A procedure for the quantification of the PCB marker congeners 28, 52, 101, 138, 153, 180 in plastic waste form the recycling of cables in the range of content between 0.1 mgkg(-1) and 20 mgkg(-1) was validated. Representative reference materials for laboratory analyses were produced using granulates from different recycling processes. The method development included aspects of grinding, sieving, homogenising as well as the comparison of extraction methods and chromatographic conditions. The validated procedure foresees grinding of the technical sample to an upper particle size limit of 0.50mm followed by extraction with an unpolar solvent such as n-hexane and gas chromatographic determination. LOD and LOQ for individual congeners ranged between 0.01 mgkg(-1) and 0.08 mgkg(-1). Electron capture detection (ECD) and mass selective detection (MS) were equivalent if chromatographic conditions were appropriately selected. The application of MS was advantageous in presence of significant amounts of interfering components which have been found in some cables. Three reference materials containing the marker congeners in the range of 0.1 mgkg(-1)-5 mgkg(-1) were submitted to a final validation intercomparison with 23 selected field laboratories using the developed method. Relative reproducibility standard deviations varied between 15% and 33%.  相似文献

Soil functional stability is the capacity of soil functions to resist and recover from an environmental perturbation and can be used to evaluate soil health. It can be influenced by the presence of xenobiotics such as herbicides. The impact of a fresh 2,4-D contamination (36 mg kg(-1) dry soil) on soil functional stability was evaluated by comparing the capacity of soil enzyme activities to resist and recover from a heat perturbation for both a clean and 2,4-D-contaminated soil. The functional stabilities of the soils (uniform sands, pH 6.9, 7% (w/w) organic matter) were calculated using the relative soil stability index (RSSI). The RSSI scores indicate the proportion of potential enzyme activity the soil retains after a perturbation compared to the potential activity of an unperturbed soil. Six extra-cellular enzyme activities (acid and alkaline phosphatases, arylsulfatase, urease, protease and beta-glucosidase) were monitored in soil microcosms during a 15-day period. During this period, a 60 degrees C heat perturbation was applied to the soil for 24 h. The activities of arylsulfatase and protease were found to be the most stable following heat perturbation obtaining the highest RSSI scores (87% and 77%, respectively). Urease activity showed the lowest RSSI score (38%). Although all enzyme activities were inhibited by the presence of 2,4-D, the RSSI results indicated that contamination lowered the stability of only three enzyme activities (arylsulfatase, beta-glucosidase and urease). The RSSI adequately described resistance, recovery and recovery rate parameters and enabled differentiation between functional stabilities of clean and contaminated soil and between different soil types.  相似文献

The aim of the present study was to determine the kinetic parameters and cholinesterase (ChE) activity in two teleost fish, the white mouth croaker Micropogonias furnieri (Scianidae) and the Madamango sea catfish Cathorops spixii (Ariidae), to verify their suitability as sentinel species of aquatic pollution by anticholinergic compounds. Individuals of each species were captured in one reference and one polluted site in the Southern Brazilian coast. Brain tissue was used as enzyme source. Inhibition kinetic parameters indicated that ChE from C. spixii collected at the reference site showed more affinity (Ka) for eserine that from those collected at the polluted site. The opposite was observed for the carbamylation constants (Kc). Thus, similar inhibition constants (Ki) were observed. M. furnieri brain showed an extremely low sensitivity to in vitro inhibition by eserine, indicating that it is not a suitable biomarker to be employed in environmental monitoring of anticholinergic compounds. Results from the present study also point to the need for considering kinetic analysis when cholinesterase activity is employed as a biomarker.  相似文献

The physiological responses of the acid-sensitive amphipod Gammarus fossarum exposed in situ to acid stress (pH4.5 and 5.5) and then transferred back to neutral water were investigated. Survival rate and haemolymph [Cl(-)] and [Na(+)] were assessed after 24, 48 and 72h of exposure in acidic streams and after a recovery period of 12, 24, 36, 48 and 60h. After 24h, exposure to slightly acidic (pH5.5) and strongly acidic water (pH4.5) led to a severe and significant depletion in haemolymph [Na(+)] and [Cl(-)] compared to organisms exposed in circumneutral water (pH7.3). However, after only a 12h-period of transfer back in neutral water and whatever the previous exposure time (24, 48 and 72h) in both slightly and strongly acidic water, haemolymph [Na(+)] and [Cl(-)] were equal or superior to the control level without associated mortality. In spite of this fast physiological recovery capacity, populations of G. fossarum living in streams undergoing episodic acid stresses were drastically affected thus, demonstrating the high acid-sensitivity of this species. We discuss the possible reasons of population regression and the absence of population recovery.  相似文献

Urban atmospheres contain complex mixtures of air pollutants including mutagenic and carcinogenic substances such as benzene, diesel soot, heavy metals and polycyclic aromatic hydrocarbons. In the frame of a European network for the assessment of air quality by the use of bioindicator plants, the Tradescantia micronucleus (Trad-MCN) test was applied to examine the genotoxicity of urban air pollution. Cuttings of Tradescantia clone #4430 were exposed to ambient air at 65 monitoring sites in 10 conurbations employing a standardised methodology. The tests revealed an elevated genotoxic potential mainly at those urban sites which were exposed to severe car traffic emissions. This bioassay proved to be a suitable tool to detect local 'hot spots' of mutagenic air pollution in urban areas. For its use in routine monitoring programmes, however, further standardisation of cultivation and exposure techniques is recommended in order to reduce the variability of results due to varying environmental conditions.  相似文献

About 20,000 Chinook salmon (Oncorhynchus tshawytscha) from Lake Ontario enter the Credit River, Ontario, Canada every fall to spawn and die. In this study, samples of muscle and eggs collected from female Chinook salmon entering the Credit River contained total PCBs, DDT compounds and other organochlorine (OC) compounds at mug/kg concentrations. Semi-permeable membrane devices (SPMDs) were deployed at a reference site above the spawning grounds and at two downstream sites at intervals over a 14-month period that spanned two spawning runs. There was an increase in the concentrations of total PCBs, total DDT and other classes of OCs in the SPMDs deployed at the two downstream sites during and after both spawning runs; indicating that the decay of salmon releases contaminants into the river. Based upon the concentrations of contaminants in the salmon tissues, approximately 75 g of total PCBs and 35 g of total DDT compounds would be transported annually into the Credit River from this source.  相似文献

Samples of the aquatic bryophyte Fontinalis antipyretica Hedw. were transplanted to different sites with the aim of characterizing the kinetics of the uptake and discharge of heavy metals in the extra and intracellular compartments. The accumulation of metals in extracellular compartments, characterized by an initial rapid accumulation, then a gradual slowing down over time, fitted perfectly to a Michaelis-Menten model. The discharge of metals from the same compartment followed an inverse linear model or an inverse Michaelis-Menten model, depending on the metal. In intracellular sites both uptake and discharge occurred more slowly and progressively, following a linear model. We also observed that the acidity of the environment greatly affected metal accumulation in extracellular sites, even when the metals were present at relatively high concentrations, whereas the uptake of metals within cells was much less affected by pH.  相似文献

Log yard run-off is a potential environmental risk, among other things because it creates an oxygen deficiency in receiving watercourses. This study was conducted to investigate the purification efficiency of soil-plant systems with couchgrass (Elymus repens) and willows (Salix sp.) when intensively irrigated with run-off from an open sprinkling system at a Norway spruce (Picea abies) log yard. The purification efficiency was determined both at the field scale (couchgrass) and in 68-L lysimeters (couchgrass and willows). Groundwater in the field and drainage water from the lysimeters were analysed for Total Organic Carbon (TOC), distillable phenols, total P, and total N. Retention of TOC, phenols and P occurred but no difference between couchgrass and willows was observed. The system had better purification capacity at the field scale than in the lysimeters.  相似文献

An acid leachable technique is employed in core samples (C1, C2 and C3) to develop a baseline data on the sediment quality for trace metals of River Uppanar, Cuddalore, southeast coast of India. Acid leachable metals (Fe, Mn, Cr, Cu, Ni, Co, Pb, Zn and Cd) indicate peak values at the sulphidic phase and enrichment of metals in the surface layers are due to the anthropogenic activities. Association of trace metals with Fe, Mn indicates their adsorption onto Fe-Mn oxyhydroxides and their correlation with S indicate that they are precipitated as metal sulphides. Factor analysis identified three possible types of geochemical associations and the supremacy of trace metals along with Fe, Mn, S and mud supports their geochemical associations. Factor analysis also signifies that anthropogenic activities have affected both the estuarine and fresh water regions of River Uppanar.  相似文献

Stream sediments from the mining and smelting district of Príbram, Czech Republic, were studied to determine the degree, sources and dispersal of metal contamination using a combination of bulk metal and mineralogical determinations, sequential extractions and Pb isotopic analyses. The highest metal concentrations were found 3-4 km downstream from the main polymetallic mining site (9800 mg Pb kg(-1), 26 039 mg Zn kg(-1), 316.4 mg Cd kg(-1), 256.9 mg Cu kg(-1)). The calculated enrichment factors (EFs) confirmed the extreme degree of contamination by Pb, Zn and Cd (EF>40). Lead, Zn and Cd are bound mainly to Fe oxides and hydroxides. In the most contaminated samples Pb is also present as Pb carbonates and litharge (PbO). Lead isotopic analysis indicates that the predominant source of stream sediment contamination is historic Pb-Ag mining and primary Pb smelting (206Pb/207Pb=1.16), while the role of secondary smelting (car battery processing) is negligible.  相似文献
