Molecular methods are useful both to monitor natural communities of bacteria, and to track specific bacterial markers in complex environments. Length-heterogeneity polymerase chain reaction (LH-PCR) and terminal restriction fragment length polymorphism (T-RFLP) of 16S rDNAs discriminate among 16S rRNA genes based on length polymorphisms of their PCR products. With these methods, we developed an alternative indicator that distinguishes the source of fecal pollution in water. We amplify 16S rRNA gene fragments from the fecal anaerobic genus Bacteroides with specific primers. Because Bacteroides normally resides in gut habitats, its presence in water indicates fecal pollution. Molecular detection circumvents the complexities of growing anaerobic bacteria. We identified Bacteroides LH-PCR and T-RFLP ribosomal DNA markers unique to either ruminant or human feces. The same unique fecal markers were recovered from polluted natural waters. We cloned and sequenced the unique markers; marker sequences were used to design specific PCR primers that reliably distinguish human from ruminant sources of fecal contamination. Primers for more species are under development. This approach is more sensitive than fecal coliform assays, is comparable in complexity to standard food safety and public health diagnostic tests, and lends itself to automation and high-throughput. The construction of artificial wetlands has become a measure increasingly applied to reduce nonpoint-source (NPS) pollution and to contribute to the restoration of eutrophic lakes and coastal waters. In a 2-yr study monitoring fluxes of particulate and dissolved phosphorus (P) in a small artificial wetland for the treatment of agricultural drainage water in Central Switzerland, water residence time was identified as the main factor controlling P retention in the system. Since most of the annual P load (62% as dissolved reactive phosphorus, DRP) was related to high discharge events, it was not average but minimum water residence time during flood events that determined the wetland's P retention. In agreement with a continuous stirred tank reactor (CSTR) model, our investigations suggest a minimum water residence time of 7 d to retain at least 50% of the bioavailable P. The investigated wetland retained only 2% of the bioavailable P, since the water residence time was shorter than 7 d during 61% of time in both years. Settling of phytoplankton rather than DRP uptake into phytoplankton limited the retention of bioavailable P. The overall retention efficiency of 23% total phosphorus (TP), corresponding to a surface related retention of 1.1 g P m(-2) yr(-1), was due to the efficient trapping of pedogenic particles. 相似文献
This paper assesses the vulnerability of schools to floods in the Nyando River catchment (3,600 km2) in western Kenya and identifies measures needed to reduce this vulnerability. It surveys 130 schools in the lower reaches, where flooding is a recurrent phenomenon. Of the primary schools assessed, 40% were vulnerable, 48% were marginally vulnerable and 12% were not vulnerable. Of the secondary schools, 8% were vulnerable, 73% were marginally vulnerable and 19% were not vulnerable. Vulnerability to floods is due to a lack of funds, poor building standards, local topography, soil types and inadequate drainage. The Constituencies Development Fund (CDF), established in 2003, provides financial support to cover school construction and reconstruction costs; CDF Committees are expected to adopt school building standards. In an effort to promote safe and resilient construction and retrofitting to withstand floods, this paper presents vulnerability reduction strategies and recommendations for incorporating minimum standards in the on‐going Primary School Infrastructure Programme Design. 相似文献
This study involves the monitoring of organic pollutants using transplanted mussels (Mytilus galloprovincialis) as bioindicator organisms and semipermeable membrane devices (SPMDs) as passive samplers. Mussels and SPMDs were deployed to marinas, shipyards and shipbreaking yards on the coastal area of Turkey and retrieved after 60 days. Polycyclic aromatic hydrocarbons (PAH), polychlorinated biphenyls (PCB) and organochlorine pesticide (OCP) compounds were analysed with high-resolution GC-MS. Total PAH concentrations in SPMDs and mussels ranged from 200 to 4740 ng g sampler?1 and from 7.0 to 1130 ng g?1 in wet weight (ww). PCB and OCP concentrations in SPMDs changed between 0.04–200 and 4.0–26 ng g sampler?1, respectively. The highest PCB (190 ng g?1 ww) and OCP (200 ng g?1 ww) concentrations in mussels were measured at shipyard stations. A strong correlation was observed between the PAH and PCB concentrations in SPMDs and mussels. Enzyme assays (acetylcholinesterase, ethoxyresorufin-O-deethylase, glutathione S-transferase, glutathion reductase and carboxylesterase activities) were performed as biomarkers to reveal the effects of pollution on the mussels. There was no clear relationship found between the enzyme levels and the pollutant concentrations in mussels. Integrated biomarker responses were calculated to interpret the overall effect of pollutants.
Conservation and protection of soil and water resources and visual aspects of landscape, as well as the promotion of biodiversity, are some of the central tasks of environmental policy development and social politics in the future. One of the main questions is: ‘which agricultural systems are able to guarantee sustained resource-conserving land use?’ Based on the ecological risk concepts of the 1970s and 1980s, a potential impact model was developed using a universal assessment algorithm derived from fuzzy logic. The model estimated the potential impact of agricultural land use on ecosystem function using a few resource indicators. Intervention intensities of agricultural land-use are set in relation to site conditions and aggregated for each of several defined potential impact categories. The interpretation with respect to risk and the calculation of potential impact values are explained. 相似文献
