The aim of this study was to follow contamination of ready to eat milk and meat products with Salmonella spp. by using the StepOne real-time polymerase chain reaction (PCR). Classical microbiological methods for detection of foodborne bacteria involve the use of pre-enrichment and/or specific enrichment, following isolation of bacteria in solid media and the final confirmation by biochemical and/or serological tests. We used the PrepSEQ Rapid Spin Sample Preparation Kit for isolation of DNA and MicroSEQ? Salmonella spp. Detection Kit for pursuance of the real-time PCR (Applied Biosystems). In samples without incubation we detected strain of Salmonella sp. in 5 out of 25 samples (swabs), as well as in the internal positive control (IPC), which was positive in all samples. This StepOne real-time PCR assay is extremely useful for any laboratory equipped by real-time PCR. It is a fast, reproducible, simple, specific and sensitive way to detect nucleic acids, which could be used in clinical diagnostic tests in the future. Our results indicated that real-time PCR assay developed in this study could sensitively detect Salmonella spp. in ready-to-eat food. This could prevent infection caused by Salmonella, and also could benefit food manufacturing companies by extending their product's shelf-life as well as saving the cost of warehousing their food products while awaiting pathogen testing results.  相似文献

The bottlenose dolphin (Tursiops truncatus) is the most common cetacean species worldwide and the only marine mammal species resident in the Croatian part of the Adriatic Sea. To gain insight into genetic diversity of bottlenose dolphins at adaptively important loci relevant to conservation, we analysed the polymorphism of major histocompatibility complex (MHC) genes, which play a key role in pathogen confrontation and clearance. Specifically, we examined the diversity of MHC class II DRA, DQA and DQB alleles in 50 bottlenose dolphins from the Adriatic Sea collected between 1997 and 2011 and in 12 animals from other Mediterranean locations. Notable variation in DQA, DQB and three-locus haplotypes was found, with all 10 DQA and 12 DQB alleles encoding unique protein products. Analysis of the ratio of non-synonymous to synonymous substitution rates suggests that positive selection acts at both highly variable loci. Phylogenetic analyses revealed trans-species polymorphism at the DQB locus, strongly indicating the influence of balancing selection in the long term. In fact, the balancing selection observed in bottlenose dolphins is higher than that reported for most other cetaceans and comparable to that seen in terrestrial mammals.  相似文献

PCB 136 is an environmentally relevant chiral PCB congener, which has been found in vivo to be present in form of rotational isomers (atropisomers). Its atropselective biotransformation or neurotoxic effects linked with sensitization of ryanodine receptor suggest that it might interact also with other intracellular receptors in a stereospecific manner. However, possible atropselective effects of PCB 136 on nuclear receptor transactivation remain unknown. Therefore, in this study, atropselective effects of PCB 136 on nuclear receptors controlling endocrine signaling and/or expression of xenobiotic and steroid hormone catabolism were investigated. PCB136 atropisomers were found to exert differential effects on estrogen receptor (ER) activation; (+)-PCB 136 was estrogenic, while (?)-PCB 136 was antiestrogenic. In contrast, inhibition of androgen receptor (AR) activity was not stereospecific. Both PCB136 stereoisomers induced the constitutive androgen receptor (CAR)-dependent gene expression; however, no significant stereospecificity of PCB 136 atropisomers was observed. PCB136 was a partial inducer of the pregnane X receptor (PXR)-dependent gene expression. Here, (?)-PCB 136 was a significantly more potent inducer of PXR activity than (+)-PCB 136. Taken together, the present results indicate that at least two nuclear receptors participating in endocrine regulation or metabolism, ER and PXR, could be regulated in an atropselective manner by chiral PCB 136. The enantioselective enrichment of PCB atropisomers in animal and human tissues may thus have significant consequences for endocrine-disrupting effects of chiral ortho-substituted PCB congeners.  相似文献

The present study was designed to reveal whether long-term consumption of bitter apricot seeds causes changes in lipid profile and other risk factors for cardiovascular diseases. The study group consisted of 12 healthy adult volunteers (5 females and 7 males). The average age of women was 41.60 ± 11.28 years and the average age of men was 36.71 ± 13.70 years. Volunteers consumed 60 mg kg?1 of body weight of bitter apricot seeds divided into 8–12 doses daily for 12 weeks. Volunteers were recruited from the general population of Slovak Republic. After 12 weeks, mean body weight of the participants increased from 77.34 to 78.22 kg (P > 0.05). The average total cholesterol levels decreased from 4.86 mmol L?1 at the beginning of the study to 4.44 mmol L?1 at the end of the study (P < 0.05). We did not observe any significant increase in high-density cholesterol (from 1.55 to 1.60 mmol L?1). The average low-density cholesterol levels decreased from 2.93 mmol L?1 at the beginning of the study to 2.31 mmol L?1 at the end of the study (P < 0.001). Concentration of triglycerides increased significantly over the 12-week intervention period from 0.84 to 1.17 mmol L?1. After the intervention, the high-sensitivity C-reactive protein level decreased from 1.92 to 1.23 mg L?1, but results were non-significant (P > 0.05). Creatine kinase serum levels increased from 2.31 to 2.77 mg L?1 (P > 0.05) over the 12-week intervention period. The results suggest that regular intake of bitter apricot seeds may be considered potentially useful for prevention of cardiovascular diseases.  相似文献

Mycobiota (fungi, lichens and myxomycetes) was examined in permanent plots following experiments of artificial drought (D) and nitrogen addition (N) and compared with untreated forest (C), in Scots pine stand planted on Arenosols. Species diversity and relationships between fungal community structure and environmental variables (plant species numbers and cover, bryophyte cover, soil and bark pH, tree mortality) were explored. Both D and N treatments lead to decrease of fungal species in general, however, responses of individual trophic and ecological groups varied. The strongest effect of the treatments was observed for soil fungi, especially mycorrhizal: numbers of fruiting species and ectomycorrhizal root tips decreased, and species composition has changed. Saprotrophic fungi reacted by changes in species composition but not in numbers. Of the studied environmental variables, the most significant effect on mycobiota had bryophyte and vascular plant cover as well as vascular plant species numbers.  相似文献

Arsenic is a toxic and carcinogenic element. Its toxicity depends on its oxidation state and its concentration. The aim of this paper is to determine, for the first time, the concentration levels of arsenic in water and sediment during the spring/summer period of 2009 in Sevojno, a region in West Serbia with a long industrial tradition, as well as to determine the model of arsenic distribution in water/sediment system and the level of its compatibility with the existing theoretical model. Adsorption is a continual process in the environment. It plays a very important role in the transport and fate of pollutants, especially in sediment. The adsorption of arsenic was examined using the Freundlich adsorption isotherm.  相似文献

The soils adjacent to an area of historical mining, ore processing and smelting activities reflects the historical background and a mixing of recent contamination sources. The main anthropogenic sources of metals can be connected with historical and recent mine wastes, direct atmospheric deposition from mining and smelting processes and dust particles originating from open tailings ponds. Contaminated agriculture and forest soil samples with mining and smelting related pollutants were collected at different distances from the source of emission in the Pb–Zn–Ag mining area near Olkusz, Upper Silesia to (a) compare the chemical speciation of metals in agriculture and forest soils situated at the same distance from the point source of pollution (paired sampling design), (b) to evaluate the relationship between the distance from the polluter and the retention of the metals in the soil, (c) to describe mineralogy transformation of anthropogenic soil particles in the soils, and (d) to assess the effect of deposited fly ash vs. dumped mining/smelting waste on the mobility and bioavailability of metals in the soil. Forest soils are much more affected with smelting processes than agriculture soils. However, agriculture soils suffer from the downward metal migration more than the forest soils. The maximum concentrations of Pb, Zn, and Cd were detected in a forest soil profile near the smelter and reached about 25 g kg − 1, 20 g kg − 1 and 200 mg kg − 1 for Pb, Zn and Cd, respectively. The metal pollutants from smelting processes are less stable under slightly alkaline soil pH then acidic due to the metal carbonates precipitation. Metal mobility ranges in the studied forest soils are as follows: Pb > Zn ≈ Cd for relatively circum-neutral soil pH (near the smelter), Cd > Zn > Pb for acidic soils (further from the smelter). Under relatively comparable pH conditions, the main soil properties influencing metal migration are total organic carbon and cation exchange capacity. The mobilization of Pb, Zn and Cd in soils depends on the persistence of the metal-containing particles in the atmosphere; the longer the time, the more abundant the stable forms. The dumped mining/smelting waste is less risk of easily mobilizable metal forms, however, downward metal migration especially due to the periodical leaching of the waste was observed.  相似文献

Because aerosol particle deposition is an important factor in indoor air quality, many empirical and theoretical studies have attempted to understand the process. In this study, we estimated the deposition rate of aerosol particles on smooth aluminum surfaces inside a test chamber. We investigated the influence of turbulent intensity due to ventilation and fan operation. We also investigated two important processes in particle deposition: turbophoresis, which is significant for micron particles, and coagulation, which is relevant to ultrafine particles (UFP diameter <0.1 μm) at high particle concentrations. Our analysis included semi-empirical estimates of the deposition rates that were compared to available deposition models and verified with simulations of an aerosol dynamics model. In agreement with previous studies, this study found that induced turbulent intensity greatly enhanced deposition rates of fine particles (FP diameter <1 μm). The deposition rate of FP was proportional to the ventilation rate, and it increased monotonically with fan speed. With our setup, turbophoresis was very important for coarse particles larger than 5 μm. The coagulation of aerosol particles was insignificant when the particle concentration was less than 104 cm?3 during fan operation. The model simulation results verified that the aerosol dynamics module incorporated in our Multi-Compartment and Size-Resolved Indoor Aerosol Model (MC-SIAM) was valid. The behavior of aerosol particles inside our chamber was similar to that found in real-life conditions with the same ventilation rates (0.018–0.39 h?1) and similar air mixing modes. Therefore, our findings provide insight into indoor particle behavior.  相似文献
