioremediation. Earthworms have also been shown to retard the binding of organic contaminants to soils, release previously soil-bound contaminants for subsequent degradation, and promote and disperse organic contaminant degrading microorganisms. This review discusses these earthworm actions upon the soil environment and how they might influence the fate and behaviour of soil associated organic contaminants, subsequently improving bioremediation potential. A fashionable field of enquiry – the relationship between expressed attitudes and actual behaviour in the context of sustainable development – is complex (Staats et al. 2004) and difficult to apply in a policy-relevant manner (Aall and Norland 2005). However, recent research indicates that the use of a household diary can be beneficial in helping to quantify household environmental impact, in educating householders about their impact, and in identifying major ‘behavioural turning points’, where householders may focus efforts to reduce their environmental impact (Hunter et al. 2006). In other words, the use of a diary by householders is a potentially powerful tool in encouraging and facilitating desired behavioural change. This paper reports on the initial findings of an innovative study that assessed the use of a household diary approach as a means of framing and collecting household environmental data and, critically, as an educational vehicle for bringing about behavioural change. Based on the initial findings, it is clear that there is some potential to develop practical policy measures that empower householders by allowing them to better grasp their environmental impact and, consequently, recoup positive implications in terms of financial savings (e.g. reducing the amount of food thrown out) and health benefits (e.g. more walking/cycling). 相似文献

Variability in the Southern Ocean is frequently reflected in changes in the abundance of Antarctic krill Euphausia superba and subsequent effects on dependent predators. However, the nature and consequences of changes in krill population dynamics
that accompany fluctuations in its abundance are essentially unknown. A conceptual model, developed from quantitative measures
of krill length in the diet of predators at South Georgia from 1991 to 1997, allowed predictions to be made about the abundance
and population structure of krill in 1998 and the consequences for predators. Consistent with model predictions, in 1998 there
was a serial change in krill population structure, low krill biomass and low reproductive performance of predators. The change
in the modal size of krill, from 56 mm in December to 42 mm in March, was apparently a result of the transport of krill into
the region. This is the first occasion when the future status and structure of the krill population at South Georgia has been
successfully predicted. By representing local krill population dynamics, which may also reflect large-scale physical and biological
processes, predators have a potential key role as indicators of environmental variation in the Southern Ocean at a range of
spatial scales.

Despite numerous reviews suggesting that microbial biosensors could be used in many environmental applications, in reality they have failed to be used for which they were designed. In part this is because most of these sensors perform in an aqueous phase and a buffered medium, which is in contrast to the nature of genuine environmental systems. In this study, a range of non-exhaustive extraction techniques (NEETs) were assessed for (i) compatibility with a naphthalene responsive biosensor and (ii) correlation with naphthalene biodegradation. The NEETs removed a portion of the total soil naphthalene in the order of methanol > HPCD > βCD > water. To place the biosensor performance to NEETs in context, a biodegradation experiment was carried out using historically contaminated soils. By coupling the HPCD extraction with the biosensor, it was possible to assess the fraction of the naphthalene capable of undergoing microbial degradation in soil. 相似文献

Advances towards sustainable land management necessitate application of a broader portfolio of decision-support tools that improve evaluation of contaminated land. Over the last decade regulators have directed concerted effort towards rationalization of risk-based contaminated land policies recognizing bioavailability and bioaccessibility as concepts to be incorporated into risk assessments. The desire for a precise and rapid method to inform consideration of bioavailability and bioaccessibility to support risk assessment of contaminated land has never been greater. This study presents a comprehensive appraisal of both emerging non-exhaustive extraction techniques (subcritical water extraction and Brij 700 extraction) developed to reflect polycyclic aromatic hydrocarbon (PAH) bioaccessibility to microorganisms as well as formerly demonstrated methodologies (the use of cyclodextrins and butanol extraction). Application of unified evaluation criteria across different techniques enabled comparison not only from the bioaccessibility prediction perspective but also analysis of economical (cost of extraction) and practical (such as extraction time) measures. Whilst the use of cyclodextrins was the best predictor of the bioaccessible fraction for the majority of compounds, other methods appeared more cost- and time-effective. Juxtaposition of the techniques presented in this study assists establishing cost-benefit trade-offs of different non-exhaustive extraction techniques and contributes to tailoring information on contaminant bioaccessibility to support risk evaluation on contaminated sites. 相似文献

Taxonomic composition, biomass as organic carbon, numerical abundance, and size distribution of the microplankton were determined at 6 Southern California nearshore locations in late May–early June, 1970. Samples were taken at approximately 5 m (10 m at one station) intervals through the upper 40 to 50 m to reveal some of the small-scale differences and levels of variability in the populations. Total microplankton biomass over all euphotic zone samples varied by more than two orders of magnitude (7.6 to 1,200 g C l-1). Average biomass at comparable sites (n=5) ranged from 48 to 240 g C l-1; biomass range within stations varied from about 5-fold to 120-fold. Total microplankton numbers varied approximately 22-fold (4.3×105 to 9.5×106 organisms l-1) over all euphotic zone samples, but the range within stations was always less than an order of magnitude. At comparable stations, nanoplankton biomass had ranges extending from 3.7-fold to 12-fold; its average percentage contribution (±1 SD) to the total microplankton biomass varied from 39±5% to 54±13%. Netplankton biomass showed a similar minimal range, but its greatest range was more than two orders of magnitude. Ranges of abundance of major taxonomic groups within stations varied considerably from about 2-fold to more than three orders of magnitude. Electron micrograph examination of the leaf and stem surfaces of Cleome viscosa L (Family Capparaceae) revealed the presence of secretory glandular trichomes with club-cylinder and cylinder morphologies. In the present study, the leaves and stems of C. viscosa were extracted with hexane and the extract was evaluated for the following biological activities: anti-bacterial, anti-fungal, contact insecticidal and nematicidal. The extract was found to be a potent anti-bacterial agent according to the thin layer chromatography autobiographic assay. Activity-directed isolation studies of the anti-bacterially active compounds led to a 14-member ring cembranoid diterpene being identified as one of the effective agents. Minimum inhibitory concentration (MIC) values (µg/spot) of 5.0 µg/spot and 1.0 µg/spot were found for the diterpene on Bacillus subtilis (Gram-positive) and Pseudomonas fluorescens (Gram-negative), respectively. The diterpene did not inhibit the growth of the fungus Cladosporium cucumerinum. The extract demonstrated a pyrethroid type of contact insecticidal activity on adult Cylas formicarius elegantulus Summer (Coleoptera: Curculionidae). The extract also had high nematicidal activity with a percentage Abbott's value of 72.69 on the plant parasitic nematode Meloidogyne incognita Chitwood; however, the extract lost its potency upon subfractionation. 相似文献

The interannual variations in abundance of the calanoid copepod Centropages typicus (Krøyer) in the North Sea are significantly positively correlated with sea-surface temperature. The same is true of Calanus helgolandicus Claus. In both cases there was a deviation from the close relationship with temperature for several years in the early to mid-1980s. Centropages typicus was less abundant than expected from 1981 to 1984, but Calanus helgolandicus recovered from a period of lower than expected abundance from 1980 to 1982, to become more abundant than expected from 1983 to 1987. The difference between the responses of the two species to climate-driven changes in seasonal cycles of phytoplankton is interpreted in relation to their biology. Geographical variations of the interannual variability and seasonal cycles of Centropages typicus in the North Sea are analysed. The centre of the population of this species in the North Sea is in the German and Southern Bights, but the variations in abundance may be partly influenced by advection from areas to the west of Britain. 相似文献
