The present study is aimed at analysing and comparing different soil enzymes in soil samples of native contaminated sites
of a Mathura refinery and adjoining agricultural land. Enzyme activities are considered as indicators of soil quality and
changes in biogeochemical function due to management or perturbations. Soil samples were collected from the premises and nearby
area of Mathura refinery, India. Biological health parameters (dehydrogenase, aryl esterase, aryl sulphatase,
\upbeta \upbeta -glucosidase, alkaline phosphatase, acid phosphatase, lipase, laccase and catalase activity) were estimated in the soil samples.
Among all the samples, sewage sludge soil showed maximum activity of enzymes, microbial biomass carbon and most probable number
of polycyclic aromatic hydrocarbon (PAH) degraders in soils spiked with three- to four-ring PAHs at 50 ppm. Available phosphorus,
potassium and nitrogen was also exceptionally high in this sample, indicating maximum microbial bioconversion due to presence
of nutrients stimulating potent PAH-degrading microorganisms. 相似文献
Sediment quality guidelines (SQGs) can be derived using different approaches and are commonly used in environmental management,
reclamation, and risk assessment. The screening-level concentration (SLC) approach has been used in Ontario, Canada, to derive
lowest effect levels (LELs) and severe effect levels for use as SQGs. This approach was adopted by the Canadian Nuclear Safety
Commission (CNSC) to set guidelines for metals (As, Cr, Cu, Pb, Mo, Ni, Se, U, and V) and radionuclides (Ra-226, Pb-210, and
Po-210) in sediment at northern Saskatchewan uranium mining and milling operations. The SLC approach is based on total metal
and radionuclide concentrations in sediment, and corresponding benthic community composition data for a specific sampling
site. In this study, sediment chemistry (total metals and radionuclides) and benthic community data from northern Saskatchewan
uranium operations were compiled and examined. Results indicate that the CNSC-derived SQGs had limited relationships to observed
effects, or lack thereof, on benthic invertebrate communities near uranium operations in Saskatchewan. The LELs were found
to correctly align with effects at 95% of the sites that had effects, on a general basis, but on an element-specific basis
many of the elements had concentrations at effect sites below their LELs. Furthermore, concentrations of the evaluated elements
exceeded at least one LEL at 60% of the no-effect sites. The high number of exceedences of LELs at reference and no-effect
sites (false-positives) calls to question the appropriateness of the CNSC-derived SQGs. It is suggested that alternatives

We used variance components to assess allocation of sampling effort in a hierarchically nested sampling design for ongoing monitoring of early life history stages of the federally endangered Devils Hole pupfish (DHP) (Cyprinodon diabolis). Sampling design for larval DHP included surveys (5 days each spring 2007–2009), events, and plots. Each survey was comprised of three counting events, where DHP larvae on nine plots were counted plot by plot. Statistical analysis of larval abundance included three components: (1) evaluation of power from various sample size combinations, (2) comparison of power in fixed and random plot designs, and (3) assessment of yearly differences in the power of the survey. Results indicated that increasing the sample size at the lowest level of sampling represented the most realistic option to increase the survey’s power, fixed plot designs had greater power than random plot designs, and the power of the larval survey varied by year. This study provides an example of how monitoring efforts may benefit from coupling variance components estimation with power analysis to assess sampling design. 相似文献
One of the highest self-reported incidence rates of acute gastrointestinal illness (AGI) in the global peer-reviewed literature occurs in Inuit communities in the Canadian Arctic. This high incidence of illness could be due, in part, to the consumption of contaminated water, as many northern communities face challenges related to the quality of municipal drinking water. Furthermore, many Inuit store drinking water in containers in the home, which could increase the risk of contamination between source and point-of-use (i.e., water recontamination during storage). To examine this risk, this research characterized drinking water collection and storage practices, identified potential risk factors for water contamination between source and point-of-use, and examined possible associations between drinking water contamination and self-reported AGI in the Inuit community of Rigolet, Canada. The study included a cross-sectional census survey that captured data on types of drinking water used, household practices related to drinking water (e.g., how it was collected and stored), physical characteristics of water storage containers, and self-reported AGI. Additionally, water samples were collected from all identified drinking water containers in homes and analyzed for presence of Escherichia coli and total coliforms. Despite municipally treated tap water being available in all homes, 77.6% of households had alternative sources of drinking water stored in containers, and of these containers, 25.2% tested positive for total coliforms. The use of transfer devices and water dippers (i.e., smaller bowls or measuring cups) for the collection and retrieval of water from containers were both significantly associated with increased odds of total coliform presence in stored water (ORtransfer device = 3.4, 95% CI 1.2–11.7; ORdipper = 13.4, 95% CI 3.8–47.1). Twenty-eight-day period prevalence of self-reported AGI during the month before the survey was 17.2% (95% CI 13.0–22.5), which yielded an annual incidence rate of 2.4 cases per person per year (95% CI 1.8–3.1); no water-related risk factors were significantly associated with AGI. Considering the high prevalence of, and risk factors associated with, indicator bacteria in drinking water stored in containers, potential exposure to waterborne pathogens may be minimized through interventions at the household level. 相似文献
Animal studies show that exposure to the environmental pollutant 3,3′,4,4′,5-pentachlorobiphenyl (PCB126) causes alterations in hepatic metals as measured in acid-digested volume-adjusted tissue. These studies lack the detail of the spatial distribution within the liver. Here we use X-ray fluorescence microscopy (XFM) to assess the spatial distribution of trace elements within liver tissue. Liver samples from male Sprague Dawley rats, treated either with vehicle or PCB126, were formalin fixed and paraffin embedded. Serial sections were prepared for traditional H&E staining or placed on silicon nitride windows for XFM. With XFM, metal gradients between the portal triad and the central vein were seen, especially with copper and iron. These gradients change with exposure to PCB126, even reverse. This is the first report of how micronutrients vary spatially within the liver and how they change in response to toxicant exposure. In addition, high concentrations of zinc clusters were discovered in the extracellular space. PCB126 treatment did not affect their presence, but did alter their elemental makeup suggesting a more general biological function. Further work is needed to properly evaluate the gradients and their alterations as well as classify the zinc clusters to determine their role in liver function and zinc homeostasis. 相似文献
Soybean meal (SBM) is a product generated from the manufacture of soybean oil and has the potential for use as a source of fermentable sugars for ethanol production or as a protein source for animal feeds. Knowing the levels of nitrogen available from ammonium is a necessary element of the ethanolic fermentation process while identifying the levels of essential amino acids such as lysine is important in determining usage as a feed source. As such the purpose of this study was to quantify total nitrogen and ammonium in the liquid fraction of hydrolyzed SBM and to evaluate total and bioavailable lysine in the solid fraction of the hydrolyzed SBM. The effects of acid concentration, cellulase and β-glucosidase on total and ammonium nitrogen were studied with analysis indicating that higher acid concentrations increased nitrogen compounds with ammonium concentrations ranging from 0.20 to 1.24 g L?1 while enzymatic treatments did not significantly increase nitrogen levels. Total and bioavailable lysine was quantified by use of an auxotrophic gfpmut3 E.coli whole-cell bioassay organism incapable of lysine biosynthesis. Acid and enzymatic treatments were applied with lysine bioavailability increasing from a base of 82% for untreated SBM to up to 97%. Our results demonstrated that SBM has the potential to serve in ethanolic fermentation and as an optimal source essential amino acid lysine. 相似文献
Remote sensing devices have been used for decades to measure gaseous emissions from individual vehicles at the roadside. Systems have also been developed that entrain diluted exhaust and can also measure particulate matter (PM) emissions. In 2015, the California Air Resources Board (CARB) reported that 8% of in-field diesel particulate filters (DPF) on heavy-duty (HD) vehicles were malfunctioning and emitted about 70% of total diesel PM emissions from the DPF-equipped fleet. A new high-emitter problem in the heavy-duty vehicle fleet had emerged. Roadside exhaust plume measurements reflect a snapshot of real-world operation, typically lasting several seconds. In order to relate roadside plume measurements to laboratory emission tests, we analyzed carbon dioxide (CO2), oxides of nitrogen (NOX), and PM emissions collected from four HD vehicles during several driving cycles on a chassis dynamometer. We examined the fuel-based emission factors corresponding to possible exceedances of emission standards as a function of vehicle power. Our analysis suggests that a typical HD vehicle will exceed the model year (MY) 2010 emission standards (of 0.2 g NOX/bhp-hr and 0.01 g PM/bhp-hr) by three times when fuel-based emission factors are 9.3 g NOX/kg fuel and 0.11 g PM/kg using the roadside plume measurement approach. Reported limits correspond to 99% confidence levels, which were calculated using the detection uncertainty of emissions analyzers, accuracy of vehicle power calculations, and actual emissions variability of fixed operational parameters. The PM threshold was determined for acceleration events between 0.47 and 1.4 mph/sec only, and the NOX threshold was derived from measurements where after-treatment temperature was above 200°C. Anticipating a growing interest in real-world driving emissions, widespread implementation of roadside exhaust plume measurements as a compliment to in-use vehicle programs may benefit from expanding this analysis to a larger sample of in-use HD vehicles.
Implications: Regulatory agencies, civil society, and the public at large have a growing interest in vehicle emission compliance in the real world. Leveraging roadside plume measurements to identify vehicles with malfunctioning emission control systems is emerging as a viable new and useful method to assess in-use performance. This work proposes fuel-based emission factor thresholds for PM and NOx that signify exceedances of emission standards on a work-specific basis by analyzing real-time emissions in the laboratory. These thresholds could be used to prescreen vehicles before roadside enforcement inspection or other inquiry, enhance and further develop emission inventories, and potentially develop new requirements for heavy-duty inspection and maintenance (I/M) programs, including but not limited to identifying vehicles for further testing. 相似文献
Because of the widespread use of silver nanoparticles in commercial products, discharges of municipal wastewater may be a point source of silver in the aquatic environment. We monitored two sites in western Lake Ontario impacted by discharges from wastewater treatment plants serving the City of Toronto. Concentrations of silver were elevated in bottom sediments and suspended sediments collected at the two sites. We also deployed two types of passive samplers in the water column at the two sites, the newly developed Carbon Nanotube Integrative Samplers for monitoring “CNIS-labile” silver and Diffusive Gradient in Thin Film samplers for monitoring “DGT-labile” silver. Results from these passive samplers indicated that the concentrations of silver at the two sites were either below detection limits or were in the ng/L range. In laboratory experiments where the sediments were re-suspended in Milli-Q water, a small proportion of the silver (i.e., <?25%) was labile and partitioned as colloidal or dissolved silver into the liquid phase after agitation. Nanoparticles tentatively identified as silver nanoparticles were detected by single-particle ICP-MS in suspension after agitation of both suspended and bottom sediments. Therefore, there is a need to assess whether silver species, including silver nanoparticles are transported from wastewater treatment plants into sediments in the aquatic environment. The study of static and dynamic behavior of environmentally conditioned fibre reinforced polymeric (FRP) composites is necessary and crucial to examine the durability, reliability and sustainability of these noble materials. FRP composites are being used all around the globe and substituting the conventional materials, starting from mini toys to large aerospace components. Present review has introduced to accumulate and understand the disseminate literature in concentrating the significance of understanding the static and dynamic behavior of FRPs with changing environmental conditionings (hygrothermal, low and high temperature, salt solution, freeze thaw, UV light) and with the interaction of different nano-fillers. Their stability and integrity in diverse service environments may be reformed by their reactions against different nature of loadings i.e. static or dynamic and the components such as fibre, matrix and fibre/matrix interfaces in those environments. The static and dynamic states of loading may come with a possible weaker region to encounter the durability and integrity of the composites. To understand the exact failure modes that correlates the position of environmentally conditioned interfaces and dynamic state of loadings, thus confusing the estimation of its overall performance and mechanical behavior. Interface reliability and durability is vital since in-service environments the degradation in the interfacial region leads to complete composite failure. Therefore, the study of combined effects of various in-service environmental conditions and the role of static and dynamic behavior on the interface will be a crucial part related to the multiaxial dynamic states of failures occurring in FRP’s. 相似文献
