土壤宏蛋白质组学在揭示土壤微生物功能、代谢与环境相互作用方面具有广阔的应用前景,但由于土壤样品的特殊性,土壤蛋白质提取步骤是限制土壤宏蛋白质组学大规模应用的瓶颈之一.本文从样品制备、提取方法、影响因素等方面综述了土壤蛋白提取方面的研究进展.一般来说,根据实验目的、蛋白种类及后续研究方法设计相应的分组成收集策略才能取得较好的提取效果.土壤总蛋白、胞内蛋白与胞外蛋白分别有不同的提取方法.总蛋白提取一般采用直接提取法;胞外蛋白不需要裂解;胞内蛋白提取方法有直接提取法和间接提取法等.裂解、浓缩、去除腐殖质的方法以及提取液、pH值的选择等也会影响提取效果.此外,简单介绍了土壤宏蛋白质组学的应用,并对今后的研究工作提出展望.表1参36  相似文献

The objective of this research was to determine the spatial distributions of childhood lead poisoning and soil lead contamination in urban Tijuana. The Bocco–Sanchez model of point-source emissions was evaluated in terms of validity and reliability. We compared the model's predicted vulnerable populations with observed cases of childhood lead poisoning in Tijuana, identified fixed point sources in the field, and analyzed 76 soil samples from 14 sites. The soil lead results were compared to the blood lead analyses performed on Tijuana children whose blood lead levels were ≥10 μg/dL, who reported that they did not use lead-glazed ceramics for cooking or storing food (n = 63). Using GIS, predicted vs observed risk areas were assessed by examining spatial patterns, including the distribution of cases per designated risk area. Chi-square analysis of expected vs observed values did not differ significantly at the p = 0.02 level, showing that the model was strikingly accurate in predicting the distribution of subjects with elevated blood lead. Results reveal that while point sources are significant, other sources of lead exposure are also important. The relative public health risk from exposure to lead in an urban setting may be assessed by distinguishing among sources of exposure and associating concentrations to blood lead levels. The results represent an iterative approach in environmental health research by linking environmental and human biomarker lead concentrations and using these results to validate an environmental model of risk to lead exposure.  相似文献

The treatment of an effluent from the production of trifluraline was studied using a 1-L, semi-batch, tank-stirred glass reactor for performing three different advanced oxidation processes (photoperoxidation, Fenton, photo-Fenton). A commercial, medium-pressure mercury lamp was used for sample irradiation. The degradation was monitored by measurements of absorptiometric color reduction, UV-visible absorption spectra, and chemical oxygen demand (COD). The obtained results showed that the photo-Fenton process was the most effective treatment for the trifluraline effluent.  相似文献

Adsorption and mobility of linuron (3-(3,4-dichlorophenyl)-1-methoxy-1-methylurea) and diuron (3-(3,4-dichlorophenyl)-1, 1-dimethylurea) were studied in clayey soils from the Gharb area (Morocco). Soils A and B were planted with sun flower (Helianthus annuus) while soil C was planted with sugar cane (Saccharum offcinarum). Adsorption was studied for linuron in soils A and B, while mobility was studied only in soil B. Adsorption data were found to fit the Freundlich equation with correlation coefficients r2 > 0.9. Freundlich coefficients (Kf, nf) were in agreement with L and S isotherm types for soils A and B, respectively. Values of Koc (195 and 102) indicate moderate adsorption. Desorption isotherms for linuron showed hysteresis for both soils. The pesticide would be more bound to soil A (H = 8.44) than to soil B (H = 4.01). The effect of alternating wet and dry conditions was tested for soils A and B. Results showed that retention would increase in soil subject to an additional wet and dry cycle. In the case of diuron isotherm was of type L in soil C. Desorption was noticeable at high concentrations and tended to decrease when concentrations diminished. Mobility of linuron was tested in polyvinyle chloride (PVC) columns, which received different treatments before their percolation. The pesticide was more mobile in a previously saturated column. In columns subject to a drying step after saturation with water, linuron mobility was greatly reduced.  相似文献

In species‐rich tropical forests, effective biodiversity management demands measures of progress, yet budgetary limitations typically constrain capacity of decision makers to assess response of biological communities to habitat change. One approach is to identify ecological‐disturbance indicator species (EDIS) whose monitoring is also monetarily cost‐effective. These species can be identified by determining individual species’ responses to disturbance across a gradient; however, such responses may be confounded by factors other than disturbance. For example, in mountain environments the effects of anthropogenic habitat alteration are commonly confounded by elevation. EDIS have been identified with the indicator value (IndVal) metric, but there are weaknesses in the application of this approach in complex montane systems. We surveyed birds, small mammals, bats, and leaf‐litter lizards in differentially disturbed cloud forest of the Ecuadorian Andes. We then incorporated elevation in generalized linear (mixed) models (GL(M)M) to screen for EDIS in the data set. Finally, we used rarefaction of species accumulation data to compare relative monetary costs of identifying and monitoring EDIS at equal sampling effort, based on species richness. Our GL(M)M generated greater numbers of EDIS but fewer characteristic species relative to IndVal. In absolute terms birds were the most cost‐effective of the 4 taxa surveyed. We found one low‐cost bird EDIS. In terms of the number of indicators generated as a proportion of species richness, EDIS of small mammals were the most cost‐effective. Our approach has the potential to be a useful tool for facilitating more sustainable management of Andean forest systems. Abstract:  The 735,000-km2 Caatinga is a mosaic of thorn scrub and seasonally dry forests, with more than 2000 species of vascular plants, fishes, reptiles, amphibians, birds, and mammals. Endemism in these groups varies from 7% to 57%. Inappropriate land use has already caused serious environmental damage and accelerating desertification, which is currently threatening about 15% of the region. Moreover, the rich and diversified biota of the Caatinga is poorly protected: only 11 reserves (<1% of the region) are strictly protected areas. A conservation strategy for the Caatinga biota should be designed to (1) avoid further habitat loss and desertification, (2) maintain key ecological services necessary for improving the living standards of the rural population, and (3) promote the sustainable use of the region's natural resources. Implementing an effective conservation agenda for the Caatinga is not an easy task but, with creativity and consistent financial support, it should be possible to nurture this unique biome and guarantee the preservation of its rich and diversified fauna and flora, and, with this, the well-being of its rural populations.  相似文献

Abstract:  Not all species are likely to be equally affected by habitat fragmentation; thus, we evaluated the effects of size of forest remnants on trophically linked communities of plants, leaf-mining insects, and their parasitoids. We explored the possibility of differential vulnerability to habitat area reduction in relation to species-specific and food-web traits by comparing species–area regression slopes. Moreover, we searched for a synergistic effect of these traits and of trophic level . We collected mined leaves and recorded plant, leaf miner, and parasitoid species interactions in five 100-m2 transects in 19 Chaco Serrano woodland remnants in central Argentina. Species were classified into extreme categories according to body size, natural abundance, trophic breadth, and trophic level . Species–area slopes differed between groups with extreme values of natural abundance or trophic specialization. Nevertheless, synergistic effects of life-history and food-web traits were only found for trophic level and trophic breadth: area-related species loss was highest for specialist parasitoids. It has been suggested that species position within interaction webs could determine their vulnerability to extinction. Our results provide evidence that food-web parameters, such as trophic level and trophic breadth, affect species sensitivity to habitat fragmentation .  相似文献
