The prenatal diagnosis of the cerebro-hepato-renal syndrome of Zellweger (CHRS) was made by assaying the levels of very long chain fatty acids (VLCFAs) in amniotic fluid cell cultures, obtained by amniocentesis at 16 1/2 weeks of pregnancy. The family-at-risk, because they had previously borne a child with CHRS, accepted these results as indications of an affected fetus, and chose to terminate the pregnancy at 20 1/2 weeks of gestation. The diagnosis was confirned by the phenotype of the aborted fetus and the presence of markedly elevated levels of VLCFAs in fetal liver homogenates. The prenatal diagnosis of CHRS, which can now readily be determined from amniotic fluid cell cultures, is an important step in genetic counselling of families-at-risk for this disease.  相似文献

Biodiversity is highly valuable and critically threatened by anthropogenic degradation of the natural environment. In response, governments have pledged enhanced protected‐area coverage, which requires scarce biological data to identify conservation priorities. To assist this effort, we mapped conservation priorities in Kenya based on maximizing alpha (species richness) and beta diversity (species turnover) of plant communities while minimizing economic costs. We used plant‐cover percentages from vegetation surveys of over 2000 plots to build separate models for each type of diversity. Opportunity and management costs were based on literature data and interviews with conservation organizations. Species richness was predicted to be highest in a belt from Lake Turkana through Mount Kenya and in a belt parallel to the coast, and species turnover was predicted to be highest in western Kenya and along the coast. Our results suggest the expanding reserve network should focus on the coast and northeastern provinces of Kenya, where new biological surveys would also fill biological data gaps. Meeting the Convention on Biological Diversity target of 17% terrestrial coverage by 2020 would increase representation of Kenya's plant communities by 75%. However, this would require about 50 times more funds than Kenya has received thus far from the Global Environment Facility.  相似文献

Single copies of tiny chromosome fragments, appearing either as single or as double minutes, were observed in a high frequency in amniotic fluid cultures of five mothers who underwent prenatal testing because of advanced age. In four cases, the minutes had arisen de novo. The minutes were later confirmed in fetal skin following termination of pregnancy in one case; in another, in cord blood following the birth of a normal boy; and in the third, in peripheral blood of a normal 3-year-old girl. In the fourth case, the minutes were not confirmed in cord blood following the birth of a normal boy. A follow-up chromosome study of the baby boy in the fifth case was not possible but the minutes were maternally transmitted.  相似文献

We describe a simulation model representing the most important human and natural factors driving land use and cover changes (LUCC) in southern Chile. We evaluate the model by examining its ability to simulate LUCC observed over the past three decades, conduct a sensitivity analysis of simulated trends to changes in important model parameters, and use the model to project likely landscape transformations over the next decade under “as usual,” “pessimistic,” and four “optimistic” scenarios. The model consists of five submodels representing LUCC on two distinct soil formations (volcanic ash and gleysols) and four major land use categories: native forest, agricultural land, shrubland, and urban land. Land use and cover sub-categories include old growth forests, secondary forests, and low and flooded shrubland. The model simulated well general historic trends in forest cover, agricultural land, shrubland, and urban land: from a forest-dominated landscape in 1976 to a landscape dominated by shrubland and agricultural land by 2007. Forest loss, forest degradation by logging and clearing for agriculture were the most important direct drivers of LUCC: forest logging and clearing were most important from 1976 to 1985, whereas after 1985 logging for firewood, driven by population growth, was most important. Sensitivity analysis indicated that model projections of general trends in the main land use and cover categories were not overly sensitive to changes in important model parameters, although further study is necessary to improve our estimates of the proportion of pasture requirements supplied by clearing low shrubland. Projections of LUCC suggested that a reduced amount of secondary forest would be left by 2017 if no actions are taken to reduce forest loss (“as usual”). Increasing population (“pessimistic scenario”) resulted in similar trajectories than those predicted by the as usual scenario, whereas reducing logging for firewood and increasing forest recruitment from shrubland could reduce loss of native forest by nearly one-third (“optimistic scenarios”). Surprisingly, shrubland exhibited the most complex and influential dynamics in all scenarios, being the immediate outcome of forest loss and the main long-term source of land for agriculture, urban expansion, and forest recovery. Few studies in Chile, or elsewhere, have considered the importance of this intermediate successional stage. Of the scenarios simulated, financial incentives targeted toward channeling shrubland into regenerated forest seemed most promising, although obstacles to such a management strategy exist.  相似文献

Germplasm collection is important to preserve and maximize genetic diversity for germplasm conservation. Tsuga dumosa ( D. Don) Eichler in Engler & Prantl. and T. chinensis var. forrestii (Downie) Silba germplasm was collected from three localities in China: Mt. Yulong, Wenfeng Temple and Mt. Dishiergu, Yunnan Province. Accessions were identified based on morphological characters and RAPD markers. The shapes of the apices and margins of needles were examined, and the length and width of needles, cones and seeds from accessions of mature plants were used to compare the morphological differences and to identify the germplasm. Molecular markers generated by randomly amplified polymorphic DNA (RAPD) were also used to characterize the taxa. Although the clustering based on RAPD markers was inconsistent with the morphological characters of the needles, based on the overall morphological characters and on RAPD markers, the accessions from Mt. Yulong and Wenfeng Temple were identified as T. chinertsis var. forrestii, and those from Mt. Dishiergu identified as T. dumosa. Taxonomic identification of the accessions was made based on morphology and by RAPD markers concurred. The results indicate that the shapes of the apices and margins of needles particularly from young plants could not be used as a possible key to identify T. dumosa and T. chinertsis var. forrestii. The growing importance of the environment and its management has simultaneously emphasized the benefits of hydroelectric power and its environmental costs. In a changing policy climate, giving importance to renewable energy development and environmental protection, conflict potential between stakeholders is considerable. Navigation of conflict determines the scheme constructed, making sustainable hydropower a function of human choice. To meet the needs of practitioners, greater understanding of stakeholder conflict is needed. This paper presents an approach to illustrate the challenges that face small-scale hydropower development as perceived by the stakeholders involved, and how they influence decision-making. Using Gordleton Mill, Hampshire (UK), as an illustrative case, soft systems methodology, a systems modeling approach, was adopted. Through individual interviews, a range of problems were identified and conceptually modeled. Stakeholder bias towards favoring economic appraisal over intangible social and environmental aspects was identified; costs appeared more influential than profit. Conceptual evaluation of the requirements to meet a stakeholder-approved solution suggested a complex linear systems approach, considerably different from the real-life situation. The stakeholders introduced bias to problem definition by transferring self-perceived issues onto the project owner. Application of soft systems methodology caused a shift in project goals away from further investigation towards consideration of project suitability. The challenge of sustainable hydropower is global, with a need to balance environmental, economic, and social concerns. It is clear that in this type of conflict, an individual can significantly influence outcomes; highlighting the need for more structured approaches to deal with stakeholder conflicts in sustainable hydropower development.  相似文献

This paper reports on the effect of aqueous and nano-particulated Pb on oxidative stress (lipid peroxidation), cytoxicity, and cell mortality. As determined by the Thiobarbituric Acid Reactive Substances (TBARS) method, only 6 h after incubation aqueous suspensions bearing nano-sized PbO2, soluble Pb(II), and brain-homogenate only suspensions, were determined to contain as much as ca. 7, 5, and 1 nmol TBARS mg protein−1, respectively. Exposure of human cells (central nervous system, prostate, leukemia, colon, breast, lung cells) to nano-PbO2 led to cell-growth inhibition values (%) ca. ≤18.7%. Finally, as estimated by the Artemia salina test, cell mortality values were found to show high-survival larvae rates. Microscopic observations revealed that Pb particles were swallowed, but caused no mortality, however.  相似文献

A battery of biomarkers has recently been developed in the earthworm Eisenia andrei. In this study, different biomarkers (i.e. Ca2+-ATPase activity, lysosomal membrane stability-LMS, lysosomal lipofuscin and neutral lipid content) were utilized to evaluate the alterations in the physiological status of animals, induced by exposure for 3 d to different sublethal concentrations of TCDD (2,3,7,8-tetrachlorodibenzo-p-dioxin) (1.5 × 10−3, 1.5 × 10−2, 1.5 × 10−1 ng mL−1) utilizing the paper contact toxicity test. Lysosome/cytoplasm volume ratio and DNA damage were also evaluated as a biomarker at the tissue level and as a biomarker of genotoxicity, respectively. Moreover, the NR retention time assay conditions were optimized for the determination of in vivo LMS in earthworm coelomocytes. The results demonstrate that LMS and Ca2+-ATPase activity were early warning biomarkers able to detect the effects of minimal amounts of TCDD and that biomarkers evaluated at the tissue level are important for following the evolution of the stress syndrome in earthworms. To evaluate the health status of the animals, an Earthworm Expert System (EES) for biomarker data integration and interpretation was developed. The EES proved to be a suitable tool able to rank, objectively, the different levels of the stress syndrome in E. andrei induced by the different concentrations of TCDD.  相似文献

One monitoring station is insufficient to characterize the high spatial variation of traffic-related heavy metals within cities. We tested moss bags (Hylocomium splendens), deployed in a dense network, for the monitoring of metals in outdoor air and characterized metals’ long-term spatial distribution and its determinants in Girona, Spain. Mosses were exposed outside 23 homes for two months; NO2 was monitored for comparison. Metals were not highly correlated with NO2 and showed higher spatial variation than NO2. Regression models explained 61-85% of Cu, Cr, Mo, Pb, Sb, Sn, and Zn and 72% of NO2 variability. Metals were strongly associated with the number of bus lines in the nearest street. Heavy metals are an alternative traffic-marker to NO2 given their toxicological relevance, stronger association with local traffic and higher spatial variability. Monitoring heavy metals with mosses is appealing, particularly for long-term exposure assessment, as mosses can remain on site many months without maintenance.  相似文献

Particulate matter (PM) emitted from cattle feedlots are thought to affect air quality in rural communities, yet little is known about factors controlling their emissions. The concentrations of PM (i.e., PM2.5, PM10, and total suspended particulates or TSP) upwind and downwind at two large cattle feedlots (KS1, KS2) in Kansas were measured with gravimetric samplers from May 2006 to October 2009 (at KS1) and from September 2007 to April 2008 (at KS2). The mean downwind and net (i.e., downwind - upwind) mass concentrations of PM2.5, PM10, and TSP varied seasonally, indicating the need for multiple-day, seasonal sampling. The downwind and net concentrations were closely related to the moisture content of the pen surface. The PM2.5/PM10 and PM2.5/TSP ratios at the downwind sampling location were also related to the moisture content of the pen surface, humidity, and temperature. Measurement of the particle size distribution downwind of the feedlot with a cascade impactor showed geometric mean diameter ranging from 7 to 18 microm, indicating that particles that were emitted from the feedlots were generally large in size.  相似文献
