Recent reports have indicated that digital PCR may be useful for the noninvasive detection of fetal aneuploidies by the analysis of cell-free DNA and RNA in maternal plasma or serum. In this review we provide an insight into the underlying technology and its previous application in the determination of the allelic frequencies of oncogenic alterations in cancer specimens. Definition, analysis, and certification of biochar quality are crucial to the agronomic acceptance of biochar. While most biochars have a positive impact on plant growth, some may have adverse effects due to the presence of phytotoxic compounds. Conversely, some biochars may have the ability to adsorb and neutralize natural phytotoxic compounds found in soil. We evaluated the effects of biochars on seedling growth and absorption of allelochemicals present in corn ( L.) residues. Corn seeds were germinated in aqueous extracts of six biochars produced from varied feedstocks, thermochemical processes, and temperatures. Percent germination and shoot and radicle lengths were evaluated at the end of the germination period. Extracts from the six biochars had no effect on percent germination; however, extracts from three biochars produced at high conversion temperatures significantly inhibited shoot growth by an average of 16% relative to deionized (DI) water. Polycyclic aromatic hydrocarbons detected in the aqueous extracts are believed to be at least partly responsible for the reduction in seedling growth. Repeated leaching of biochars before extract preparation eliminated the negative effects on seedling growth. Biochars differ significantly in their capacity to adsorb allelochemicals present in corn residues. Germination of corn seeds in extracts of corn residue showed 94% suppression of radicle growth compared to those exposed to DI water; however, incubation of corn residue extracts with leached biochar for 24 h before initiating the germination test increased radicle length 6 to 12 times compared to the corn residue extract treatments. Germination tests appear to be a reliable procedure to differentiate between effects of different types of biochar on corn seedling growth. 相似文献

Toxicity and hazard of a mixture of SSRIs to zooplankton communities evaluated in aquatic microcosms
The toxicity and hazard of a mixture of selective serotonin reuptake inhibitors (SSRIs), including fluoxetine, fluvoxamine, and sertraline, to zooplankton communities were evaluated using 120,00l outdoor microcosms. Acute (day 4) and chronic (day 35) zooplankton abundance and species richness were assessed for Rotifera, Cladocera, and Copepoda. For acute SSRI exposures, rotifers were the most sensitive zooplankton taxa to changes in abundance (predicted no effect concentration (PNEC)=19 nM); however, no effects in zooplankton species richness were observed for this treatment period. A decrease in Copepoda abundance and species richness was observed following chronic exposures of SSRIs (PNEC=9.1 nM). A 99th-centile predicted environmental concentration (PEC=0.51 nM) yielded HQs at least two orders of magnitude below 1. Therefore, mixtures of SSRIs do not appear to present a hazard to zooplankton communities at environmentally relevant concentrations. 相似文献

Does local competition increase the coexistence of species in intransitive networks? 总被引:1,自引:0,他引:1
Competitive intransitivity, a situation in which species' competitive ranks cannot be listed in a strict hierarchy, promotes species coexistence through "enemy's enemy indirect facilitation." Theory suggests that intransitivity-mediated coexistence is enhanced when competitive interactions occur at local spatial scales, although this hypothesis has not been thoroughly tested. Here, we use a lattice model to investigate the effect of local vs. global competition on intransitivity-mediated coexistence across a range of species richness values and levels of intransitivity. Our simulations show that local competition can enhance intransitivity-mediated coexistence in the short-term, yet hinder it in the long-term, when compared to global competition. This occurs because local competition slows species disaggregation, allowing weaker competitors to persist longer in the shifting spatial refuges of intransitive networks, enhancing short-term coexistence. Conversely, our simulations show that, in the long-term, local competition traps disaggregated species in unfavorable areas of the competitive arena, where they are excluded by superior competitors. As a result, in the long-term, global intransitive competition allows a greater number of species to coexist than local intransitive competition. 相似文献

A stochastic programming formulation considering Conditional-Value-at-Risk (CVaR) is developed for the optimal placement of gas detectors in petrochemical process facilities. A rigorous gas dispersion simulator, FLACS, is used to generate release scenario data for a real process geometry. We consider two problem formulations: minimization of expected detection time and minimization of expected detection time subject to a restriction on CVaR across the scenario set. The extensive form of each stochastic program is formulated in Pyomo and solved using CPLEX. Considering all scenarios, we compare key values and histograms of detection times for both formulations. Minimizing the mean detection time only can lead to optimal detector placements with a good expected behavior, but unacceptable worst-case behavior. The formulations that minimize or constraint CVaR produce sensor placements with significantly better worst-case behavior and fewer scenarios having high detection times. Considering these results, a strong case for the use of optimal sensor placement using stochastic programming considering CVaR is made for improving safety systems. 相似文献

Prenatal diagnosis for Tay-Sachs disease was performed on 25 patients using chorionic villus sampling (CVS). Nineteen were diagnosed as normal, and six were affected. Normal villus extracts had both hexosaminidase (hex) A and B activity, as determined by Cellogel and polyacrylamide gel electrophoresis, while extracts from affected fetuses had only hex B activity. Compared to cultured amniotic fluid cells or fibroblasts, villi contained less hex A. Hex A levels in fresh villi and cultured trophoblasts were roughly comparable. 相似文献

46,XY, −22,+t(22;22)(p11;q11) or i(22q) was diagnosed in 15/15 cells from two cultures from the amniotic fluid culture of a 31-year-old patient whose fetus demonstrated cystic hygroma on ultrasound. Cytogenetic studies performed on fetal skin from the abortus revealed the same karyotype as that seen on amniocentesis, but the placenta demonstrated a 46,XY,46,XY, −22,+t(22;22) or i(22q) mosaicism, with 65 per cent of the cells being 46,XY. This case provides an example of placental mosaicism for a normal male karyotype, while the fetus demonstrated non-mosaic trisomy 22. 相似文献

We investigated whether exposure to environmentally relevant polybrominated diphenyl ethers (PBDEs) causes immunomodulation in captive nestling American kestrels (Falco sparverius). Eggs within each clutch, divided by laying sequence, were injected with safflower oil or penta-BDE congeners-47, -99, -100, and -153 dissolved in safflower oil (18.7 microg sigmaPBDEs/egg) approximating Great Lakes birds. For 29 days, nestlings consumed the same PBDE mixture (15.6+/-0.3 ng/g body weight per day), reaching sigmaPBDE body burden concentrations that were 120x higher in the treatment birds (86.1+/-29.1 ng/g ww) than controls (0.73+/-0.5 ng/g ww). PBDE-exposed birds had a greater PHA response (T-cell-mediated immunity), which was negatively associated with increasing BDE-47 concentrations, but a reduced antibody-mediated response that was positively associated with increasing BDE-183 concentrations. There were also structural changes in the spleen (fewer germinal centers), bursa (reduced apoptosis) and thymus (increased macrophages), and negative associations between the spleen somatic index and sigmaPBDEs, and the bursa somatic index and BDE-47. Immunomodulation from PBDE exposure may be exacerbated in wild birds experiencing greater environmental stresses. 相似文献
