We characterized 32 cold-adapted, psychrophilic and cold-tolerant, yeast strains isolated from alpine habitats with regard to their taxonomy, growth temperature profile, and ability to degrade phenol and 18 phenol-related mono-aromatic compounds at 10 degrees C. Twenty of the strains were identified by sequencing of the ribosomal ITS region as seven species of the basidiomycota: Cryptococcus terreus (three strains), Cryptococcus terricola (one strain), Rhodosporidium lusitaniae (two strains), Rhodotorula creatinivora (10 strains), Rhodotorula ingeniosa (one strain), Mastigobasidium intermedium (one strain), and Sporobolomyces roseus (two strains). Twelve strains sharing closely related ITS sequences could not be identified to the species level; according to their ITS sequence they are included in the Microbotryomycetidae. These 12 strains were psychrophilic (no growth at temperatures above 20 degrees C); one-third of these strains did not grow above 15 degrees C. None of the 32 strains utilized any of the highly volatile mono-aromatic compounds (benzene, toluene, ethylbenzene, nitrobenzene, o-xylene, m-xylene, and p-xylene) as the sole carbon source. Non/low volatile aromatic compounds were degraded in the following order: phenol>hydroquinone>resorcinol>benzoate>catechol>salicylate>p-cresol>m-cresol. o-Cresol, guaiacol, p-nitrophenol, or p-nitrotoluene were not utilized for growth. R. creatinivora strains degraded up to seven compounds, whereas C. terricola and S. roseus strains degraded only two compounds. The toxicity of the compounds was determined via growth inhibition in the presence of toxicants and nutrients at 10 degrees C. R. creatinivora strains were characterized by higher IC50 values than other species, S. roseus was the most sensitive species. The most toxic compounds were the xylene isomers, ethylbenzene, p-nitrophenol, and m-cresol. There was a relation between the chemical structure of the compounds and their toxicity, whereas a relation between the toxicity of the compounds and the ability of the yeasts strains to utilize these compounds for growth was only detected in some cases.  相似文献

Proactive corporations have been looking at methods to determine the real costs of doing business, including environmental costs using environmental cost accounting (ECA) methodology. Many companies do not track or measure environmental costs and therefore do not know their true environmental costs. Unfortunately, conventional accounting practices rarely tease out environmental costs, and the costs associated with environmental compliance remain hidden in general overhead accounts. ECA can bring the world of business and the environment closer. When we have a better handle on environmental costs, we can use this information to better prioritize environmental projects, identify cost improvement projects, and allocate environmental costs to products. In this article we explore how some companies approach ECA and show how to initiate an ECA program.  相似文献

Domoic acid (DA), the amnesic shellfish toxin, is a food-web-transferred algal toxin that has been detected in many marine organisms from copepods to whales. However, cephalopods, which are important members of the food chain, have never been implicated in DA transfer or accumulation. Here, we present data showing relevant values of DA detected in the common octopus (Octopus vulgaris) from the Portuguese continental coast. Even though DA is hydrophilic and is not expected to be accumulated in the tissues, DA was always detected in our octopus tissue samples. Tissue distribution of DA revealed that the digestive gland and the branchial hearts are the main organs of DA accumulation. Highly variable DA concentrations, ranging from 1.1 to 166.2 g DA g–1, were observed in the digestive glands. Low levels of DA were detected in the digestive tract (stomach and intestine) and could be a consequence of high digestion rates or a result of non-exposure to toxic vectors during the sampling period. In fact, octopus prey, such as bivalves, crustaceans and fishes, are known to occasionally work as DA vectors. Consequently, DA uptake into octopus tissues is likely sporadic. Similar low levels were detected in the kidney, gills, systemic heart, posterior salivary glands and mantle, and no DA was found in either the gonads or the ink sac. These data are the necessary first step towards achieving an understanding of the accumulation of phycotoxins in O. vulgaris.Communicated by S.A. Poulet, Roscoff  相似文献

Summary. Worker policing in honeybees predicts the evolution of a mechanism to discriminate between queenand worker-born eggs. Although it has been postulated that this discrimination is based on an egg recognition pheromone, neither the chemistry nor the glandular source were elucidated. To verify whether egg discrimination might be based on structural differences, we compared the ultrastructure surface of queen-laid diploid and haploid eggs to that of worker-laid eggs using SEM. Only small differences between the different types of eggs were found. Thus, at least based on the fine structure of the egg surface, queen eggs are indistinguishable from worker-laid eggs.To explore the chemosensory hypothesis for egg discrimination, we conducted a detailed comparative chemical analysis of the different egg types. The coating of all egg types was dominated by linear alkanes, but queen eggs, diploid and haploid, differed from those of workers on two accounts: 1. The diversity of compounds found on queenlaid eggs was much greater than found on worker-laid eggs, mainly due to the number of hydrocarbons. 2. Acetates of some fatty alcohols, alkenes and especially monomethylalkanes were characteristic to queen eggs. The origin of the two latter substances and the acetates is still unknown. Whether these compounds constitute the signal that enables police workers to discriminate between queen- and workerborn eggs remains to be investigated.  相似文献

Experiments reported here were carried out to investigate the use of acoustic cues by palmate newts (Lissotriton helveticus) for orientation and to study whether this behavior is learned, or whether two populations of palmate newts that cohabit with different frog species (Iberian green frog, Rana perezi, and European common brown frog, Rana temporaria) show different phonotactic preferences. The orientation tests consisted of presenting a control stimulus (white noise), a sympatric acoustic stimulus (calls of R. perezi or R. temporaria, depending on the origin of newts), or an allopatric stimulus (calls of natterjack toads, Bufo calamita, or R. perezi). Newts were released in a circular arena, while the acoustic stimuli were presented outside of the circular arena in four different compass orientation directions (0, 90, 180 and 270°). In this study, we show that L. helveticus performed positive phonotaxis toward the calls of R. perezi only when both species shared habitat, orienting randomly when R. perezi was absent from the newt’s natal population. Newts from both populations oriented randomly when exposed to the allopatric and control acoustic stimuli. These results suggest, for the first time, that recognition of the sympatric heterospecific calls could be learned. However, newts sharing the breeding pond with a population of R. temporaria oriented randomly when exposed to the calls of this species. The fact that the breeding seasons of R. temporaria and L. helveticus do not overlap in time does not allow the use of R. temporaria calls as a guidance mechanism for migrating individuals of L. helveticus.  相似文献

BACKGROUND: Plant cell cultures may serve as biosensors for the detection of heavy metals and other toxic substances. Standard culture media and protocols are frequently utilised, but in these media no care is usually taken to control the influence of hormones and nutrients on the reaction of the enzymes or m under consideration as parts of the sensor. The present paper investigates the influence of media composition on the reaction of spruce cells towards heavy metals. METHODS: Spruce cell cultures were grown in a standard medium, either i) alone, ii) containing 0.3% sucrose or iii) containing 3% sucrose and the hormones BAP and NAA. The cell cultures were then incubated in medium with fungal elicitor, H2O2, CdSO4 (50 to 500 microM), or, alternatively, with a standard heavy metal mixture containing 80 microM Na2HAsO4, 150 microM CdSO4 and 200 microM PbCl2. RESULTS: Depending on the nutrient status and hormone availability, large differences in glutathione contents and the GSH/GSSG ratio were observed. However, the cellular redox state seemed to remain more or less constant. Glutathione S-transferase activity was determined with four substrates, and high induction rates for the conjugation of three substrates were observed when hormones were omitted from the media. 1,2-epoxy-nitrophenoxy-propane conjugation was highest in starving cell in the presence of hormones, showing a transient GST induction, with highest rates occurring after 16 hrs following incubation; the induction effect was lost after 24 hrs. CONCLUSION: A medium containing 3% sucrose and both hormones (BAP and NAA) appears to be most favourable for cellular growth as well as the expression of a basis level of detoxification enzymes and antioxidants. With this combination, early responses towards heavy metals at low concentration can be monitored. RECOMMENDATIONS AND PERSPECTIVE: Plant cell cultures are valuable tools for the bioindication of heavy metals and toxic xenobiotics. If standard media and protocols are utilised, the influence of hormones and nutrients on the reaction of the biosensor have to be evaluated thoroughly.  相似文献
