Sulfate reducing bacteria (SRB) are important mercury methylators in sediments, but information on mercury methylators in other compartments is ambiguous. To investigate SRB involvement in methylation in Amazonian periphyton, the relationship between Hg methylation potential and SRB (Desulfobacteraceae, Desulfobulbaceae and Desulfovibrionaceae) abundance in Eichhornia crassipes and Polygonum densiflorum root associated periphyton was examined. Periphyton subsamples of each macrophyte were amended with electron donors (lactate, acetate and propionate) or inhibitors (molybdate) of sulfate reduction to create differences in SRB subgroup abundance, which was measured by quantitative real-time PCR with primers specific for the 16S rRNA gene. Mercury methylation and demethylation potentials were determined by a stable isotope tracer technique using 200HgCl and CH3202HgCl, respectively. Relative abundance of Desulfobacteraceae (<0.01-12.5%) and Desulfovibrionaceae (0.01-6.8%) were both highly variable among samples and subsamples, but a significant linear relationship (p < 0.05) was found between Desulfobacteraceae abundance and net methylmercury formation among treatments of the same macrophyte periphyton and among all P. densiflorum samples, suggesting that Desulfobacteraceae bacteria are the most important mercury methylators among SRB families. Yet, molybdate only partially inhibited mercury methylation potentials, suggesting the involvement of other microorganisms as well. The response of net methylmercury production to the different electron donors and molybdate was highly variable (3-1104 pg g−1 in 12 h) among samples, as was the net formation in control samples (17-164 pg g−1 in 12 h). This demonstrates the importance of community variability and complexity of microbial interactions for the overall methylmercury production in periphyton and their response to external stimulus.  相似文献

Concentrations and sources of Dechlorane Plus in sewage sludge   总被引:1,自引:0,他引:1  
Sewage sludge from 31 urban Spanish wastewater treatment plants (WWTP) was analyzed for the emerging halogenated flame retardant Dechlorane Plus (DP). Concentrations of the two major isomers in the technical mixture, syn and anti, ranged between 0.903-19.2 and 1.55-75.1 ng g−1 dry weight, respectively. Overall, concentrations of DP were lower than those of polybrominated diphenyl ethers (PBDEs) (9.10-995 ng g−1 dry weight) and this is likely related to the higher usage of brominated flame retardants. The average ratio of the syn isomer to total DP (fsyn) was 0.28 ± 0.05, which is similar to that of the commercial mixture. Comparing different wastewater treatment methods, we found lower concentrations in those using biological nitrogen and phosphorous elimination, suggesting that DP is susceptible to microbial degradation and that anti-DP is more so, given the enrichment of syn-DP in the sewage sludge. Principal components analysis revealed significant positive correlation (r = 0.619, p < 0.05) between total DP concentrations with the contribution of industrial input to waste streams. This implies release of DP is related to industrial activity, likely stemming from the use of the technical product during manufacture of consumer goods. However, use and disposal of products containing DP could not be dismissed. According to our knowledge, this is the first report on DP in WWTP sludge.  相似文献

This work demonstrates the success of a recently developed technique in chemical amplification, non-biological inhibition-based sensing (NIBS), for the detection of toxic arsenic compounds. Screening for toxic arsenic compounds is especially important due to their prevalence in wastewater and water sources. The detection method presented in this work amplifies the chemical response of toxic arsenic compounds by developing a sensor chemistry where the analyte inhibits, rather than enhances, the rate of a catalytic reaction. This technique mimics the work done with enzyme inhibition; however, using non-biological molecules allows for selective detection without the shelf-life issue associated with biological molecules. Using NIBS we find that we can enhance the sensitivity of the system by two orders of magnitude with no apparent loss in selectivity. This work demonstrates the versatility of NIBS, showing that the technique can be of general use for the detection of toxic compounds.  相似文献

This paper reports genotoxicity and toxicity data in water samples collected in Sinos River, an important water course in the hydrographic region of Guaíba Lake, Rio Grande do Sul State, south of Brazil. This river is exposed to intense anthropic influence by numerous shoes, leather, petrochemical, and metallurgy industries. Water samples were collected at two moments (winter 2006 and spring 2006) at five sites of Sinos River and evaluated using in vitro V79 Chinese hamster lung fibroblasts (cytotoxicity, comet assay and micronucleus test) and Allium cepa test (toxicity and micronucleus test). Comet and micronucleus tests revealed that water samples collected exerted cytotoxic, toxic, genotoxic and mutagenic effects. The results showed the toxic action of organic and inorganic agents found in the water samples in all sites of Sinos River, for both data collections. The main causes behind pollution were the domestic and industrial toxic discharges. The V79 and A. cepa tests were proved efficient to detect toxicity and genotoxicity caused by complex mixtures. This study also showed the need for constant monitoring in sites with strong environmental degradation caused by industrial discharges and urban sewages.  相似文献

Differentiation of nitrous oxide emission factors for agricultural soils   总被引:3,自引:0,他引:3  
Nitrous oxide (N2O) direct soil emissions from agriculture are often estimated using the default IPCC emission factor (EF) of 1%. However, a large variation in EFs exists due to differences in environment, crops and management. We developed an approach to determine N2O EFs that depend on N-input sources and environmental factors. The starting point of the method was a monitoring study in which an EF of 1% was found. The conditions of this experiment were set as the reference from which the effects of 16 sources of N input, three soil types, two land-use types and annual precipitation on the N2O EF were estimated. The derived EF inference scheme performed on average better than the default IPCC EF. The use of differentiated EFs, including different regional conditions, allows accounting for the effects of more mitigation measures and offers European countries a possibility to use a Tier 2 approach.  相似文献

Several major breeding areas have been defined for the South American sea lion (Otaria flavescens) along the Atlantic Ocean including the Uruguayan and Patagonian coasts. Together with a documented and severe reduction in population sizes caused by commercial hunting in the last century, these areas show opposite population trends. While Patagonian populations are recovering since hunting ceased, Uruguayan populations are declining. In this context, population genetic structure and genetic diversity were studied for the first time with both nuclear (microsatellites) and mitochondrial (control region) markers together. Alternative scenarios were found for both markers. While mitochondrial marker showed geographically structured populations, the nuclear loci showed a lack of geographical structure. These opposite patterns in genetic structure could be explained by female phylopatry and high male dispersion. The reduction in population size caused by commercial hunting did not leave a detectable footprint of bottleneck at the genetic level.  相似文献

Intraguild predation (IGP) has been explained in terms of competitor-removal, food-stress and predator-removal hypotheses. Only the first two hypotheses have been fairly well studied. To test the predator-removal hypothesis as a force determining IGP in avian predators, we performed a field experiment to simulate the presence of an IG predator (eagle owl Bubo bubo dummy) in the surrounding of the nests of four potential IG prey (black kite Milvus migrans, red kite Milvus milvus, booted eagle Aquila pennata and common buzzard Buteo buteo). To discard the possibility that an aggressive reaction towards the eagle owl was not related to the presence of the IG predator, we also presented a stuffed tawny owl Strix aluco, which is a potential competitor but cannot be considered an IG predator of the studied diurnal raptors considered in the experiment. While almost always ignoring the tawny owl, raptors chiefly showed an interspecific aggressive behaviour towards their IG predator. Our results seem to support the predator-removal hypothesis, as the IG prey may take advantage of the diurnal inactivity of the IG predator to remove it from their territory. However, the recorded behaviour may be also considered as a special variety of mobbing (i.e. a prey’s counter-strategy against its predator), where the mobber is sufficiently powerful to escalate predator harassment into deliberate killing attempts. In their turn, eagle owls can respond with an IG predatory behaviour aimed at removing IG prey species which are highly aggressive mobbers.  相似文献

The present study investigated the influence of 241Am on microbial growth and the degradation of organic waste. Leachate samples collected in a lysimeter were periodically analyzed for bacterial growth, under both aerobic and anaerobic conditions. 241Am inhibited bacterial growth, and the degradation of organic matter was delayed in comparison with the control. Minimal inhibitory concentration assays and survival curves revealed that it inhibits the growth of Pseudomonas putida F1. The assay also revealed that 241Am is more toxic than 238U, Zn2+ and Cd2+. This study further led to the finding of four new radionuclide-tolerant bacterial strains: Flavobacterium spp., Pseudomonas gladioli, Chryseobacterium indologenes and Ochrobactrum anthropi. The survival curves of P. gladioli, C. indologenes revealed that these bacteria are resistant to metal as consortia.  相似文献

An major research area in environmental chemistry is the development of methods for the analysis of biomarkers. Metallothioneins are used as biomarkers in studies of heavy metals exposure in water, because metallothioneins are synthesized and accumulated when organisms are exposed to toxic concentrations of pollutants. In this work, simple and sensitive voltammetric methods were developed for metallothionein and copper (II) determinations in fish liver Lepomis gibbosus. Both analytical methodologies were optimized and applied to samples extracted from individuals previously submitted to sub-lethal toxicological trials with copper sulphate (CuSO4) and cadmium chloride (CdCl2). The obtained results showed that both methods are very precise, sensitive, and involve simple sample preparation processes. Moreover, metallothioneins showed better correlation with the toxic exposure than Cu2+. To the best of our knowledge, this is the first time that hepatic metallothioneins and Cu2+ contents are voltammetrically determined in order to be compared in their function as heavy metal biological indicators.  相似文献

Polder lakes in Flanders are stagnant waters that were flooded by the sea in the past. Several of these systems are colonized by exotic species, but have hardly been studied until present. The aim of the present study was: (1) to assess the influence of exotic macrobenthic species on the outcome of the Multimetric Macroinvertebrate Index Flanders (MMIF) and (2) to use classification trees for evaluating to what extent physical-chemical characteristics affect the presence of exotic species.In total, 27 mollusc and 10 macro-crustacean species were present in the monitored lakes of which respectively five and four were exotic. The exclusion of the exotic species from the MMIF resulted in a significant decline of this ecological index (−0.03 ± 0.04; p = 0.00). This elimination often resulted into a lower ecological water quality class and more samples were classified into the bad and poor ecological water quality classes.Single-target classification trees for Gammarus tigrinus and Potamopyrgus antipodarum were constructed, relating environmental parameters and ecological status (MMIF) to the occurrence of both exotic invasive species. The major advantages of using single-target classification trees are the transparency of the rule sets and the possibility to use relatively small datasets. However, this classification technique only predicts a single-target attribute and the trees of the different species are often hard to integrate and use for water managers. As a solution, a multi-target approach was used in the present study. Exotic molluscs and crustaceans communities were modelled based on environmental parameters and the ecological status (MMIF) using multi-target classification trees. Multi-target classification trees can be used in management planning and investment decisions as they can lead to integrated decisions for the whole set of exotic species and avoid the construction of many models for each individual species. These trees provide general insights concerning the occurrence patterns of individual crustaceans and molluscs in an integrated way.  相似文献
