The published results on nanoparticles cytotoxicity and genotoxicity such as titanium dioxide nanoparticles (TiO(2) NPs) are inconsistent, and often conflicting and insufficient. Since different parameters may have impact on the toxicity results, there is need to lay stress on detailed characterization of NPs and the use of different testing conditions for assessment of NPs toxicity. In order to investigate whether dispersion procedures influence NP cytotoxicity and genotoxicity, we compared two protocols giving TiO(2) NP dispersions with different stability and agglomeration states. Detailed primary and secondary characteristics of both TiO(2) NP dispersions in culture media were carried out before toxicological testing; TK6 human lymphoblast cells, EUE human embryonic epithelial cells and Cos-1 monkey kidney fibroblasts were used to assess cytotoxicity (by trypan blue exclusion, proliferation activity and plating efficiency assays) and genotoxicity (by the comet assay). DNA strand breaks were detected by the alkaline comet assay. DNA oxidation lesions (especially 8-oxo-7,8-dihydroguanine, 8-oxoG) were measured with a modified comet assay including incubation with specific repair enzyme formamidopyrimidine DNA glycosylase (FPG). The TiO(2) NPs dispersion with large agglomerates (3 min sonication and no serum in stock solution) induced DNA damage in all three cell lines, while the TiO(2) NPs dispersed with agglomerates less than 200 nm (foetal serum in stock solution and sonication 15 min) had no effect on genotoxicity. An increased level of DNA oxidation lesions detected in Cos-1 and TK6 cells indicates that the leading mechanism by which TiO(2) NPs trigger genotoxicity is most likely oxidative stress. Our results show that the dispersion method used can influence the results of toxicity studies. Therefore at least two different dispersion procedures should be incorporated into assessment of cyto- and genotoxic effects of NPs. It is important, when assessing the hazard associated with NPs, to establish standard testing procedures and thorough strategies to consider the diverse conditions relevant to possible exposures. 相似文献

<Emphasis Type="Italic">Eubacterium rangiferina</Emphasis>, a novel usnic acid-resistant bacterium from the reindeer rumen 总被引:1,自引:0,他引:1
Reindeer are able to eat and utilize lichens as an important source of energy and nutrients. In the current study, the activities of antibiotic secondary metabolites including usnic, antranoric, fumarprotocetraric, and lobaric acid commonly found in lichens were tested against a collection of 26 anaerobic rumen bacterial isolates from reindeer (Rangifer tarandus tarandus) using the agar diffusion method. The isolates were identified based on their 16S ribosomal ribonucleic acid (rRNA) gene sequences. Usnic acid had a potent antimicrobial effect against 25 of the isolates, belonging to Clostridiales, Enterococci, and Streptococci. Isolates of Clostridia and Streptococci were also susceptible to atranoric and lobaric acid. However, one isolate (R3_91_1) was found to be resistant to usnic, antranoric, fumarprotocetraric, and lobaric acid. R3_91_1 was also seen invading and adhering to lichen particles when grown in a liquid anaerobic culture as demonstrated by transmission electron microscopy. This was a Gram-negative, nonmotile rod (0.2-0.7 x 2.0-3.5 microm) with a deoxyribonucleic acid G + C content of 47.0 mol% and main cellular fatty acids including 15:0 anteiso-dimethyl acetal (DMA), 16:0 iso-fatty acid methyl ester (FAME), 13:0 iso-3OH FAME, and 17:0 anteiso-FAME, not matching any of the presently known profiles in the MIDI database. Combined, the phenotypic and genotypic traits including the 16S rRNA gene sequence show that R3_91_1 is a novel species inside the order Clostridiales within the family Lachnospiraceae, for which we propose the name Eubacterium rangiferina. This is the first record of a rumen bacterium able to tolerate and grow in the presence of usnic acid, indicating that the rumen microorganisms in these animals have adapted mechanisms to deal with lichen secondary metabolites, well known for their antimicrobial and toxic effects. 相似文献

of vegetation fuels and methods of their mapping as well as maximum utilization of forest inventory information and Geographic
Information System (GIS). (2) Elaborate a technique of proper wildfire monitoring including estimation of vegetation damage.
(3) Improve daily rating of regional fire danger. (4) Improve fire-preventive arrangement of the territory covered by vegetation,
the main goal being creation of favourable conditions for active fire management. (5) Choose the main direction in elaboration
of fire-fighting means and methods taking into account their universality, simplicity, reliability, etc. (6) Elaborate an
improved technique for estimation of economic effectiveness of the wildfire control system. (7) Develop international cooperation

Technical product harvesting (TEPHA) is a newly developing interdisciplinary approach in which bio-based production is investigated from a technical and ecological perspective. Society‘s demand for ecologically produced and sustainably operable goods is a key driver for the substitution of conventional materials like metals or plastics through bio-based alternatives. Technical product harvesting of near net shape grown components describes the use of suitable biomass for the production of technical products through influencing the natural shape of plants during their growth period. The use of natural materials may show positive effects on the amount of non-renewable resource consumption. This also increases the product recyclability at the end of its life cycle. Furthermore, through the near net shape growth of biomass, production steps can be reduced. As a consequence such approaches may save energy and the needed resources like crude oil, coal or gas. The derived near net shape grown components are not only considered beneficial from an environmental point of view. They can also have mechanical advantages through an intrinsic topology optimization in contrast to common natural materials, which are influenced in their shape after harvesting. In order to prove these benefits a comprehensive, interdisciplinary scientific strategy is needed. A three-dimensional chemical transport model (PMCAMx) was used to investigate changes in fine particle (PM2.5) concentrations in response to changes in sulfur dioxide (SO2) and ammonia (NH3) emissions during July 2001 and January 2002 in the eastern United States. A uniform 50% reduction in SO2 emissions was predicted to produce an average decrease of PM2.5 concentrations by 26% during July but only 6% during January. A 50% reduction of NH3 emissions leads to an average 4 and 9% decrease in PM2.5 in July and January, respectively. During the summer, the highest concentration of sulfate is in South Indiana (12.8 microg x m(-3)), and the 50% reduction of SO2 emissions results in a 5.7 microg x m(-3) (44%) sulfate decrease over this area. During winter, the SO2 emissions reduction results in a 1.5 microg x m(-3) (29%) decrease of the peak sulfate levels (5.2 microg x m(-3)) over Southeast Georgia. The maximum nitrate and ammonium concentrations are predicted to be over the Midwest (1.9 (-3)g x m(-3) in Ohio and 5.3 microg x m(-3) in South Indiana, respectively) in the summer whereas in the winter these concentrations are higher over the Northeast (3 microg x m(-3) of nitrate in Connecticut and 2.7 microg x m(-3) of ammonium in New York). The 50% NH3 emissions reduction is more effective for controlling nitrate, compared with SO2 reductions, producing a 1.1 microg x m(-3) nitrate decrease over Ohio in July and a 1.2 microg x m(-3) decrease over Connecticut in January. Ammonium decreases significantly when either SO2 or NH3 emissions are decreased. However, the SO2 control strategy has better results in July when ammonium decreases, up to 2 microg x m(-3) (37%), are predicted in South Indiana. The NH3 control strategy has better results in January (ammonium decreases up to 0.4 microg x m(-3) in New York). The spatial and temporal characteristics of the effectiveness of these emission control strategies during the summer and winter seasons are discussed. 相似文献

Sorption, desorption, and degradation of (4-chloro-2-methylphenoxy)acetic acid in representative soils of the Danubian Lowland, Slovakia 总被引:1,自引:0,他引:1
Herbicide leaching through soil into groundwater greatly depends upon sorption-desorption and degradation phenomena. Batch adsorption, desorption and degradation experiments were performed with acidic herbicide MCPA and three soil types collected from their respective soil horizons. MCPA was found to be weakly sorbed by the soils with Freundlich coefficient values ranging from 0.37 to 1.03 mg1−1/n kg−1 L1/n. It was shown that MCPA sorption positively correlated with soil organic carbon content, humic and fulvic acid carbon contents, and negatively with soil pH. The importance of soil organic matter in MCPA sorption by soils was also confirmed by performing sorption experiments after soil organic matter removal. MCPA sorption in these treated soils decreased by 37-100% compared to the original soils. A relatively large part of the sorbed MCPA was released from soils into aqueous solution after four successive desorption steps, although some hysteresis occurred during desorption of MCPA from all soils. Both sorption and desorption were depth-dependent, the A soil horizons exhibited higher retention capacity of the herbicide than B or C soil horizons. Generally, MCPA sorption decreased in the presence of phosphate and low molecular weight organic acids. Degradation of MCPA was faster in the A soil horizons than the corresponding B or C soil horizons with half-life values ranging from 4.9 to 9.6 d in topsoils and from 11.6 to 23.4 d in subsoils. 相似文献

The reactivity of the herbicide cycloxydim (CD) toward singlet oxygen was studied in organic solution and on wax films to mimic the leaf surface. Experiments in solution were conducted in acetonitrile using phenalenone as a sensitizer. For the experiments in the solid state, phenalenone was included in paraffinic wax films and CD deposited at the film surface. By laser flash photolysis we observed that the triplet of phenalenone reacts with CD with a bimolecular rate constant of (9.6 ± 1)×106 M−1 s−1. However, scavenging experiments using β-carotene as a singlet oxygen quencher showed that the observed CD degradation in aerated acetonitrile involves singlet oxygen essentially. The bimolecular rate constant of reaction of CD with singlet oxygen was evaluated to (1.0 ± 0.2)×107 M−1 s−1. Phenalenone included in the wax films also significantly increased the rate of CD photodegradation, the involvement of singlet oxygen being very likely. The photoproducts formed via the singlet oxygen pathway resulted from the oxidation of the thiol group and/or the cyclohexene ring. This study should help to better understand the fate of CD after its spraying on crops. 相似文献

Citizen science has the potential to expand the scope and scale of research in ecology and conservation, but many professional researchers remain skeptical of data produced by nonexperts. We devised an approach for producing accurate, reliable data from untrained, nonexpert volunteers. On the citizen science website www.snapshotserengeti.org , more than 28,000 volunteers classified 1.51 million images taken in a large‐scale camera‐trap survey in Serengeti National Park, Tanzania. Each image was circulated to, on average, 27 volunteers, and their classifications were aggregated using a simple plurality algorithm. We validated the aggregated answers against a data set of 3829 images verified by experts and calculated 3 certainty metrics—level of agreement among classifications (evenness), fraction of classifications supporting the aggregated answer (fraction support), and fraction of classifiers who reported “nothing here” for an image that was ultimately classified as containing an animal (fraction blank)—to measure confidence that an aggregated answer was correct. Overall, aggregated volunteer answers agreed with the expert‐verified data on 98% of images, but accuracy differed by species commonness such that rare species had higher rates of false positives and false negatives. Easily calculated analysis of variance and post‐hoc Tukey tests indicated that the certainty metrics were significant indicators of whether each image was correctly classified or classifiable. Thus, the certainty metrics can be used to identify images for expert review. Bootstrapping analyses further indicated that 90% of images were correctly classified with just 5 volunteers per image. Species classifications based on the plurality vote of multiple citizen scientists can provide a reliable foundation for large‐scale monitoring of African wildlife. 相似文献

A water quality model (MERES) is applied in the multi-purpose reservoir of Plastiras in Greece. The model was calibrated with
limited field data using the standard trial and error procedure; next, it was applied for four scenarios of reservoir operation,
which correspond to four values of minimum water levels (MWL). Model results were combined with landscape aesthetics to establish
a “safe” MWL in the reservoir. MWL was proposed to be +784 m, a value that ensures a very good water quality by retaining
the reservoir in the status of “a little beyond oligotrophic” and a very good quality of landscape with an acceptable area
of dead zones; it also ensures an annual release of 127.5 hm3 of water within a reliability level of 90%. This quantity can be considered as acceptable from the water supply point of
