Molecular techniques and modelling are presented as powerful tools required in the performance of efficient soil and water bioremediation systems. An Escherichia coli CC118-D strain was constructed by inserting the Klebsiella pneumoniae hpa B gene, coding for the unstable 3,4-dihydroxyphenylacetate 2,3-dioxygenase, into its chromosome. When the constructed strain was immobilized, both enzyme stability and viability increased along the studied period, in absence of antibiotic. We proposed this strategy as an approach to overcoming plasmid instability and to enhance enzyme activity and stability, avoiding antibiotic utilization. A model was developed to understand and predict the behaviour of bacteria and pollutants in a bioreactor system, considering: fluid dynamics, molecular/cellular scale processes and biofilm formation.  相似文献

High zinc ion concentrations produce an inhibitory effect on the naturally occurring hemolytic activity of theHolothuria polii coelomic fluid and coelomocyte lysate. The naturally occurring hemagglutinating activity remains unaffected. The inhibition appears reversible, suggesting a non-covalent interaction of the ions with the hemolysins. Moreover, as demonstrated by SDS-PAGE and immunoblotting analysis, the Zn++ treatment does not produce molecular weight variations in the coelomic fluid and coelomocyte lysate hemolytic patterns. The level of specialization of the entomopathogenic nematode Steinernema scapterisci with its native Xenorhabdus symbiont was investigated by testing (1) the influence of non-native bacterial strains on nematode fitness within an insect-host (Galleria mellonella) and (2) specificity of the association between the nematode infective juveniles and non-native bacteria. All non-native Xenorhabdus spp. or Photorhabdus spp. strains tested were mutualistically associated with other entomopathogenic nematodes in nature. We showed that most of the Xenorhabdus spp. strains tested led to an insignificant difference of the nematode's fitness compared to the one obtained with the native bacterium. Conversely, Photorhabdus spp. strains almost entirely abolished nematode reproduction. The phylogenetic analysis of bacterial strains tested, showed that there was a negative correlation between S. scapterisci's reproduction rate with a bacterial strain and the genetic distance of this bacterial strain from the native one. We also showed that the native bacterium was the only one which was transmitted by S. scapterisci's infective juveniles. All these results, suggested a specialization between S. scapterisci and its native Xenorhabdus. As the same phenomenon was already demonstrated in the association between S. carpocapsae and X. nematophila, specialization between partners would not be an exception in entomopathogenic nematode-bacteria interactions. Nevertheless, S. scapterisci showed a dramatically higher compatibility with non-native Xenorhabdus spp. strains than did S. carpocapsae, suggesting differences in the co-evolutionary processes between nematodes and bacteria in these two model systems.Table 1 List of the bacterial strains, native nematode species with their geographical origin, accession numbers of bacterial 16S rDNA partial sequences and number of combination experiments for each bacterium tested

Development of an ELISA for the detection of bromoxynil in water   总被引:4,自引:0,他引:4  
For development of an indirect competitive enzyme-linked immunosorbent assay (ELISA) for the nitrile herbicide bromoxynil, the polyclonal antibodies raised against 2,6-dibromo-4-cyano-phenoxyacetic acid (hapten) conjugated to bovine serum albumin (BSA) by the N-hydroxysuccinimide-activated ester method. Antiserum with a sufficiently high titer to provide the determinations of targeted compounds was obtained only 77 days after the primary immunization. Antiserum A2 was applied to the residual analysis of some water samples, under optimized ELISA condition, the quantitative working range was from 10 to 500 ppb with a limit of detection of 5 ppb. Cross-reactivity to structurally similar agrochemicals and related chemicals was determined. The antiserum showed little cross-reactivity with 2,6-dibromophenol and bromoxynil octanoate ester which have a dibromophenol group as common structure, but showed no cross-reactivity with other herbicides. Each water sample (river water, tap water, purified water, and bottled water) had a matrix effect and was investigated by adding Tween20 in the assay buffer. These four kinds of water samples were fortified with bromoxynil at several concentration levels and were directly analyzed with only dilution with an equal volume of antiserum solution, the mean recovery was 102.3%, and the mean coefficient of variation was 5.96%. The proposed ELISA turned out to be a powerful tool for monitoring of residual bromoxynil in water samples at trace level.  相似文献

In this study, metals (Be, Cr, Mn, Fe, Ni, Cu, Zn, Ag, Cd, Pb and Hg) in the fine-grained fraction (<63 microm) from 12 sites at different locations in northern San Francisco Bay over a year period from March 2000 to March 2001 were analyzed after acid extraction. The results showed that metal concentrations in the sediments varied from site to site, whereas some of them were found elevated with respect to the sediment of Tomales Bay, CA, which has little contamination history, indicating an enrichment of the metals in the sediment samples analyzed. Sediment toxicity and bioaccumulation evaluation by a clam species, Macoma nasuta, exposed to the sediment samples collected from the six sampling sites was carried out. The results showed that the sediment samples tested significantly reduced clam survival. Toxicity of the sediments to the clam was, in part, related to elevated metal concentrations in the sediments. In order to examine geochemistry of the metals and to understand potential correlations between metal concentrations and geochemical matrix elements of the sediments, bioavailability and toxicity of the metals, detailed analysis of metal concentrations associated with total organic carbon and the Fe-oxy-hydroxides in the sediment samples was performed. The analysis showed that sediment geochemistry appeared to influence metal bioavailability and may have important impacts on the toxicity of these metals to the clam.  相似文献

During 1998 regional surveys were conducted to evaluate natural radiation exposure of people in the vicinity of the six Spanish nuclear power stations. Indoor radon, external gamma dose rates outdoors and indoors and radioactivity in soil were measured in these surveys. The highest mean annual effective dose to the population was found in the surroundings of the Almaraz nuclear power plant in the province of Cáceres and was mainly due to the presence of high radon concentrations in homes. In order to make a more accurate assessment of the dose coming from the radon in this area, a new and more extensive surgery on indoor radon with a total of 380 measurements was carried out in 2000 in the Campo Ara?uelo region around the Almaraz nuclear power station. From the results obtained in this survey, a population-weighted mean annual effective dose from radon exposure of 1.7 mSv y(-1), 44% lower than that previously reported in 1998, was estimated for the whole Campo Ara?uelo region. The maximum dose value due to radon exposure, about 4 mSv y(-1), was reached in the so-called La Vera area located in the north of this region. The relationship between the indoor radon concentrates experimentally measured, the geological characteristics of this geographic area and the information provided by the radiation map of the Spanish MARNA Project is also analyzed, confirming that La Vera area should be considered as a high radon level area.  相似文献

The concept of participation in rural development has been evolutionary for the past two decades with those involved, such as development agencies and governments, particularly in rural water supply, re-evaluating their active role. The move towards effective community participation has encouraged a shift from the traditional top-down to a bottom-up approach whereby there is a decentralisation of unevenly distributed resources and power to empower a community and allow mobility of ‘people participation’. The Molinos water project is the first large-scale development project of its kind introduced into the village of Molinos in an under-developed area of Chile, where there has been no tradition of people participation. The project objective was to implement a low technology, low budget water treatment plant to the village of Molinos. Various aspects have hindered the continued development of the project including both technical and financial. In terms of people participation, the initial approach used was the top-down approach. There was a failure to fully integrate the community or inform the community in a formal manner about the project and consult them regarding key project issues. This case study illustrates that the lack of comprehensive consultation and the low level of participation of the community on the participatory scale does not achieve much in terms of people-centred benefits. For governance at the local level to be effective, participation should be inclusive and communicative so as to enhance transparency throughout the project lifetime. In the present work the effect of the alkyl chain length and the position of the sulfophenyl substituent of the linear alkylbenzene sulfonates (LAS) on their anaerobic biodegradability have been investigated. Degradation kinetics of the linear alkyl benzene sulfonates homologues, 2C10LAS, 2C12LAS and 2C14LAS, have been studied. It has been also investigated the effect of the isomer type on the degradation rate of the LAS molecule through the comparative study of the 2C10LAS and 5C10LAS isomers. Batch anaerobic biodegradation tests were performed using sludge from the anaerobic digester of a wastewater treatment plant as microorganisms source. Ultimate biodegradation was evaluated from the biogas production whereas primary biodegradation was determined by specific analysis of the surfactant. LAS homologues and isomers showed a negligible primary biodegradation under anaerobic conditions. Furthermore, analysis of sulfophenyl carboxilates (SPC) by LC–MS indicated a low and constant level of these LAS degradation metabolites over the test period. These data are consistent with a minimal transformation of the LAS parent molecule in the anaerobic digesters. On the other hand, the addition of the shortest alkyl chain length homologues, decyl and dodecylbenzene sulfonates, reduces the biogas production whereas the most hydrophobic homologue, the tetradecylbenzene sulfonate, enhances the biogas production. This LAS homologue seems to increase the availability of organic compounds sorbed on the anaerobic sludge promoting their biodegradation.  相似文献
