借碱洗脱法证明:具有致突变能力的致癌剂1,2-二溴乙烷和肼在经过代谢活化后,均剂理相关地引起L1210细胞中DNA的股间交联。相反,动物致癌试验表现阴性的突变剂溴乙烷,以及通过各种致癌动物试验均显示阴性的传统常用致突变剂羟胺,在代谢活化以后的同样条件下均不能引起L1210细胞中DNA的股间交联。这证明了双区理论的致癌观点,即致癌剂必须是双官能烷化剂,其同时引起互补碱对的交联将引起癌变。相反,股内单一碱基的变异则可能引起致突变作用。并用APCI/SIM(大气压化学电离/选择离子质谱)证明:1,2-二溴乙烷与DNA碱基对的模型反应,与致突实验一致主要引起G-C对的交联和发生G-C→A-T突变。 相似文献

Phytoplankton diversity and abundance in estuarine systems are controlled by many factors. Salinity, turbidity, and inorganic nutrient concentrations and their respective ratios have all been proposed as principal factors that structure phytoplankton diversity and influence the emergence of potentially toxic species. Although much work has been conducted on temperate estuaries, less is known about how phytoplankton diversity is controlled in tropical, monsoonal systems that are subject to large, seasonal shifts in hydrology and to rapidly changing land use. Here, we present the results of an investigation into the factors controlling phytoplankton species composition and distribution in a tropical, monsoonal estuary (Bach Dang estuary, North Vietnam). A total of 245 taxa, 89 genera from six algal divisions were observed. Bacillariophyceae were the most diverse group contributing to 51.4 % of the microalgal assemblage, followed by Dinophyceae (29.8 %), Chlorophyceae (10.2 %), Cyanophyceae (3.7 %), Euglenophyceae (3.7 %) and Dictyochophyceae (1.2 %). The phytoplankton community was structured by inorganic nutrient ratios (DSi:DIP and DIN:DIP) as well as by salinity and turbidity. Evidence of a decrease in phytoplankton diversity concomitant with an increase in abundance and dominance of certain species (e.g., Skeletonema costatum) and the appearance of some potentially toxic species over the last two decades was also found. These changes in phytoplankton diversity are probably due to a combination of land use change resulting in changes in nutrient ratios and concentrations and global change as both rainfall and temperature have increased over the last two decades. The subject of energy choices and public policy is one that has interested me at a professional level. The outlook for energy supply for Americans is uncertain. Will the OPEC crank the per barrel price of world oil up again this fall by three or four dollars? We don’t know. Will the cost of finding and developing new domestic oil reserves continue to rise, and will the domestic production continue to fall as it has been doing since 1970? We don’t know. Would regulating the price of old oil and natural gas entering into interstate commerce bring forth significantly increased supplies? We don’t know. The chances for reducing oil consumption—or at least checking the rate of increase—are equally uncertain. Will still higher prices, brought on by whatever means, cause people and industries to use less oil? We don’t know. Will demand shift away from oil and natural gas to cheaper, more plentiful energy sources? We don’t know. Will penalties, taxes, or exhortations get the motor vehicle industry to produce more gasoline efficient cars and trucks? We don’t know. We may not even get a chance to try. Lipophilic contaminants are present in the environment and bioaccumulate in the food chain. Therefore, their intake via animal fat of various sources was assessed for three age groups of the Flemish population, participating in a large biomonitoring program of the Flemish government. In total, 1636 adolescents (14-15 years), 1186 mothers (18-44 years), and 1586 adults (50-65 years) participated in the study and completed a semi quantitative food frequency questionnaire. Individual consumption data were combined, via a so-called simple distribution approach, with recent data on polychlorinated dibenzo-p-dioxins/furans and dioxin-like polychlorinated biphenyls, measured via the chemical-activated luciferase gene expression (CALUX) bio-assay in food items available on the Flemish market. The median (95th percentile) estimated intakes of dioxin-like contaminants were 2.24 (4.61), 2.09 (4.26), and 1.74 (3.53) pg CALUX-TEQ kg(-1) bw d(-1) for, respectively adolescents, mothers and adults. These data are in the same range as those found in other European studies. The CALUX-TEQ results of respectively 59.8%, 53.7% and 36.2% of the adolescent, mother and adult population exceed the tolerable weekly intake (TWI) of 14 pg WHO-TEQ kg(-1) bw w(-1), as derived by the Scientific Committee on Food [Scientific Committee on Food, 2001.
