Copepods have been widely used to evaluate toxicity of metals present in marine environments. However, a technical difficulty is to understand the possible routes of metal uptake and to identify in which tissues or organs metals are being accumulated. Traditional techniques are hard to be employed once each organ has to be analyzed separately. Autoradiography is an alternative technique to circumvent this limitation, since metal distribution in tissues can be visualized and quantified, even in small organisms like copepods. In the present study, accumulation and distribution of 64Cu in the copepod Calanus hyperboreus was studied using autoradiography. Copepods were exposed for 2 h to copper (2.3 mg L−1; 1.08 MBq 64Cu mg−1 Cu) and then allowed to depurate for 2 h in clean seawater. Total 64Cu was determined by gamma-spectrometry after a metal exposure and a depuration period. 64Cu distribution was determined based on images generated by autoradiography. Metal accumulation was observed on all external surfaces of the copepods, being accumulated mostly on the ventral region, followed by dorsal, urossoma and internal regions. After depuration, radioactivity levels had a decrease in the sum of external body surface. Our results show that copper uptake by C. hyperboreus is fast and that a non-negligible proportion of the accumulated metal can reach internal tissues, which may lead to detrimental physiological effects. Moreover, whole-body autoradiography was demonstrated to be an efficient technique to study copper accumulation and body distribution in a very small organism such as the copepod C. hyperboreus.  相似文献

The ever-increasing demand for determining pesticides at low concentration levels in different food matrices requires a preliminary step of pre-concentration which is considered a crucial stage. Recently, the parameter of “greenness” during sample pre-concentration of pesticides in food matrices is as important as selectivity in order to avoid using harmful organic solvents during sample preparation. Developing new green pre-concentration techniques is one of the key subjects. Thus, to reduce the impact on the environment during trace analysis of pesticides in food matrices, new developments in pre-concentration have gone in three separate directions: the search for more environmentally friendly solvents, miniaturization and development of solvent-free pre-concentration techniques. Eco-friendly solvents such as supercritical fluids, ionic liquids and natural deep eutectic solvents have been developed for use as extraction solvents during pre-concentration of pesticides in food matrices. Also, miniaturized pre-concentration techniques such as QuEChERS, dispersive liquid–liquid micro-extraction and hollow-fiber liquid-phase micro-extraction have been used during trace analysis of pesticides in food samples as well as solvent-free techniques such as solid-phase micro-extraction and stir bar sorptive extraction. All these developments which are aimed at ensuring that pesticide pre-concentration in different food matrices is green are critically reviewed in this paper.  相似文献

Many air pollution control regulations limit the emission of visible effluents, based on the visual observation of “equivalent opacity.” Because of difficulties encountered in using visual observation, the Texas Air Control Board developed a method of calibration which made it possible to use an instrumental method for measuring visible emissions. A legal regulation based on this instrumental method has been in effect for almost two years. Despite minor difficulties in calibration and maintenance, results have been satisfactory. The use of the instrumental method avoids many of the difficulties inherent in using a regulation based on visual observation, and continued use of the instrumental method is anticipated.  相似文献

Pacific salmon are particularly susceptible to copper (Cu)-induced olfactory injuries that can ultimately inhibit neurobehaviors critical to survival. However, the molecular mechanisms underlying Cu-mediated olfactory impairment remain poorly understood. In the present study, we conducted a short-term Cu exposure at levels relevant to urban runoff (5, 25 and 50 ppb) , and investigated the roles of impaired olfactory signal transduction and induced apoptosis as underlying mechanisms of olfactory injury. Increased cell death in the olfactory epithelium was evident in coho receiving 4 h exposures to 25 and 50 ppb Cu. Expression of olfactory marker protein (omp), a marker of mature olfactory sensory neurons, also decreased at 50 ppb Cu. Immunohistochemical analysis of coho olfactory epithelium demonstrated a loss of type 3 adenylate cyclase (ACIII) in the apical olfactory epithelium cilia at all levels of Cu exposure, suggesting an inhibitory effect of Cu in olfactory signaling. Accompanying the loss of ACIII in Cu-exposed coho were reduced intracellular cyclic guanosine monophosphate (cGMP) levels in the olfactory rosettes. Collectively, these results support a linkage among the initial steps of olfactory signaling in Cu-induced salmon olfactory injury, and suggesting that monitoring olfactory cGMP levels may aid in the assessment of salmon olfactory injury.  相似文献

ABSTRACT A synthetic storm rainfall hyetograph for a one-year design frequency is derived from the one-year intensity-duration curve developed for Cincinnati, Ohio. Detailed rainfall data for a three-year period were collected from three raingages triangulating the Bloody Run Sewer Watershed, an urban drainage areas of 2380 acres'in Cincinnati, Ohio. The advancement of the synthetic storm pattern is obtained from an analysis of the antecedent precipitation immediately preceding the maximum period of three selected durations. Rains which produced excessive runoff at least for some duration were considered only. The same approach can be used for other design frequencies. The purpose of this study is to provide synthetic storm hyetographs to be used as input in deterministic mathematical models simulating urban storm water runoff for the design, analysis and possible surcharge prediction of sewer systems.  相似文献

Amiodarone treatment in pregnancy might be difficult to handle because of the long half-life of the drug (14–28 days up to 2 months) and because it reduces maternal and neonatal thyroid activity. Although short-term use in pregnancy has been described in cases of fetal supraventricular tachycardia, there are few reports on the chronic use of the drug. In this paper we describe our experience with amiodarone treatment in two pregnant sisters with familial dilatative cardiomyopathy and ventricular malignant extrasystole. Prolonged administration of amiodarone (400–200 mg/die) since the beginning of pregnancy did not have any adverse effects; maternal and neonatal thyroid function was normal, as was the neurological and motor development of the neonates.  相似文献

The zinc binding characteristics of natural organic matter (NOM) from several representative surface waters were studied and compared. NOM samples were concentrated by reverse osmosis. The samples were treated in the laboratory to remove trace metals. Square wave anodic stripping voltammetry (SWASV) was used to study zinc complexing properties of those NOM samples at fixed pH, ionic strength, and dissolved organic carbon (DOC) concentrations. Experimental data were compared to the predictions from the Windermere Humic Aqueous Model (WHAM) Version VI. At the same pH, ionic strength, and temperature, the zinc titration curves for NOM samples from different surface water sources tested in our study almost overlapped each other, indicating similarity in zinc binding properties of the NOM. A discrete two-site model gave good fits to our experimental titration data. Non-linear fitting by FITEQL 4.0 shows that the conditional zinc binding constants at the same pH are similar for NOM from different sources, indicating that zinc complexation characteristics of the NOM used in our study do not depend on their origin and one set of binding parameters can be used to represent Zn-NOM complexation for NOM samples from those different surface water sources representing geographically diverse locations. In addition, the total ligand concentrations (L(1,T), L(2,T), and L(T)) of all NOM show no observable gradation with increasing pH (L(1,T)=2.06+/-0.80 mmol/g carbon; L(2,T)=0.12+/-0.04 mmol/g carbon; L(T)=2.18+/-0.78 mmol/g carbon), while the conditional binding constants of zinc by NOM (logK(ZnL)(c)) show a linear increase with increasing pH(logK(1)(c)(pH=6.0)=4.69+/-0.25; logK(1)(c)(pH=7.0)=4.94+/-0.10; logK(1)(c)(pH=8.0)=5.25+/-0.006; logK(2)(c)(pH=6.0)=6.29+/-0.13; logK(2)(c)(pH=7.0)=6.55+/-0.08; logK(2)(c)(pH=8.0)=6.86+/-0.023) with a slope of ca. 0.28, indicating the zinc-NOM complexes become more stable at higher pH. The WHAM VI predicted free zinc ion activities at high zinc concentrations agree with our experimental results at pH 6.0, 7.0, and 8.0. However, the zinc binding of these NOM samples is over estimated by WHAM VI at zinc concentrations below 10(-6) M at pH 8.0.  相似文献

Flax (Linum ustitatissimum L.) is the source of natural fibers that provides biobased products for a variety of existing markets, but considerable processing and cleaning is required. Flax fibers, and bast fibers generally, are produced in the outer regions of the stem between bark and inner core tissues and require retting, which is the microbial separation of fiber from nonfiber tissues, as the first and most limiting stage of processing. Enzyme retting offers a method to overcome disadvantages of the current method, i.e., dew-retting, for high- and consistent-quality fibers with tailored properties for specific applications. Using chemical analyses, microscopy, and microspectroscopy, sites of carbohydrates, aromatics, and waxes plus cutins were identified in flax stems and their relationship to effective enzyme retting determined. Aromatics occur mostly in the inner, core tissues, with the fibers containing only small amounts located sporadically in cell corners of fiber bundles. Therefore, effective retting using enzymes to separate the aromatic-containing tissues from the fibers, but not to degrade aromatic compounds per se, is required. Waxes and cutin in the epidermal regions are effective barriers to enzyme penetration, and mechanical disruption facilitates enzyme penetration into the stems. Pectinases, with chelators to remove Ca++ and destabilize pectin molecules, remove matrix compounds holding fibers within the stem and have been used in effective formulations to ret flax stems.  相似文献
