Enteric viruses monitoring in surface waters requires the concentration of viruses before detection assays. The aim of this study was to evaluate different methods in terms of recovery efficiencies of bacteriophage PP7 of Pseudomonas aeruginosa, measured by real-time PCR, using it as a viral control process in water analysis. Different nucleic acid extraction methods (silica–guanidinium thiocyanate, a commercial kit (Qiagen Viral RNA Kit) and phenol–chloroform with alcohol precipitation) exhibited very low recovery efficiencies (0.08–4.18 %), being the most efficient the commercial kit used for subsequent experiments. To evaluate the efficiency of three concentration methods, PBS (as model for clean water) and water samples from rivers were seeded to reach high (HC, 106 pfu ml?1) and low concentrations (LC, 104 pfu ml?1) of PP7. Tangential ultrafiltration proved to be more efficient (50.36?±?12.91, 17.21?±?9.22 and 12.58?±?2.35 % for HC in PBS and two river samples, respectively) than adsorption–elution with negatively charged membranes (1.00?±?1.34, 2.79?±?2.62 and 0.05?±?0.08 % for HC in PBS and two river samples, respectively) and polyethylene glycol precipitation (15.95?±?7.43, 4.01?±?1.12 and 3.91?±?0.54 %, for HC in PBS and two river samples, respectively), being 3.2–50.4 times more efficient than the others for PBS and 2.7–252 times for river samples. Efficiencies also depended on the initial virus concentration and aqueous matrixes composition. In consequence, the incorporation of an internal standard like PP7 along the process is useful as a control of the water concentration procedure, the nucleic acid extraction, the presence of inhibitors and the variability of the recovery among replicas, and for the calculation of the sample limit of detection. Thus, the use of a process control, as presented here, is crucial for the accurate quantification of viral contamination.  相似文献

The lower portion of the Reconquista River is highly polluted. However, little is known about the state of the high and middle basins. The aims of this work were to assess the water quality on the high and middle Reconquista River basins and to determinate if the presence of a reservoir in the river has a positive effect on the water quality. We conducted a seasonal study between August 2009 and November 2010 at the mouth of La Choza, Durazno, and La Horqueta streams at the Roggero reservoir—which receives the water from the former streams—at the origin of the Reconquista River and 17 km downstream from the reservoir. We measured 25 physical and chemical parameters, including six heavy metal concentrations, and performed a multivariate statistical analysis to summarize the information and allow the interpretation of the whole data set. We found that the Durazno and La Horqueta streams had better water quality than La Choza, and the presence of the reservoir contributed to the improvement of the water quality, allowing oxygenation of the water body and processing of organic matter and ammonia. The water quality of the Reconquista River at its origin is good and similar to the reservoir, but a few kilometers downstream, the water quality declines as a consequence of the presence of industries and human settlements. Therefore, the Roggero reservoir produces a significant improvement of water quality of the river, but the discharge of contaminants downstream quickly reverses this effect.  相似文献

The European Water Framework Directive (WFD) establishes a framework for the protection of estuarine and coastal waters, with the most important objective being to achieve 'good ecological status' for all waters, by 2015. Hence, Member States are establishing programmes for the monitoring of water quality status, through the assessment of ecological and chemical elements. These monitoring programmes can be of three types: surveillance monitoring; operational monitoring (both undertaken on a routine basis); and investigative monitoring (carried out where the reason of any exceedance for ecological and chemical status is unknown). Until now, nothing has been developed in relation to investigative monitoring and no clear guidance exists for this type of monitoring, as it must be tackled on a 'case-by-case' basis. Consequently, the present study uses slag disposal from a blast furnace, into a coastal area, as a case-study in the implementation of investigative monitoring, according to the WFD.In order to investigate the potential threat of such slags, this contribution includes: a geophysical study, to determine the extent of the disposal area; sediment analysis; a chemical metal analysis; and an ecotoxicological study (including a Microtox test and an amphipod bioassay). The results show that metal concentrations are several times above the background concentration. However, only one of the stations showed toxicity after acute toxicological tests, with the benthic communities being in a good status. The approaches used here show that contaminants are not bioavailable and that no management actions are required with the slags.  相似文献

The ecological importance of mysid (Crustacea: Mysidacea) populations in coastal food webs is not well understood. Although the 10 or more species of epibenthic mysids found in Clayoquot Sound, BC, Canada, form the primary prey resource for seasonally abundant gray whales (Eschrichtius robustus), little is known about their life history. Here, Holmesmysis sculpta dominates multi-species swarms, however there are several potential routes to this state, one of which is higher embryo production. Reproductive capacity is key to the resiliency of local mysid populations and species diversity, in this study I compare one aspect, brood size for the four most commonly found species, H. sculpta, Neomysis rayii, Exacanthomysis davisi, and Columbiaemysis ignota. The number of embryos per brood was found to vary significantly between species; however, individual length is a stronger determinant of brood size. Here, I report previously unknown life history attributes of coastal mysid species, with important consequences for community structure and local marine food webs.  相似文献

The introduction of non-native species has accelerated due to increasing levels of global trade and travel, threatening the composition and function of ecosystems. Upon arrival and successful establishment, biological invaders begin to spread and often do so with considerable assistance from humans. Recreational areas can be especially prone to the problem of accidental non-native species transport given the number of visitors that arrive from geographically diverse areas. In this paper, we examine camping permit data to the Apostle Islands National Lakeshore in northwestern Wisconsin, USA, from 1999 to 2007 relative to gypsy moth distribution, phenology and outbreak data. During this time, gypsy moth populations became established in this area ahead of the moving population front of the gypsy moth, suggesting anthropogenic introduction. The permit data revealed that the majority of visitors arrived from outside of the gypsy moth established area. However, there was a consistent yearly trend of visitors that arrived from areas of high gypsy moth populations and who arrived during the gypsy moth life stage (egg masses) most likely to be successfully introduced. Using available data on the gypsy moth and its relationship to camping permit data, we describe how recreational managers could optimize park strategies to mitigate unwanted introductions of the gypsy moth as well as develop analogous strategies for managing other biological invaders in recreational areas.  相似文献

This study develops and tests a leadership model that focuses on managers' motivation for attempting the leadership of change. The construct of leadership self‐efficacy (LSE) is defined, and a measure comprising three dimensions (direction‐setting, gaining followers' commitment, and overcoming obstacles to change) is developed. Based on Bandura's (1986) social cognitive theory, the primary hypothesis is that high LSE managers will be seen by direct reports as engaging in more leadership attempts. Relationships are also proposed between LSE and several factors that are expected to influence this confidence judgment. Managers' organizational commitment and crisis perceptions are modelled as potential moderators of the relationship between LSE and leadership attempts. The model was tested through surveys distributed to managers (n = 150) and their direct reports (n = 415) in a real estate management company and an industrial chemicals firm. Positive relationships (p < 0.05) were found between the first two dimensions of LSE and managers' leadership attempts. An interaction effect involving organizational commitment was discovered for the LSE/overcoming obstacles dimension (p < 0.05). Several positive relationships were found between LSE dimensions and proposed antecedents, including self‐esteem (p < 0.05), subordinates' performance abilities (p < 0.05), and managers' job autonomy (p < 0.05). In this article we apply and test a methodology to estimate cumulative frequency distribution for air pollutant concentration from wind-speed data. We use the inverse relationship after Simpson et al. (Atmospheric Environment, 19, 75–82, 1985) between the opposing percentile values in the statistical distributions for air pollutant concentrations and wind-speed data. This relationship is valid, irrespective of the statistical distributions of both variables, if an inverse relationship between them is also applicable. The available data are five years of 8-h average carbon monoxide concentration and 8-h mean wind-speed, observed in Buenos Aires (Argentina). The performance of the obtained empirical expressions in estimating cumulative frequency distributions for 8-h CO is statistically evaluated. The results show that it is possible to obtain an acceptable cumulative frequency distribution for 8-h CO concentration at the site if the cumulative frequency distribution for wind-speed is known. Q–Q plots show a good agreement between estimated and observed values. From our data, the mean relative error of the estimations was found to be as much as 8.0%.  相似文献
