A total of 310 individuals (124 males, 185 females, 1 indeterminate) of the octopod Octopus salutii caught throughout the year by bottom trawlers from 25 to 800 m depth in the western Mediterranean were analysed to study some general aspects of its biology. Octopus sizes ranged from 4.0 to 13.0 cm ML and 3.5 to 16.5 cm ML in males and females, respectively. The species inhabits the lower continental shelf and upper slope, primarily between 250 and 500 m depth. The scarcity of small-sized individuals in the samples and the predominance of males and females >8 and >9 cm ML, respectively, suggest two interpretations: (1) juveniles inhabit grounds inaccessible to, or unsampled by, trawlers, or they avoid capture owing to behavioural aspects; or (2) the species meet in the trawling grounds for reproduction, because in the sizes described the majority of individuals have begun sexual maturation. The reproductive period was found to occur during spring and summer. The stomach content analysis revealed a diet composed of 33 different prey items belonging to three major taxonomic groups (crustaceans, fishes and cephalopods). Quantitatively, crustaceans (Decapoda Reptantia and Natantia groups) were the most important prey, appearing in 87% of the stomachs, followed by fish (25%) and cephalopods (10%). Finally, the paper presents data on mantle length–total weight, gonad weight–total weight and ligulae length–total weight relationships, and utilises different indexes (gonadosomatic, hepatosomatic, fullness weight and emptiness) to describe changes through the three maturity stages of both sexes.Communicated by S.A. Wet weight to ash-free dry weight conversion factors were calculated for the bivalve Mytilus galloprovincialis Lamarck; specimens were collected in November 1989 in the Magra Estuary (eastern Ligurian Sea, Italy). Ash-free dry weight was determined by four different methods: (A) mechanical separation of shell and body; (B) shell dissolution with HCl; (C) dissolution of organic matter with NaClO; (D) incineration of the whole organism. The results obtained by these four methods were significantly different, indicating that caution is necessary when comparing biomass data obtained by different methods. Incineration and decalcification with HCl seem to be the best procedures: they not only combine accuracy with speed, but are also the most universally applicable, being equally suitable for soft-bodied or hard-bodied organisms. Mechanical separation can lead to an underestimation of the biomass, whereas treatment with NaClO can lead to its overestimation. However, no method can be considered the best in absolute terms, the final choice depending on the particular weight measurement required (e.g. shell-free dry weight) and the nature of the organism concerned (e.g. an organism with a large carbonate content).  相似文献

Samples of Halobates robustus Barber (Heteroptera: Gerridae) from the Galápagos Islands were analysed by optical emission spectrometry. The levels (in g g-1 dry weight) of Zn (134), Cu (155), Pb (< 1), Cd (7), and Cr (3) were not significantly different among insects of different sexes or developmental stages. The low natural levels of Cd in H. robustus from the relatively unpolluted environment of the Galápagos Islands are compared to the high concentrations of Cd in Halobates spp. from relatively polluted regions. Since the measured levels of Cd in their natural zooplankton food rarely exceed 10 g g-1, and very little of the Cd is found in the soft tissues, the high Cd concentrations (100 to 200 g g–1) in some seaskater species have evidently been derived by drinking from the surface microlayer of the seawater.  相似文献

The composition of amorphous and condensed soil/sediment organic matter (SOM) domains was investigated for one soil sample and four sediment samples. These samples were oxidized with persulfate to remove amorphous SOM, before and after which the composition of SOM was studied by thermogravimetric analysis, pyrolysis-GC/MS, and cross polarization magic angle spinning 13C-NMR. Comparison of the SOM composition before and after oxidation showed that condensed SOM was more thermostable and less polar than amorphous SOM. Condensed SOM was relatively low in O-alkyl C and carboxyl C and it was likely to contain only small amounts of labile organic components (carbohydrates, peptides, fatty acids). Apart from these general characteristics, the composition of the condensed and amorphous domains appeared to be highly dependent on the origin and nature of the SOM investigated. Condensed domains in relatively undecomposed SOM were enriched in aliphatic C, whereas condensed domains in relatively weathered SOM were enriched in aromatic C. Altogether, the compositional changes upon persulfate oxidation were similar to the compositional changes upon humification, which supports the idea that weathered SOM is more condensed than the original material.  相似文献

We investigated a case of massive feto-maternal bleeding by using negative magnetic cell sorting (MACS) and fluorescent in situ hybridization (FISH). A 37-year-old pregnant woman had an uncomplicated amniocentesis for advanced maternal age at 16 weeks' gestation. The fetal karyotype was 46, XY. At 19 weeks' gestation, she had a minor car accident and slight vaginal bleeding. A subsequent Kleihauer-Betke test showed a 140 ml feto-maternal haemorrhage. Serial sonographic examinations indicated a normal fetus and placenta. We performed FISH analysis on maternal peripheral blood at 25 weeks. Anti-CD45 and MACS were used to deplete maternal leucocytes, enriching the proportion of fetal nucleated erythrocytes present. The isolated cells were analysed by using dual-colour FISH with X and Y specific probes. Approximately 65 800 nucleated cells were obtained after MACS depletion. A total of 234 cells were analysed by FISH. The results revealed that 70 of the nucleated cells (30 per cent) were male with one X and one Y signal. Among these cells, six male metaphases were observed in spontaneously dividing cells.  相似文献

Eight pregnancies at risk for cystic fibrosis have been monitored by first-trimester prenatal diagnosis with DNA amplification analysis. The polymerase chain reaction (PCR) was used in all cases to amplify the region detected by KM 19. In two cases, the region detected by CS·7, another DNA probe tightly linked to the CF locus, was also examined. The results of the PCR determinations were confirmed using the Southern blotting procedure, by segregation analysis of restriction fragment length polymorphisms (RFLPs) relative to XV-2c, J3·11, metH, metD, and KM19 probes.  相似文献

Trisomy 21 (T21) is the most common autosomal aneuploidy that is associated with intellectual disability. It is the focus of many prenatal screening programs across the globe. Pregnant women who receive a prenatal diagnosis of T21 in their fetus currently have the option of continuing or terminating their pregnancy, but no fetal treatment is available. In this paper, we review compelling morphogenetic, cellular, and molecular studies that, taken together, suggest that there is an important window of opportunity during fetal life to positively impact brain development to improve postnatal neurocognition and behavior. Although substantial progress has been made in understanding the basic neurobiology of Down syndrome (DS), the majority of pre-clinical trials is currently focused on adults. There are a number of challenges in the identification and development of novel antenatal therapies for DS, including the lack of toxicity and teratogenicity for the pregnant woman and the fetus, evidence that the compounds can cross the placenta and achieve therapeutic levels, and the demonstration of clinical improvement. Preliminary experiments in mouse models suggest that prenatal treatment of DS is an achievable goal. © 2013 John Wiley & Sons, Ltd.  相似文献
