There is increasing interest in continual monitoring of air for the presence of inhalation health hazards, such as particulate matter, produced through combustion of fossil fuels. Currently there are no means to rapidly evaluate the relative toxicity of materials or to reliably predict potential health impact due to the complexity of the composition, size, and physical properties of particulate matter. This research evaluates the feasibility of utilizing cell cultures as the biological recognition element of an inhalation health monitoring system. The response of rat lung type II epithelial (RLE-6TN) cells to a variety of combustion derived particulates and their components has been evaluated. The focus of the current work is an evaluation of how particles are delivered to a cellular sensing array and to what degree does washing or grinding of the particles impacts the cellular response. There were significant differences in the response of these lung cells to PM's of varying sources. Mechanical grinding or washing was found to alter the toxicity of some of these particulates; however these effects were strongly dependent on the fuel source. Washing reduced toxicity of oil PM's, but had little effect on those from diesel or coal. Mechanical grinding could significantly increase the toxicity of coal PM's, but not for oil or diesel.  相似文献

Enhanced biological phosphorus removal (EBPR) from wastewater relies on the enrichment of activated sludge with phosphorus-accumulating organisms (PAOs). The presence and proliferation of glycogen-accumulating organisms (GAOs), which compete for substrate with PAOs, may be detrimental for EBPR systems, leading to deterioration and, in extreme cases, failure of the process. Therefore, from both process evaluation and modeling perspectives, the estimation of PAO and GAO populations in activated sludge systems is a relevant issue. A simple method for the quantification of PAO and GAO population fractions in activated sludge systems is presented in this paper. To develop such a method, the activity observed in anaerobic batch tests executed with different PAO/GAO ratios, by mixing highly enriched PAO and GAO cultures, was studied. Strong correlations between PAO/GAO population ratios and biomass activity were observed (R2 > 0.97). This served as a basis for the proposal of a simple and practical method to quantify the PAO and GAO populations in activated sludge systems, based on commonly measured and reliable analytical parameters (i.e., mixed liquor suspended solids, acetate, and orthophosphate) without requiring molecular techniques. This method relies on the estimation of the total active biomass population under anaerobic conditions (PAO plus GAO populations), by measuring the maximum acetate uptake rate in the presence of excess acetate. Later, the PAO and GAO populations present in the activated sludge system can be estimated, by taking into account the PAO/GAO ratio calculated on the basis of the anaerobic phosphorus release-to-acetate consumed ratio. The proposed method was evaluated using activated sludge from municipal wastewater treatment plants. The results from the quantification performed following the proposed method were compared with direct population estimations carried out with fluorescence in situ hybridization analysis (determining Candidatus Accumulibacter Phosphatis as PAO and Candidatus Competibacter Phosphatis as GAO). The method showed to be potentially suitable to estimate the PAO and GAO populations regarding the total PAO-GAO biomass. It could be used, not only to evaluate the performance of EBPR systems, but also in the calibration of potential activated sludge mathematical models, regarding the PAO-GAO coexistence.  相似文献

We have carried out kinetic studies to characterize the heterogeneous decay of octamethylcyclotetrasiloxane (D4) and decamethylcyclopentasiloxane (D5) in the presence of representative mineral dust aerosol in order to obtain a better understanding of the atmospheric fate of these siloxanes. The heterogeneous chemistry of D4 and D5 with various mineral dusts was studied in an environmental aerosol reaction chamber using FTIR absorption spectroscopy to monitor the reaction. The apparent heterogeneous uptake coefficient, γapp, for D4 and D5 with various mineral dusts was measured under dry conditions and as a function of relative humidity (RH). In addition, the effect of initial D4 and D5 concentration on the rate and yield of the reaction was examined. The uptake coefficient, γapp, for D4 and D5 was similar for the most reactive aerosols tested, with kaolinite ≈hematite > silica. Limited uptake onto carbon black and calcite surfaces was observed for either siloxane. Reaction with hematite and kaolinite resulted in multilayer coverages, suggesting extensive polymerization of D4 and D5 on the aerosol surface.  相似文献

In this paper, the European Union's Water Framework Directive 2000/60/EC (WFD) that is intended to foster protection of water resources is examined, focusing on the improvement of ecological and chemical quality of surface and groundwater. The WFD includes the concept of full cost recovery (FCR) in accordance with the Polluter-Pays Principle, as one of the tools of an adequate and sustainable water resource management system. The WFD defines three different costs associated with water: resource costs (RC), financial costs (FC), and environmental costs (ECs).The FCR of water is examined from a biophysical perspective using emergy evaluation to: (1) establish resource values of water from different sources, (2) establish the full economic costs associated with supplying water, and (3) the societal costs of water that is used incorrectly; from which the resource costs, financial costs, and environmental costs, respectively, can be computed. Financial costs are the costs associated with providing water including energy, materials, labor and infrastructure. The emergy based monetary values vary between 0.15 and 1.73 €/m3 depending on technology. The emergy based, global average resource value (from which resource costs can be computed) is derived from two aspects of water: its chemical potential and its geopotential. The chemical potential monetary value of different sources such as rain, groundwater, and surface water derived from global averages of emergy inputs varies from 0.03 to 0.18 €/m3, depending on source, and the geopotential values vary from 0.03 to 2.40 €/m3, depending on location in the watershed. The environmental costs of water were averaged for the county of Spain and were 1.42 €/m3.Time of year and spatial location within the watershed ultimately influence the resource costs (computed from emergy value of chemical potential and geopotential energy) of water. To demonstrate this spatial and temporal variability, a case study is presented using the Foix watershed in northeastern Spain. Throughout the year, the resource value of water varies from 0.21 to 3.17 €/m3, depending on location within the watershed. It is concluded that FCR would benefit from the evaluation of resource costs using spatially and temporally explicit emergy accounting.  相似文献

A major challenge in ecology is to understand how the millions of species on Earth are organized into biological communities. Mechanisms promoting coexistence are one such class of organizing processes, which allow multiple species to persist in the same trophic level of a given web of species interactions. If some mechanism promotes the coexistence of two or more species, each species must be able to increase when it is rare and the others are at their typical abundances; this invasibility criterion is fundamental evidence for species coexistence regardless of the mechanism. In an attempt to evaluate the level of empirical support for coexistence mechanisms in nature, we surveyed the literature for empirical studies of coexistence at a local scale (i.e., species found living together in one place) to determine whether these studies satisfied the invasibility criterion. In our survey, only seven of 323 studies that drew conclusions about species coexistence evaluated invasibility in some way in either observational or experimental studies. In addition, only three other studies evaluated necessary but not sufficient conditions for invasibility (i.e., negative density dependence and a trade-off in performance that influences population regulation). These results indicate that, while species coexistence is a prevalent assumption for why species are able to live together in one place, critical empirical tests of this fundamental assumption of community structure are rarely performed. These tests are central to developing a more robust understanding of the relative contributions of both deterministic and stochastic processes structuring biological communities.  相似文献

The background noise generated in large social aggregations of calling individuals is a potent source of auditory masking for animals that communicate acoustically. Despite similarities with the so-called "cocktail-party problem" in humans, few studies have explicitly investigated how non-human animals solve the perceptual task of separating biologically relevant acoustic signals from ambient background noise. Under certain conditions, humans experience a release from auditory masking when speech is presented in speech-like masking noise that fluctuates in amplitude. We tested the hypothesis that females of Cope's gray treefrog (Hyla chrysoscelis) experience masking release in artificial chorus noise that fluctuates in level at modulations rates characteristic of those present in ambient chorus noise. We estimated thresholds for recognizing conspecific advertisement calls (pulse rate=40-50 pulses/s) in the presence of unmodulated and sinusoidally amplitude modulated (SAM) chorus-shaped masking noise. We tested two rates of modulation (5 Hz and 45 Hz) because the sounds of frog choruses are modulated at low rates (e.g., less than 5-10 Hz), and because those of species with pulsatile signals are additionally modulated at higher rates typical of the pulse rate of calls (e.g., between 15-50 Hz). Recognition thresholds were similar in the unmodulated and 5-Hz SAM conditions, and 12 dB higher in the 45-Hz SAM condition. These results did not support the hypothesis that female gray treefrogs experience masking release in temporally fluctuating chorus-shaped noise. We discuss our results in terms of modulation masking, and hypothesize that natural amplitude fluctuations in ambient chorus noise may impair mating call perception.  相似文献

To aid in the management and conservation of Southwestern Willow Flycatcher (Empidonax traillii extimus, hereafter “Flycatcher”), we developed numerous models of flycatcher breeding habitat at Roosevelt Lake, AZ. For model development and testing, we compiled 10 years of flycatcher territory data that were obtained from intensive fieldwork between 1996 and 2005. We identified riparian vegetation annually in the project area from Landsat Thematic Mapper images, and extracted floodplain features from a digital elevation model. We created a novel class of temporal (i.e., multiyear) variables by characterizing the stability and variability in breeding habitat over a 6-year time interval. We used logistic regression to determine associations between environmental variables and flycatcher territory occurrence, and to test specific hypotheses. We mapped the probability of territory occurrence with a GIS and determined model accuracies with a classification table and a 10-year population database. Environmental features that were associated with breeding flycatchers included floodplain size, proximity to water, and the density, heterogeneity, age and stability of riparian vegetation. Our best model explained 79% of the variability in the flycatcher breeding population at Roosevelt Lake. The majority of predicted flycatcher habitat formed between 1996 and 2004 on an exposed lakebed ~3 years after water levels receded during a prolonged drought. A high correlation between annual reservoir levels and predicted breeding habitat (r = ?0.82) indicates that we can create and manage habitat for conservation purposes. Our predictive models quantify and assess the relative quality of flycatcher breeding habitat remotely, and can be used to evaluate the effectiveness of habitat restoration activities. Numerous techniques we developed can be used to characterize riparian vegetation and patch dynamics directly off of satellite imagery, thereby increasing its utility for conservation purposes.  相似文献

Caribbean coral reefs are increasingly dominated by macroalgae instead of corals due to several factors, including the decline of herbivores. Yet, virtually unknown is the role of crustacean macrograzers on coral reef macroalgae. We examined the effect of grazing by the Caribbean king crab (Mithrax spinosissimus) on coral patch reef algal communities in the Florida Keys, Florida (USA), by: (1) measuring crab selectivity and consumption of macroalgae, (2) estimating crab density, and (3) comparing the effect of crab herbivory to that of fishes. Mithrax prefers fleshy macroalgae, but it also consumes relatively unpalatable calcareous algae. Per capita grazing rates by Mithrax exceed those of most herbivorous fish, but Mithrax often occurs at low densities on reefs and its foraging activities are reduced in predator-rich environments. Therefore, the effects of grazing by Mithrax tend to be localized and when at low density contribute primarily to spatial heterogeneity in coral reef macroalgal communities.  相似文献

Multi-scale resource selection modeling is used to identify factors that limit species distributions across scales of space and time. This multi-scale nature of habitat suitability complicates the translation of inferences to single, spatial depictions of habitat required for conservation of species. We estimated resource selection functions (RSFs) across three scales for a threatened ungulate, woodland caribou (Rangifer tarandus caribou), with two objectives: (1) to infer the relative effects of two forms of anthropogenic disturbance (forestry and linear features) on woodland caribou distributions at multiple scales and (2) to estimate scale-integrated resource selection functions (SRSFs) that synthesize results across scales for management-oriented habitat suitability mapping. We found a previously undocumented scale-specific switch in woodland caribou response to two forms of anthropogenic disturbance. Caribou avoided forestry cut-blocks at broad scales according to first- and second-order RSFs and avoided linear features at fine scales according to third-order RSFs, corroborating predictions developed according to predator-mediated effects of each disturbance type. Additionally, a single SRSF validated as well as each of three single-scale RSFs when estimating habitat suitability across three different spatial scales of prediction. We demonstrate that a single SRSF can be applied to predict relative habitat suitability at both local and landscape scales in support of critical habitat identification and species recovery.  相似文献
