We used an immunofluorescence technique to investigate the effects of varying light regimes on ribulose 1,5-bisphosphate
carboxylase/oxygenase (Rubisco) in individual cells of the chlorophyte Dunaliella tertiolecta Butcher. These studies were carried out between September 1995 and February 1996. The population was heterogeneous with respect
to Rubisco localization in the pyrenoid, i.e. in some cells Rubisco staining was highly concentrated in the pyrenoid while
in others it was evenly distributed throughout the chloroplast stroma. When light intensity was varied sevenfold, the fraction
of the cell population that displayed distinct Rubisco staining in the pyrenoid (PR-index) was correlated with light intensity,
although the average Rubisco abundance per cell or per total cellular protein appeared fairly constant. In darkness, or when
treated with 3-(3,4-dichlorophenyl)-1,1-dimethyl urea (DCMU), the PR-index decreased markedly during the first 4 and 3 h,
respectively, and then remained at low levels, while the cell division cycle progression remained unaffected. We conclude
that D. tertiolecta probably possesses an adaptive mechanism, i.e. the redistribution of Rubisco between the pyrenoid (probably the active site
of Rubisco activation and CO2 fixation) and the stroma (probably a reservoir of deactivated Rubisco that is readily available for transport to the pyrenoid
and activation), to respond to variations in irradiance or photosynthetic inhibition. Our results also suggest that this mechanism
is insensitive to slight variations in growth irradiance and to seasonal changes in photoperiod and temperature.

We studied the influence of hydroxypropylcyclodextrins (HPCDs) on the toxicity of some mixtures. Using the Photobacterium phosphoreum toxicity test, the joint toxicological effect for Mixture I (containing p-nitrobenzaldehyde and 1-nitronaphthalene) and Mixture II (containing p-nitrobenzaldehyde and malononitrile) were determined in water and in aqueous solutions of HPCDs. The results indicate that, although the toxicological joint effect for Mixture I (simple addition) differs from that of Mixture II (synergism), alpha- and beta-HPCD can significantly reduce the toxicity of the test compounds, whereas gamma-HPCD has only a slight effect. Explanations for these observations are given that invoke the molecular structure of the individual chemicals as well as the structures of HPCDs. This provides information to assist the application of HPCDs in remediation of environmental pollution. 相似文献

Degradation of di-butyl-phthalate by soil bacteria 总被引:2,自引:0,他引:2
Twelve Gram-positive phthalate ester degraders were isolated from soil. Using Biolog GP2 plates, eight of them were identified as belonging to the Corynebacterium-Mycobacterium-Nocardia group, while the remaining four were unidentifiable. When cultured in the presence of di-butyl-phthalate (DBP) in basal salts solution, five of these isolates accomplished more than 90% of DBP degradation within 48 h (fast group), three were placed in the medium group, and the remaining four were placed in the slow group which caused less than 30% of DBP degradation within the same period of time. A 420 bp DNA fragment was amplified from six isolates and none of them fell within the slow group. When compared with the large subunit of phthalate dioxygenase gene (phtA) of Arthrobacter keyseri, 83% and 91% similarities were evident in the nucleotide and amino acid sequences, respectively. However, no correlation between cell surface hydrophobicity and phthalate degradation ability was evident. Six surfactants (Brij 30, Brij 35, Tergitoltype NP-10, Triton N-101, Triton X-100 and SDS) were tested for their abilities to increase degradation rate. When added at the critical micellar concentration (CMC), they all displayed strong growth inhibition against the three bacteria tested, with Brij 30 been the least toxic to isolates G2 and G11, and Brij 35 had the least inhibitory effect for G1. When half the CMC of Brij 30 was incorporated into the basal salts, the inhibitory effect on DBP degradation remained. Soil helped to minimize surfactant toxicity of surfactant and increase the degradation potential of some of the test bacteria. When DBP-amended soil had been aged for three months, decreases in bioavailability were observed but the effect varied tremendously between different organisms. For isolates G1, G2, G5, G7 and G17 the aging effects were almost non-exist. The present study indicates that selection of a suitable degrader may minimize the undesired effect of aging on bioremediation process. 相似文献

This study was designed to investigate the transportation, distribution, and bioaccumulation of PCBs in various environmental media and compartments using an ecologically simulated growth chamber. Spatial and temporal trends of PCBs in the growth chamber were discussed. The release of PCB congeners in soil was affected by the amount of rainfall with the transporting direction moving away from PCBs contaminated point. Two pathways of PCBs accumulation in plants were the uptake of roots and the deposition on shoots/leaves. There were 29 PCB congeners been found in the lalang grass. Higher concentrations of lower chlorinated PCBs were identified than higher chlorinated PCBs because of relatively higher vapor pressure for lower chlorinated congeners. After 10months of monitoring, PCBs were detected in water samples which were contributed by the release of PCBs from leached soil. Analysis of sediment showed that the percentages of low- and mid-chlorinated biphenyls were decreased 1% and 13%, respectively compared to the increase (14%) of high-chlorinated biphenyls. The increase of high-chlorinated PCBs was possibly caused by their low hydrophilicity which had resulted higher adsorption rate in sediment. All of five species of fish had been found significant amount of PCBs accumulation ( summation operatorPCBs: 21.7-102.1 microg/g-lipid). The concentrations of PCBs in fish were varied significantly among species. The range of bioaccumulation factors (BAFs) among different species of fish could be as much as 5 times depending on the consumption habits of fish. An abandoned pentachlorophenol plant and nearby area in southern Taiwan was heavily contaminated by dioxins, impurities formed in the PCP production process. The investigation showed that the average serum PCDD/Fs of residents living nearby area (62.5 pg WHO-TEQ/g lipid) was higher than those living in the non-polluted area (22.5 and 18.2 pg WHO-TEQ/g lipid) (P<0.05). In biota samples, average PCDD/F of milkfish in sea reservoir (28.3 pg WHO-TEQ/g) was higher than those in the nearby fish farm (0.15 pg WHO-TEQ/g), and Tilapia and shrimp showed the similar trend. The average daily PCDD/Fs intake of 38% participants was higher than 4 pg WHO-TEQ/kg/day suggested by the world health organization. Serum PCDD/F was positively associated with average daily intake (ADI) after adjustment for age, sex, BMI, and smoking status. In addition, a prospective cohort study is suggested to determine the long-term health effects on the people living near factory. 相似文献

This study investigates the pozzolanic reactions and compressive strength of the blended cement manufactured using synthetic slag obtained from municipal solid waste incinerator (MSWI) cyclone ash and scrubber ash as partial replacement of portland cement. The synthetic slag was made by co-melting the MSWI scrubber ash and cyclone ash mixtures at 1400 degrees C for 30 min. Following pulverization, the different types of slag were blended with cement as cement replacement at ratios ranging from 10 to 40 wt %. The synthetic slag thus obtained was quantified, and the characteristics of the slag-blended cement pastes were examined. These characteristics included the pozzolanic activity, compressive strength, hydration activity, crystal phases, species, and microstructure at various ages. The 90-day compressive strength developed by slag-blended cement pastes with 10 and 20 wt % of the cement replaced by the synthetic slag outperformed ordinary portland cement by 1-7 MPa. X-ray diffraction species analyses indicated that the hydrates in the slag-blended cement pastes were mainly portlandite, the calcium silicate hydrate gels, and calcium aluminate hydrate salts, similar to those found in ordinary portland cement paste. Differential thermal and thermogravimetric analysis also indicated that the slag reacted with portlandite to form calcium silicate hydrate gels. 相似文献

Bioaerosols from wastewater treatment processes are a significant subgroup of atmospheric aerosols. In the present study,airborne microorganisms generated from a wastewater treatment station(WWTS) that uses an oxidation ditch process were diminished by ventilation.Conventional sampling and detection methods combined with cloning/sequencing techniques were applied to determine the groups,concentrations,size distributions,and species diversity of airborne microorganisms before and after ventilation. There were 3021 ± 537 CFU/m3 of airborne bacteria and 926 ± 132 CFU/m3 of airborne fungi present in the WWTS bioaerosol.Results showed that the ventilation reduced airborne microorganisms significantly compared to the air in the WWTS. Over 60% of airborne bacteria and airborne fungi could be reduced after4 hr of air exchange. The highest removal(92.1% for airborne bacteria and 89.1% for fungi) was achieved for 0.65–1.1 μm sized particles. The bioaerosol particles over 4.7 μm were also reduced effectively. Large particles tended to be lost by gravitational settling and small particles were generally carried away,which led to the relatively easy reduction of bioaerosol particles0.65–1.1 μm and over 4.7 μm in size. An obvious variation occurred in the structure of the bacterial communities when ventilation was applied to control the airborne microorganisms in enclosed spaces. 相似文献
