This study examined the adverse effects of TiO2 nanoparticle (nano-TiO2) on the kidney and liver of Wistar rats. Changes of serum biochemical parameters and pathological lesions indicated that liver and kidney were significantly affected in animals treated with 50?mg?kg?1 of nano-TiO2. The inverse relationship between the level of reactive oxygen species and the activities of superoxide dismutase, catalase, and glutathione peroxidase indicates that nano-TiO2 induces oxidative stress. A significant increase in the apoptosis of liver and kidney in a dose-dependent manner was also observed. The ultrastructural observations confirmed the internalization of nano-TiO2 and their direct involvement in the mitochondria-mediated cytotoxicity. Data indicated that nano-TiO2 induce oxidative stress which produces genotoxicity such as oxidative DNA damage, micronuclei (MN) induction, and cell apoptosis in liver and kidney. 相似文献

The present investigation was aimed to determine the effect of chronic exposure of carbendazim on cellular changes in testes of male goats. The goats were randomly divided into two groups, control and treatment (N?=?7 each). The treatment group was exposed to carbendazim at the dose rate of 50?mg?kg?1 body weight per day orally, once daily for 180 consecutive days. Testes were removed from control and experimental animals surgically on the 90th, 120th, and 180th day. On the 180th day of the treatment, a maximum number of seminiferous tubules became atrophic, and vacuolization of germinal epithelium, and sloughing of the germinal epithelium and sertoli cells was more marked as compared to days 90 and 120. Some of the seminiferous tubules were devoid of germinal cells. These pathological findings were supported by ultrathin and electron microscopic examination. On the basis of the present investigation it could be concluded that chronic exposure of carbendazim in male goats did not allow proper maturation of testis. 相似文献

A method is reported for the determination of methyl violet in the range of 10–120 nmol L?1. The method is based on the catalytic effect of silver nanoparticles (AgNPs) on the oxidation reaction of methyl violet by potassium bromate in acid medium. The reaction is followed spectrophotometrically by measuring the change in absorbance () at 620 nm using a fixed time method. The reaction variables were optimized in order to achieve highest sensitivity. The 3б criterion detection limit was 5 nmol L?1, and the relative standard deviation for ten replicate determinations at a concentration of methyl violet of 15 nmol L?1 was 0.97% (n = 10). The Sonbhadra district in the Singrauli area of Uttar Pradesh, India, has many coal mines and thermal power plants and is a critically polluted area. Many residents of this area reported adverse health conditions which may be linked to metal pollution, especially of mercury investigated here.In May 2012, samples of water (23), soil (7), blood, hair, and nails from persons showing adverse health conditions selected at random were collected and analyzed for total mercury by atomic absorption spectrometry.Twenty percent drinking water samples contained mercury from 3 to 26 μg L?1 (3–26 times the permissible limit). Soil samples had 0.5–10.1 mg kg?1 Hg.The average concentrations of mercury in human blood, hair, and nails were found to be 34 μg L?1, 7.4 mg kg?1, and 0.8 mg kg?1, respectively. Mercury concentrations in the blood of these persons were 45 and 28 μg L?1 on average in the case of men and women. This is much higher than the safe level of 5.8 μg L?1 set by the United States Environmental Protection Agency (USEPA).It was concluded that all residents of Sonbhadra sampled could be suffering from mercury toxicity as the area is polluted by Hg released from the coal-fired thermal power plants. 相似文献

The aim of the present study was to evaluate the potential toxicity and general mechanisms involved in single walled carbon nanotubes (SWCNTs)-induced cytotoxicity using human embryonic kidney cell line (HEK293) cells. Carbon nanotubes (coded as CNT) used in this study were synthesized by the chemical vapor deposition method. To elucidate the possible mechanisms underlying SWCNT-induced cytotoxicity, cell viability, cell membrane damage (lactate dehydrogenase activity (LDH) assay), reduced glutathione (GSH), interleukin-8 (IL-8) and lipid peroxidation products levels were quantitatively assessed following SWCNT exposure for 48 hr using HEK293 cells. Exposure of cells to SWCNT at 3–300 μg/ml produced significant reduction in cell viability in a concentration-dependent manner. The IC50 value of SWCNT was found to be 87.58 μg/ml. Exposure of HEK cells to SWCNT at 10–100 μg/ml resulted in concentration-dependent cell membrane damage, increased production of IL-8, elevated levels of thiobarbituric acid reactive substances like malondialdehyde and decreased intracellular GSH levels. In summary, exposure to SWCNT resulted in a concentration-dependent cytotoxicity in cultured HEK293 cells that was associated with increased oxidative stress. 相似文献

Binding affinity of metanil yellow and its breakdown product p‐aminodiphenylamine to serum proteins has been studied employing chromatographic separation on Sephadex G‐200 and by paper and polyacrylamide gel electrophoresis. Metanil yellow has more affinity towards albumin than to globulins. The complexing is presumably through electrostatic forces. p‐Aminodiphenylamine on the other hand, preferably binds to globulin fractions of serum protein. However, a stable binding with BSA alone was also observed. The binding was quite stable and was accompanied by a shift in absorbance from 430 nm to 500 nm. Aspartic acid moiety of protein was found to be one of the units involved in the binding of p‐ADPA to proteins. 相似文献

Differential Pulse Anodic Stripping Voltammetry (DPASV) was employed to measure Apparent Copper Complexing Capacity (Lt) and Stability Constants (Ks) of related cupric complexes of cell exudates of the phytoplanktonic alga Dunaliella salina. Three natural seawater samples collected 0.1, 2, 5 miles off the Tyrrhenian coast were used as culture medium. The measurements were carried out 1h and 7 days after the inoculum. Furthermore, on one culture the medium was passed through C18‐SEP‐PAK cartridges in order to separate the cell exudates. The results show that Dunaliella salina influences the value of Lt but does not affect the value of Ks . The C18SEP‐PAK retain only 50% of the ligands produced by the cells and capable of complexing copper. 相似文献

The interaction of HCH (50 mg/kg) and dietary protein levels on microsomal drug metabolizing enzymes system and liver lipids were studied in the rats for 90 days. The results indicated that rats fed a lower protein diet and HCH has a higher rate of mortality, lower rate of growth and an increased liver weight. A significant induction in the hepatic microsomal aminopyrine‐N‐demethylase, p‐nitroanisole‐O‐dealkylase, benzo(a)pyrene hydroxylase and glutathione‐S‐transferase activity was observed in pesticide treated animals as compared to control animals. The pathological changes observed in liver of HCH treated animals consisted mainly of necrosis and fatty degeneration of hepatocytes. HCH also induced the significant accumulation of cholesterol, triglycerides, phospholipid and total lipid in liver in low protein diet animals. Protein accelerates the metabolism of HCH, resulting in a decrease of HCH concentration with the increase of dietary protein level. A close correlation existed between lipid accumulation, induction of drug metabolizing enzyme system and deposition of HCH in liver. 相似文献

The genotoxicity of chromium chloride was investigated in cells of D7 strain of Saccharomyces cerevisiae harvested either from logarithmic or stationary growth phase. A weak induction of mifotic gene conversion and point reverse mutation was obtained when the incubations were performed using phosphate buffer. No genetic effect was observed when the incubations were performed using Tris‐HCl buffer. The experiments with 51Cr radiotracer demonstrated that Cr3+ ion enters the yeast cells and binds to DNA even if the incubation mixture was performed with Tris HCl buffer. This behaviour could be due to the highest concentration of CrCl3 that cause some damages to cytoplasmatic membrane. 相似文献

An anayltical procedure based on a sequential extraction scheme is applied to samples of suspended particulates from an urban area of Barcelona. This area presents high levels of air pollution owing to automotive emissions and industrial activity. The procedure allows the partitioning of particulate trace metals into four fractions: soluble and exchangeable, bound to carbonates and iron‐manganese oxides, bound to organic matter and residual. In this work, the chemical associations of Fe, Pb, Cu, Mn, Cr and Cd have been investigated. In the analysed samples, only the cadmium is predominantly in the first fraction whereas the other metals are largely associated to carbonates and Fe‐Mn oxides. 相似文献
