The aim of this work was to elucidate the role of nitrate as a terminal electron acceptor on the biodegradation of NPEO. We have characterized the products of NPEO degradation by mixed microbial communities in anaerobic batch tests by means of HPLC, 1H NMR and GC–MS. Anaerobic degradation of NPEO was strictly dependent on the presence of nitrate. Within seven days of anoxic incubation, NP2EO appeared as the major degradation product. After 21 days, NP was the main species detected, and was not degraded further even after 35 days. Nitrate concentration decreased in parallel with NPEO de-ethoxylation. A transient accumulation of nitrite was observed within the time period in which NP formation reached its maximum production. The observed generation of nonylphenol coupled to nitrate reduction suggests that the microbial consortium possessed an alternate pathway for the degradation of NPEO, which was not accessible under aerobic conditions.  相似文献

Sulfonamide antibiotics coming from both human and veterinary medicine are among the most common emerging pollutants in freshwater. The present paper shows the successful application of passive sampling using POCIS in combination with an immunochemical ELISA technique and HPLC/MS/MS analysis to study the distribution of sulfonamides in streams around small towns in the Czech Republic, as well as around a major agglomeration of the city of Brno, including its waste water treatment plant (WWTP). Results indicated the presence of sulfonamides at most studied sites with concentrations ranging from <20 up to 736 ng of sulfamethoxazole equivalents per POCIS. Very high levels were detected in both the influent and effluent of the Brno WWTP with maxima > 8000 ng SMX per POCIS. All samplers collected down-stream of the studied towns and WWTPs clearly showed an increase in sulfonamide drug residues. Higher concentrations were determined in rivers at the city of Brno agglomeration. In agreement with other available studies, these findings indicate low efficiency of conventional WWTPs to eliminate polar pharmaceuticals such as sulfonamides. Good performance and correlation with the LC/MS results, as well as ease of use, indicate good potential for the immunochemical ELISA technique to become the screening tool for sulfonamide determination in surface waters including passive samplers.  相似文献

The interactions of cetyltrimethylammonium (CTA) with mitochondria have been investigated. We confirm, as already observed in a previous paper, that this compound behaves as proton carrier (or uncoupler) of the oxidative phosphorylation, but evidences suggest that this compound enhances the membrane permeability to many other compounds such as sucrose. We conclude therefore that CTA as a detergent enhances membrane permeability to all ions including protons. Some evidences are also given that the inhibitory effect of CTA on the mitochondrial respiratory chain is a consequence of the swelling induced.  相似文献

Understanding population declines has been the objective of a wide range of ecological studies. When species have become rare such studies are complicated because particular behavior or life history traits may be the cause but also the result of the decline of a species. We approached this problem by studying species' characteristics on specimens that were collected before the onset of their decline and preserved in natural history museums. In northwestern Europe, some bumble bee species declined dramatically during the 20th century whereas other, ecologically similar, species maintained stable populations. A long-standing debate focuses on whether this is caused by declining species having stricter host plant preferences. We compared the composition of pollen loads of five bumble bee species with stable populations and five with declining populations using museum specimens collected before 1950 in Belgium, England, and The Netherlands. Prior to 1950, the number of plant taxa in pollen loads of declining species was almost one-third lower than that in stable species even though individuals of stable and declining species generally originated from the same areas. There were no systematic differences in the composition of pollen loads between stable and declining species, but the plant taxa preferred by declining species before 1950 had experienced a stronger decline in the 20th century than those preferred by stable species. In 2004 and 2005, we surveyed the areas where bumble bees had been caught in the past and compared the composition of past and present pollen loads of the stable, but not of the by now locally extinct declining species. The number of collected pollen taxa was similar, but the composition differed significantly between the two periods. Differences in composition reflected the major changes in land use in northwestern Europe but also the spread of the invasive plant species Impatiens glandulifera. The main question now is why declining species apparently were not able to switch to less preferred food plants when stable species were. This study shows that natural history collections can play an important role in improving our understanding of the ecological mechanisms driving species population change.  相似文献

The concentrations of gas-phase PCBs were measured from March 2002 to June 2003 at two sites of the Venice Lagoon and at one site of the Euganei hills. The aims of this study were to evaluate the various gas-phase PCB sources, the spatial and temporal variability of PCB concentrations in the gas-phase that enter the Venice Lagoon atmosphere and the influence of the air temperature on PCB trends. The highest annual average concentration of summation PCBs was observed at the station directly influenced by "urban" sources with values about 3 and 1.5 times higher compared to the concentrations found at the stations where "marine" sources and "not subjected to direct local sources" were respectively sampled from. The temporal trends of summation PCBs concentrations were similar at the three sampling stations corresponding to seasonal temperature changes. Greater concentrations occurred in the summer and first autumn months while the lower ones occurred in late autumn and winter. The temperature dependences were investigated using plots of the natural logarithm of the summation PCBs concentrations vs. reciprocal mean temperatures.  相似文献

A comprehensive comparative validation for two different types of dissolved oxygen (DO) analyzers, amperometric and optical, is presented on two representative commercial DO analyzers. A number of performance characteristics were evaluated including drift, intermediate precision, accuracy of temperature compensation, accuracy of reading (under different measurement conditions), linearity, flow dependence of the reading, repeatability (reading stability), and matrix effects of dissolved salts. The matrix effects on readings in real samples were evaluated by analyzing the dependence of the reading on salt concentration (at saturation concentration of DO). The analyzers were also assessed in DO measurements of a number of natural waters. The uncertainty contributions of the main influencing parameters were estimated under different experimental conditions. It was found that the uncertainties of results for both analyzers are quite similar but the contributions of the uncertainty sources are different. Our results imply that the optical analyzer might not be as robust as is commonly assumed; however, it has better reading stability, lower stirring speed dependence, and typically requires less maintenance. On the other hand, the amperometric analyzer has a faster response and wider linear range. Both analyzers seem to have issues with the accuracy of temperature compensation. The approach described in this work will be useful to practitioners carrying out DO measurements for ensuring reliability of their measurements.  相似文献

Edible and medicinal macrofungi used in human diet represent not only important sources of nutritive elements but toxic substances as well (heavy metals and radionuclides). Radioactivity levels of four radionuclides (40K, 137Cs, 226Ra, 228Ra) were determined in the basidiomata (fruiting bodies of a Basidiomycetes) of six lignicolous (Fomitopsis pinicola, Ganoderma applanatum, Hericium clathroides, Megacollybia platyphylla, Pluteus cervinus, Trametes gibbosa) and three mycorrhizal (Boletus luridus, Boletus sp. 1, Boletus sp. 2) species as well as their soil (wood) substrates by gamma spectrometry (high-resolution high-purity germanium (HPGe) detector). The aim was to investigate their ability for radionuclide absorption according to transfer factors (from soil and wood), to predict potential bioindicator species as well as species with potential risk for human use. Samples were taken during years 2011 and 2012, at two sites in forest ecosystem of Tara Mountain (Serbia). Observed concentration ranges per dry weight were as follows: 29–3,020 Bq/kg (40K), 21.9–735 Bq/kg (137Cs), 3–39 Bq/kg (226Ra), and 2.0–18 Bq/kg (228Ra). Obtained results indicate that the type of basidiome (fleshy/tough), most likely due to a different metabolic rate, has a very important role in radionuclide accumulation. The highest activity concentrations of all analyzed radionuclides were found in species with fleshy basidiomata—P. cervinus, H. clathroides, M. platyphylla, and Boletus species. A species-specific influence on radionuclide uptake was more prominent comparing to habitat differences and the role of fungal trophic mode. No significant variations were observed regarding radionuclide activity among the same fungal species from different sampling sites.  相似文献
