Bioaugmentation of contaminated sites with microbes that are adapted or genetically engineered for degradation of specific toxic compounds is an area that is currently being explored as a clean-up option. Biomarkers have been developed to track the survival and efficacy of specific bacteria that are used as inocula for bioremediation of contaminated soil. Examples of biomarkers include the luc gene, encoding firefly luciferase and the gfp gene, encoding the green fluorescent protein (GFP). The luc gene was used to tag different bacteria used for bioremediation of gasoline or chlorophenols. The bacteria were monitored on the basis of luciferase activity in cell extracts from soil. The gfp gene was also used to monitor bacteria during degradation of chlorophenol in soil, based on fluorescence of the GFP protein. Other biomarkers can also be used for monitoring of microbial inocula used for bioaugmentation of contaminated sites. The choice of biomarker and monitoring system depends on the particular site, bacterial strain and sensitivity and specificity of detection required.  相似文献

Focusing on a 2.2 million hectare area surrounding the Lolo National Forest in western Montana, USA, we illustrate a GIS method for predicting patterns of human use on public lands and highlighting potential for impacts on fish and wildlife species. Data inputs include human population count (derived from the 1990 Census), roads and trails, and the predicted distributions of bull trout (Salvelinus confluentus) and 41 terrestrial vertebrates of special concern. Because results highlight areas where conflicts between humans and resources may occur, they are of potential use to land managers. This approach can be applied wherever data are available, and inputs can be varied according to the topics of interest.  相似文献

A site in mid-western Sweden contaminated with chlorinated solvents originating from a previous dry cleaning facility, was investigated using conventional groundwater analysis combined with compound-specific isotope data of carbon, microbial DNA analysis, and geoelectrical tomography techniques. We show the value of this multidisciplinary approach, as the different results supported each interpretation, and show where natural degradation occurs at the site. The zone where natural degradation occurred was identified in the transition between two geological units, where the change in hydraulic conductivity may have facilitated biofilm formation and microbial activity. This observation was confirmed by all methods and the examination of the impact of geological conditions on the biotransformation process was facilitated by the unique combination of the applied methods. Within climate change impact research, the consideration of socioeconomic processes remains a challenge. Socioeconomic systems must be equipped to react and adapt to global change. However, any reasonable development or assessment of sustainable adaptation strategies requires a comprehensive consideration of human-environment interactions. This requirement can be met through multi-agent simulation, as demonstrated in the interdisciplinary project GLOWA-Danube (GLObal change of the WAter Cycle; ). GLOWA-Danube has developed an integrated decision support tool for water and land use management in the Upper Danube catchment (parts of Germany and Austria, 77,000 km2). The scientific disciplines invoked in the project have implemented sixteen natural and social science models, which are embedded in the simulation framework DANUBIA. Within DANUBIA, a multi-agent simulation approach is used to represent relevant socioeconomic processes. The structure and results of three of these multi-agent models, WaterSupply, Household and Tourism, are presented in this paper. A main focus of the paper is on the development of global change scenarios (climate and society) and their application to the presented models. The results of different simulation runs demonstrate the potential of multi-agent models to represent feedbacks between different water users and the environment. Moreover, the interactive usage of the framework allows to define and vary scenario assumptions so as to assess the impact of potential interventions. It is shown that integrated modelling and scenario design not only provide valuable information, but also offer a platform for discussing complex human-environment-interactions with stakeholders.  相似文献

In the buoyancy and turbulence-driven atmospheric circulations (BTDAC) that occur over urban areas where the approach means wind speeds are very low (less than turbulent fluctuations and typically < 3 m/sec), the surface temperatures are significantly higher than those in the external rural areas, and the atmosphere above the mixing layer is stably stratified. In this paper, the mechanisms of BTDAC formation are studied through laboratory experiments and modelling, with additional low-level inflow from external rural areas and a divergent outflow in the opposite direction in the upper part of the mixed layer. Strong turbulent plumes in the central region mix the flow between lower and higher levels up to the inversion height. There are shear-driven turbulent eddies and weaker buoyant plumes around the periphery of the urban area. As the approach flow is very weak, the recirculating streamlines within the dome restrict the ventilation, and the dispersion of pollution emitted from sources below the inversion height leading to a rise in the mean concentration. Low-level air entrained from rural areas can, however, improve ventilation and lower this concentration. This trend can also be improved if the recirculating structure of the BTDAC flow pattern over urban areas breaks down as a result of the surface temperature distribution not being symmetrical, or as the approach wind speed increases to a level comparable with the mean velocity of circulation, or (except near the equator) the urban area is large enough that the Coriolis acceleration is significant.  相似文献
