Water samples from six bays were taken over a 5-year period (1988 to 1992) to determine the distribution and abundance of loricate choanoflagellates in coastal Newfoundland, and to assess the impact that these organisms might have on this cold ocean food web. Scanning electron microscopy was used to study the morphology of these flagellates, allowing us to identify 11 species of loricate choanoflagellates. Parvicorbicula socialis (Meunier) Deflandre was the most abundant species (80 × 103 cells l−1), particularly during the spring diatom bloom. Single-cell species, such as Bicosta spini fera (Throndsen) Leadbeater and Calliacantha natans (Grontved) Leadbeater, were found more commonly after the spring diatom bloom in the summer months. Many of the single-cell choanoflagellates were attached to bacteria-rich microaggregates and debris in the water column and in unpoisoned sediment traps. The P. socialis cell flux was calculated to be 5.3 × 106 cells m−2 d−1 in late May sediment traps. P. socialis in the upper 100 m of the water column was removing 0.3% of the standing crop of bacteria each day (April/May), and the equivalent of 7.4% of the daily bacterial production over the water column. Diel studies of P. socialis in Conception Bay suggest that the sharp decline in population numbers observed in midnight samples may be related to the high number of grazing zooplankton observed during the same period. Pelagic tunicate and zooplankton fecal pellets were found to contain large numbers of choanoflagellate costae, thus providing a direct link from the microbial loop to the macrozooplankton. The dispersion of material released from a point source immediately upwind of an obstacle array has been examined in a hydraulic flume with a low level of background turbulence. The main purpose of the experiments was to examine the interaction of the plume and the internal boundary layer (IBL) created over the obstacle array. The obstacle array consisted of 11 rows of cubes at 16% packing density in a staggered arrangement. Plume dispersion was measured using flow visualization with Rhodamine dye and also with a thermal tracer technique. During the experiments the source release height was varied between z = 0 and z = 4H, where H is the obstacle height. For the low-level releases, the upper boundary of the plume followed the growth of the IBL over the array. For higher level releases (z/H 2) the rate of plume growth was much reduced until the point downstream where it descended into the IBL, after which it experienced the intense turbulent mixing within the array. This suggests that the urban lateral spread parameter y should be a strong function of height in situations where the turbulence level in the ambient approach flow is low. These results highlight the importance of the ambient turbulence even in strongly obstacle-affected dispersion.  相似文献

Norovirus outbreaks are associated with the consumption of contaminated shellfish, and so efficient methods to recover and detect infectious norovirus in shellfish are important. The Proteinase K digestion method used to recover norovirus from shellfish, as described in the ISO 15216, would be a good candidate but its impact on the virus capsid integrity and thus infectivity was never examined. The aim of this study was to assess the impact of the Proteinase K digestion method, and of the heat treatment component of the method alone, on norovirus (genogroups I and II) and MS2 bacteriophage capsid integrity. A slightly modified version of the ISO method was used. RT-qPCR was used for virus detection following digestion of accessible viral RNA using RNases. MS2 phage infectivity was measured using a plaque assay. The effect of shellfish digestive glands (DG) on recovery was evaluated. In the presence of shellfish DG, a reduction in MS2 phage infectivity of about 1 log10 was observed after the Proteinase K digestion method and after heat treatment component alone. For norovirus GII and MS2 phage, there was no significant loss of genome following the Proteinase K digestion method but there was a significant 0.24 log10 loss of norovirus GI. In the absence of shellfish DG, the reduction in MS2 phage infectivity was about 2 log10, with the addition of RNases resulting in a significant loss of genome for all tested viruses following complete Proteinase K digestion method and the heat treatment alone. While some protective effect from the shellfish DG on viruses was observed, the impact on capsid integrity and infectivity suggests that this method, while suitable for norovirus genome detection, may not completely preserve virus infectivity.  相似文献

ABSTRACT: The population in the Jemez y Sangre Water Planning Region of New Mexico has reached the point at which the demand for water exceeds available supplies, particularly when precipitation is below average, as has frequently occurred in recent years. The desire to develop a sustainable water supply that relies on renewable supplies in wet years and preserves the water in storage for times of drought motivated a diverse set of stakeholders in the region to participate in regional water planning. The planning effort culminated in development of the Jemez y Sangre Regional Water Plan, which was adopted by municipal and county governments in the region. The plan assesses and compares water supply and demand in the region and recommends alternatives for protecting and restoring the existing water supply and addressing the pending gap between supply and demand anticipated by the year 2060. To convey to decision makers the alternatives available to solve the future water shortage, option charts were developed to portray the amount of water that could be obtained or conserved through their implementation. The option charts show that the projected gap between supply and demand cannot be met through one alternative only, but will require a combination of alternatives.  相似文献

ABSTRACT: We suggest that a diagnostic procedure, not unlike that followed in medical practice, provides a logical basis for stream channel assessment and monitoring. Our argument is based on the observation that a particular indicator or measurement of stream channel condition can mean different things depending upon the local geomorphic context and history of the channel in question. This paper offers a conceptual framework for diagnosing channel condition, evaluating channel response, and developing channel monitoring programs. The proposed diagnostic framework assesses reach‐level channel conditions as a function of location in the channel network, regional and local biogeomorphic context, controlling influences such as sediment supply and transport capacity, riparian vegetation, the supply of in‐channel flow obstructions, and disturbance history. Field assessments of key valley bottom and active channel characteristics are needed to formulate an accurate diagnosis of channel conditions. A similar approach and level of understanding is needed to design effective monitoring programs, as stream type and channel state greatly affect the type and magnitude of channel response to changes in discharge and sediment loads. General predictions are made for five channel types with respect to the response of various stream characteristics to an increase in coarse sediment inputs, fine sediment inputs, and the size and frequency of peak flows, respectively. These predictions provide general hypotheses and guidance for channel assessment and monitoring. However, the formulation of specific diagnostic criteria and monitoring protocols must be tailored to specific geographic areas because of the variability in the controls on channel condition within river basins and between regions. The diagnostic approach to channel assessment and monitoring requires a relatively high level of training and experience, but proper application should result in useful interpretation of channel conditions and response potential.  相似文献

A binary response estimation procedure using contingent valuation data is proposed for estimating the incremental environmental benefits associated with the use of constructed wetlands to control agricultural waste-water runoff. The model uses two binary responses per individual while circumventing the non-independence problems associated with follow-up valuation questions. ABSTRACT: This study evaluated the use of a modified pebble count procedure (Bevenger and King, 1995) to detect cumulative watershed effects on headwater streams in the Routt National Forest in northcentral Colorado. The 42 sample reaches were stratified by disturbance (reference or disturbed) and geologic terrene (granitic or mixed sedimentary-volcanic). Water surface slope was a significant control on the number of fine particles in the reference reaches in both terranes, and the data from the disturbed reaches were adjusted accordingly. The disturbed reaches in the granitic terrene generally had a higher percentage of fine particles, and the adjusted number of fine particles was significantly correlated with the number of road crossings. Disturbed reaches in the sedimentary-volcanic (s-v) terrane generally did not have significantly more fine particles, nor were the adjusted numbers of fine particles significantly correlated with any management index. The lack of significant trends in the s-v streams is probably due to differences in weathering between the two rock types, and the location of the sample reach relative to sedimentary outcrops. Two other procedures were also used to assess cumulative watershed effects, with the Pfankuch channel stability rating yielding stronger and more consistent differences between the reference and the disturbed streams than the Tarzwell substrate ratio. We conclude that it may be difficult to define a standard reference condition, and that the number of road crossings is more strongly correlated with the number of fine particles than equivalent clearcut area.  相似文献
