Polychlorinated biphenyls (PCBs) are present in the environment as complex mixtures, which make it challenging to identify PCB congeners that may be subject to active transport processes. Here we employ a transgenic mouse model in combination with multivariate analyses to investigate if chiral PCBs 91, 95, 132, 136, 149, 174, 176 and 183 are subject to active (enantioselective) transport by multidrug resistance (MDR) transporters. A synthetic PCB mixture containing these congeners was administered orally to female FVB or mdr1a/1b knockout mice. Due to the short half-life of chiral PCB congeners, mice were euthanized after 24 h and PCB concentrations and enantiomeric fractions were determined in selected tissues and excreta. Principal component analysis did not reveal differences between wild-type and mdr1a/1b knockout mice. However, Hotelling T2-test revealed significantly lower PCB concentrations and a more pronounced enantiomeric enrichment in the adipose tissue of mdr1a/1b knockout mice. These differences are due to higher body weights and higher fecal fat contents of mdr1a/1b knockout mice. Analysis of the enantiomeric fractions of PCBs 91, 95, 136, 149 and 174 showed a significant enantiomeric enrichment for all five congeners in wild-type and mdr1a/1b knockout mice. Overall, by studying a PCB mixture in a transgenic mouse model in combination with a multivariate data reduction approach, PCBs 91, 95, 136, 149 and 174 could be excluded as substrates of multidrug resistance transporters 1a/b.  相似文献

Results are presented for 137Cs, 90Sr and plutonium activity concentrations in more than 20 samples of terrestrial invertebrates, including species of beetles, ants, spiders and millipedes, collected in the highly contaminated area of the Chernobyl exclusion zone. The majority of samples were collected in Belarus, with some also collected in the Ukraine. Three other samples were collected in an area of lower contamination. Results show that seven samples exceed an activity concentration of 100 kBq/kg (ash weight - a.w.) for 137Cs. The maximum activity concentration for this isotope was 1.52 ± 0.08 MBq/kg (a.w.) determined in ants (Formica cynerea). Seven results for 90Sr exceeded 100 kBq/kg (a.w.), mostly for millipedes. Relatively high plutonium activity concentrations were found in some ants and earth-boring dung beetles. Analyses of activity ratios showed differences in transfer of radionuclides between species. To reveal the correlation structure of the multivariate data set, the Partial Least-Squares method (PLS) was used. Results of the PLS model suggest that high radiocesium activity concentrations in animal bodies can be expected mainly for relatively small creatures living on the litter surface. In contrast, high strontium activity concentrations can be expected for creatures which conduct their lives within litter, having mixed trophic habits and a moderate lifespan. No clear conclusions could be made for plutonium.  相似文献

The paper presents the application of the hyphenated technique - high-performance liquid chromatography with atomic absorption spectrometry detection with hydride generation (HPLC-HG-AAS) - in the determinations of inorganic forms of arsenic: As(III) and As(V) in the exchangeable fraction of flood deposits. The separation of analytical signals of the determined arsenic forms was obtained using an ion-exchange column in a chromatographic system with the atomic absorption spectrometer as a detector, at the determination limits of 5 ng g−1 for As(III) and 10 ng g−1 for As(V). Flood deposits were collected after big flood event in valley of the Warta river which took place in summer 2010. Samples of overbank deposits were taken in Poznań agglomeration and vicinity (NW Poland). The results of determinations of arsenic forms in the exchangeable fraction of flood deposits allowed indication of a hypothetical path of deposits migration transported by a river during flood and environmental threats posed by their deposition by flood.  相似文献

The paper presents the results of year-long measurements of radon (222Rn) concentration inside 129 buildings in Poland in relation to the geological conditions of their foundation. The authors took into account the division of the country into tectonic units, as well as the lithology of the rocks forming the bedrock of these buildings. As expected, the highest value of mean annual 222Rn concentration (845 Bq/m3) was recorded in a building situated in the area of the Sudetes, while the highest geometric mean (characteristic of the expected log-normal data distribution) was calculated based on measurements from buildings located within the East-European craton, in the area of Mazury-Podlasie monocline, where it reached 231 Bq/m3. Such results reflect geological conditions - the occurrence of crystalline rocks (especially U- and Ra-enriched granites and orthogneisses) on the surface in the Sudetes, and of young post-glacial sediments containing fragments of Scandinavian crystalline rocks, also enriched with U and Ra, in the area of Mazury-Podlasie monocline. However, the least expected result of the investigations was finding out that, contrary to the hitherto widespread belief, none of the major tectonic units of Poland can be excluded from the list of those containing buildings with mean annual 222Rn concentration exceeding 200 Bq/m3. The mean annual concentration of radon for all the buildings were much higher than the mean concentration value (49.1 Bq/m3) of indoor radon in Poland quoted so far. These results cast a completely new light on the necessity to perform measurements of radon concentration in residential buildings in Poland, no more with reference to small areas with outcrops of crystalline rocks (especially the Sudetes, being the Polish fragment of the European Variscan belt), but for all the major tectonic units within Poland.  相似文献

Characterization of bacterial communities at heavy-metal-contaminated sites   总被引:2,自引:0,他引:2  
The microbial community in soil samples from two long-term contaminated sites was characterized by using culture-dependent and culture-independent methods. The two sites investigated contained high amounts of heavy metals and were located in the upper Silesia Industrial Region in southern Poland. The evaluation of the aerobic soil microbial population clearly demonstrated the presence of considerable numbers of viable, culturable bacteria at both sites. A high fraction of the bacterial population was able to grow in the presence of high amounts of metals, i.e. up to 10 mM Zn2+, 3 mM Pb2+ or 1 mM Cu2+. Site 1 contained significantly (P < 0.05) lower bacterial numbers growing in the presence of 10 mM Zn2+ than site 2, while the opposite was observed for bacteria tolerating 1 mM Cu2+. This coincided with the contents of these two metals at the two sites. Ecophysiological (EP) indices for copiotrophs (r-strategists) and oligotrophs (K-strategists) pointed to high bacterial diversity at both sites. Fluorescence in situ hybridization (FISH) analysis indicated that Actinobacteria and Proteobacteria represent the physiologically active fraction of bacteria at the two sites. Shannon diversity (H′) indices for FISH-detected bacterial phylogenetic groups were not significantly different at the two sites.  相似文献

The present study deals with the application of the self-organizing map (SOM) technique in the exploration of spatiotemporal dynamics of spring and stream water samples collected in the Chocho?owski Stream Basin located in the Tatra Mountains (Poland). The SOM-based classification helped to uncover relationships between physical and chemical parameters of water samples and factors determining the quality of water in the studied high-mountain area. In the upper part of the Chocho?owski Stream Basin, located on the top of the crystalline core of the Tatras, concentrations of the majority of ionic substances were the lowest due to limited leaching. Significantly higher concentration of ionic substances was detected in spring and stream samples draining sedimentary rocks. The influence of karst-type springs on the quality of stream water was also demonstrated.  相似文献

Atmospheric concentrations of benzene, toluene, ethylbenzene, and xylenes (o-xylene and m,p-xylene) were assessed in the Tricity area (Gdańsk-Sopot-Gdynia, Poland) with the use of two types of passive samplers: permeation (homemade passive samplers) and diffusive (Radiello and Orsa 5). Samples were collected during 2008 at selected sites in the Tricity area at monitoring stations belonging to the agency of Regional Air Quality Monitoring Foundation. The field study was conducted to compare the performance of these two different types of passive samplers. A statistical approach was formulated, and the experimental data were evaluated using the paired t test, Wilcoxon signed rank-sum test, and Friedman analysis of variance. All the statistical results confirm the hypothesis that the differences between the performances of the three sampling devices are highly significant. Despite the fact that data obtained with the homemade passive sampler indicated that the results were higher compared with those for the Radiello and Orsa 5 diffusive samplers, the authors note that all differences between the homemade permeation sampler and the Radiello and Orsa 5 diffusive samplers are positive.  相似文献

In this study the values of subcooled vapor pressures (log PL) were estimated for 209 trans chloroazobenzenes (Ct-ABs) that fill some gaps in analytical and experimental data on these compounds. There are 209 chloro derivatives of trans azobenzenes that are relatively stable and more environmentally relevant than 209 chloro cis congeners. The calculations models were based on the Quantitative Structure-Property Relationship (QSPR) scheme using the semi-empirical method (PM6) in molecular package (MOPAC) software and density functional theory (DFT) method using B3LYP functional and 6-311++G** basis set) in Gaussian 03 software method and the artificial neural networks (ANNs) prediction. The values of log PL predicted by models used varied between ?3.94 to ?2.66 for Mono-; ?4.85 to ?2.97 for Di-; ?5.18 to ?3.17 for Tri-; ?6.02 to ?3.77 for Tetra-; ?6.64 to ?4.64 for Penta-; ?7.36 to ?4.76 for Hexa-; ?7.54 to ?5.79 for Hepta-; ?7.75 to ?6.64 for Octa-; ?7.89 to ?7.44 for Nona-Ct-Abs; and ?8.09 and ?8.13 for Deca-Ct-AB. Based on these values Ct-ABs can be grouped localized among relatively low (log PL ?4 to ?2) and low (log PL < ?4) mobile Persistent Organic Pollutants (POPs). Both the calculation methods employed were characterized by similar prediction ability of subcooled vapor pressure values of Ct-ABs, while those of PM6 are much more efficient due to a cheaper hardware used and around 300-fold less time spent on calculations.  相似文献

Azoxystrobin (methyl(2E)-2-{2-[6-(2-cyanophenoxy)pyrimidin-4-yloxy] phenyl}-3-methoxyacrylate) is an active ingredient used to protect crops against fungal diseases. The experience of the Polish control laboratory indicates relatively frequent cases of counterfeit plant protection products (PPPs) containing this active substance. The present study aimed to use chemometric methods to model chemical fingerprints obtained by different chromatographic techniques to verify the original formulation of PPPs containing the active substance azoxystrobin. The pesticides used in the study came from different sources (including stores and warehouses), were manufactured at a different time and came from different production batches. The results obtained with the HPLC-DAD and HS-GC-MS techniques were then modeled using principal component analysis (PCA) and soft independent modeling by class analogy (SIMCA) classifier. The proposed approach has been confirmed as useful for verifying the authenticity of PPPs and can be used in the routine control testing of SC pesticides containing azoxystrobin.  相似文献
